    Notes: Summary Pullulan, a microbial polysaccharide, was employed with polyethylene glycol (PEG) to form an aqueous two-phase system. The phase diagram of the PEG-Pullulan systems as well as the partition of enzymes depended strongly on the molecular weight of PEG. This behavior provided a basis for the separation of cellulase from β-galactosidase. The extractive hydrolysis of lactose, using a commercial preparation of β-galactosidase, could also be performed in a PEG-Pullulan system (16% PEG 1400, 14% Pullulan). The enzyme was effectively contained in the bottom phase while a high percentage of the glucose produced could be removed in the top phase. The enzyme activity was preserved very well after several repeated hydrolyses. Furthermore, it was demonstrated that the PEG-Pullulan system was capable of retaining Trichoderma reesei in the bottom phase, allowing the production of cellulase which could be intermittently removed in the top phase. Such a system permitted a semicontinuous production scheme although the enzyme production was observed to decrease after five cycles. In view of the low cost and low viscosity of pullulan, this polysaccharide could be considered a suitable replacement for dextran, a component widely used in aqueous two-phase formulation.

    Notes: Using a well-mixed and temperature-led vessel, the diffusion characteristics of various solutes into spherical κ-carrageenan gel beads were experimentally investigated. The diffusion coefficient of glucose was markedly affected by the glucose concentration and the operating temperature. In all cases the diffusivity obtained was noticeably smaller than that of glucose in pure water. The experimental data also indicated an inverse relationship between the diffusivity and the polymer concentration used in the gel preparation. As well, the glucose diffusivity was affected by the presence of other solutes in the glucose solution. Electrolytes such as ammonium sulfate, KCl, and CaCl2 were observed to enhance the diffusion coefficient. On the other hand, the addition of arginine or bovine serum albumin had an adverse effect on the diffusivity. No diffusion of albumin into the gel beads was observed, and such a solute created a significant mass transfer resistance during the diffusion process.

    Notes: An amperometric biosensor for hypoxanthine was constructed by forming a layer of crosslinked xanthine oxidase on a platinum electrode, followed by electropolymerization of a submonolayer film of resorcinol and para-diaminobenzene. The fabricated electrodes were evaluated for speed of response, sensitivity, and reusability. Optimal performance was obtained with enzyme-based electrodes sparsely covered with film which was formed by electropolymerization in less than 6 min. The resulting electrodes exhibited linear response to hypoxanthine in the. range 5-300 μM with a response time of 2 min. Application of the biosensor in monitoring hypoxanthine content of fish extracts yielded results which agreed well with spectrophotometric assays using soluble xanthine oxidase. The biosensor was stable for 60 days when stored at 4°C in phosphate buffer and it could be used continuously for 6 h with over 50 assays.

    Notes: The dielectric, dynamic mechanical, and melting (denaturation) behavior of reconstituted collagen has been investigated. Dynamic mechanical properties are reported for the range of temperature from -100 to 220°C. Dielectric properties are reported for the range of temperature from -120 to 90°C. Possible origins of dynamic mechanical and dielectric relaxations are discussed. Effect of moisture content on mechanical and dielectric behavior is also presented. The melting process of collagen immersed in diluents as well as that of dry collagen were studied by the techniques of hot-stage polarizing microscopy, differential thermal analysis, thermal mechanical analysis, and small-angle light scattering. It was concluded that the melting point of dry collagen is about 217°C.

    Notes: Cyclodextrin-modified capillary electrophoresis has been developed for separation and analysis of benzene and its derivatives. The procedure used a mixture of negatively charged sulfobutyl ether-β-cyclodextrin (SBβCD) and neutral hydroxypropyl-β-cyclodextrin (HPβCD) to effect differential distribution (partitioning) of the aromatic hydrocarbons between the buffer and CD phases. In 80 mM phosphate buffer, containing 15 mM SBβCD and 5 mM HPβCD, benzene, toluene, ethyl benzene and three xylene isomers (BTEXs) were well resolved with a number of theoretical plates well above 100 000, for 50 cm of effective length. Some halogenated benzenes were also observed to separate well from the BTEX components to indicate their suitability as an internal standard for BTEX analyses. Equilibrium complexation models were used for investigating the effect of the cyclodextrin(s) at different concentrations and temperature on the electrophoretic mobility.
