    Notizen: Summary The current-voltage (I−V) relations of the rheogenic Na-sugar cotransport mechanism at the apical membrane ofNecturus small intestine were determined from the relations between the electrical potential difference across the apical membrane, Ψ mc , and that across the entire epithelium, Ψ ms , when the latter was varied over the range ±200 mV, (i) under steady conditions in the presence of galactose and (ii) after the current across the apical membrane carried by the cotransporter,I SNa m , is blocked by the addition of phloridzin to the mucosal solution.I SNa m was found to be strongly dependent upon Ψ mc over the range −50 mV 〈 Ψ mc 〈E SNa m whereE SNa m is the “zero current” or “reversal” potential. Over the range of values of Ψ mc encountered under physiological conditions the cotransporter may be modeled as a conductance in series with an electromotive force so thatI SNa m =g SNa m (E SNa m − Ψ mc ) whereg SNa m is the contribution of this mechanism to the conductance of the apical membrane and is “near constant”. In several instancesI SNa m “saturated” at large hyperpolarizing or depolarizing values of Ψ mc . The values ofE SNa m determined in the presence of 1, 5, and 15mm galactose strongly suggest that if the Na-galactose cotransporters are kinetically homogeneous, the stoichiometry of this coupled process is unity. Finally, the shapes of the observedI−V relations are consistent with the predictions of a simple kinetic model which conforms with current notions regarding the mechanico-kinetic properties of this cotransport process.

Membrane Biol. 73:157–175, 1983) the current-voltage (I–V) relations of the Na-entry step across the apical membrane of short-circuitedNecturus urinary bladder in the presence of varying mucosal Na concentrations are (i) time-independent between 20–90 msec and (ii) conform to the Goldman-Hodgkin-Katz constant field flux equation for a single cation over a wide range of voltages. In contrast, theI–V relations of the basolateral membrane under these conditions are (i) essentially linear between the steady-state, short-circuited condition and the reversal potential (E s ); and (ii) are decidedly time-dependent withE s increasing and the slope conductance,E s , decreasing between 20 and 90 msec after displacing the transepithelial electrical potential difference. Evidence is presented that this time-dependence cannot be attributed entirely to the electrical capacitance of the tissue. The values ofg s determined at 20 msec are linear functions of the short-circuit current,I sc, confirming the relations reported previously, which were obtained using a more indirect approach. The values ofE s determined at 20 msec are significantly lower than any reasonable estimate of the electromotive force for K across the basolateral membrane, indicating that this barrier possesses a significant conductance to other ions which may exceed that to K. In addition, these values increase linearly with decreasingI sc and approach the value of the electrical potential difference across the basolateral membrane observed when Na entry across the apical membrane is blocked with amiloride or when Na is removed from the mucosal solution. A possible explanation for the time-dependence ofE s andg s is offered and the implications of these findings regarding the interpretation of previous microelectrophysiologic studies of epithelia are discussed.

    Notizen: Summary The electrophysiologic properties of rabbit colonic epithelial cells were investigated employing microelectrode techniques. Under open-circuit conditions, the transepithelial electrical potential difference (PD) averaged 20 mV, serosa positive, and the intracellular electrical potential (ψ mc ) averaged −32 mV, cell interior negative with respect to the mucosal solution; under short-circuit conditions,ψ mc averaged −46 mV. The addition of amiloride to the mucosal solution abolishes the transepithelialPD and active Na transport, andψ mc is hyperpolarized to an average value of −53 mV. These results indicate that Na entry into the mucosal cell is a conductive process which, normally, depolarizesψ mc . The data obtained were interpreted using a double-membrane equivalent electrical circuit model of the “active Na transport pathway” involving two voltage-independent electromotive forces (emf's) and two voltage-independent resistances arrayed in series. Our observations are consistent with the notions that: (a) The emf's and resistances across the mucosal and baso-lateral membranes are determined predominantly by the emf (64mV) and resistance of the Na entry process and the emf (53 mV) and resistance of the process responsible for active Na extrusion across the baso-lateral membranes: that is, the electrophysiological properties of the cell appear to be determined solely by the properties and processes responsible for transcellular active Na transport. The emf of the Na entry process is consistent with the notion that the Na activity in the intracellular transport pool is approximately one-tenth that in the mucosal solution or about 14mM. (b) In the presence of amiloride, the transcellular conductance is essentially abolished and the total tissue conductance is the result of ionic diffusion through paracellular pathways. (c) The negative intracellular potential (with respect to the mucosal solution) is due primarily to the presence of a low resistance paracellular “shunt” pathway which permits electrical coupling between the emf at the baso-lateral membrane and the potential difference across the mucosal membrane; in the absence of this shunt, the “well-type” electrical potential profile characteristic of rabbit colonic cells would be ‘converted’ into a “staircase-type” profile similar to those reported for frog skin and toad urinary bladder by some investigators.

    Notizen: Summary In this paper we employ the data described in the previous paper (I) to derive the current-voltage (I-V) relations of the basolateral membrane, the amiloride-insensitive “leak” pathway across the apical membrane, and the parallel pathways across rabbit descending colon. The results indicate that: a) The resistance of the basolateral membrane is independent of the electrical potential difference across that barrier over the range −8 to 67 mV and averaged 195 Ωcm2. The electromotive force across this barrier averaged 50 mV under control conditions and 48 mV in the presence of amiloride. The origin of this difference is discussed. b) The resistance of the parallel pathways averaged 351 Ωcm2 and was independent of the transepithelial electrical potential difference over the range −170 to +90 mV. The conductance of these pathways can be reasonably well accounted for by the partial ionic conductances of Na, K and Cl reported previously. c) The resistance of the amiloride-insensitive pathway across the apical membrane averaged 1667 Ωcm2 and the electromotive force across this pathway averaged −51 mV. These values are in excellent agreement with those determined by others. The ionic nature of this “leak” pathway remains to be elucidated.
