    Notizen: : Renewed (de novo) synthesis of methane gas was shown to occur when samples of lake sediment were dispersed on glass beads and incubated in a helium atmosphere at 23°C. Under the above conditions, sediment samples from hardwater and softwater lakes generated up to 440 nanomoles and 80 nanomoles per ml of sediment per two days, respectively. At the time of collection, sediment samples possessed approximately similar amounts of “native” methane. Nitrate, sulfate, and acetylene were shown to suppress methane synthesis by sediment incubated as described.

    Notizen: Abstract Kjeldahl nitrogen (N), total N and forms of inorganic N (ammoniacal (NH4)-N, nitrate (NO3)-N and nitrite (NO2)-N) were measured in a range of animal manures. The manures include fresh samples of poultry manure, sheep manure, horse manure, dairy slurry and pig slurry and composted poultry manure. Kjeldahl N was measured by standard micro-Kjeldahl digestion. For total N measurements, NO3-N and NO2-N were recovered during Kjeldahl digestion by pretreatments with various oxidizing and reducing agents. Inorganic forms of N were measured by extraction with 2M KCl solution. Kjeldahl digestion alone allowed measurement only of organic N and NH4-N. Amongst various modifications to the Kjeldahl, pretreatment with either acidified (H2SO4) Zn-CrK(SO4)2 or acidified (H2SO4) reduced Fe achieved complete recovery of NO3-N. Nitrite N was only recovered by first oxidising the NO 2 - to NO 3 - with KMnO4 followed by reduction to NH4-N with acidified (H2SO4) reduced Fe. More than 95% of the total N in fresh animal manure was present as organic N and NH4-N which were recovered by the standard Kjeldahl digestion. In the case of fresh manures there was no difference between the amount of total N measured by the Kjeldahl digestion and its modified methods. However composting of poultry manure or drying of poultry manure, pig slurry and dairy slurry resulted in an increase in NO3-N which was not recovered during Kjeldahl digestion alone. Under these conditions the total N could be measured by pretreating the samples with KMnO4 and reduced Fe prior to Kjeldahl digestion. Drying of animal manures caused a decrease in organic N and NH4-N, especially in poultry, pig and dairy manures. There was a slight increase in NO3-N; but most of the decrease in N content with drying was attributed to the volatilization loss of ammonia (NH3). Amongst various drying methods examined air drying caused maximum loss of N as NH3 whereas freeze drying caused minimum loss of N. This suggests that fresh animal manures can be freeze dried for analysis of N which causes minimum loss of N.

    Notizen: Summary An incubation experiment was conducted to study the changes that occur in the K status of soil due to earthworm activity. Samples of Tokomaru silt loam soil were inoculated with the common pasture earthworm species Aporrectodea caliginosa and incubated for 21 days. Aliquots of moist soil were analyzed for exchangeable K by leaching with neutral molar ammonium acetate at 1:50 soil solution ratio. Extraction with boiling 1 M nitric acid at 1:100 soil solution ratio for 20 min was used to determine available non-exchangeable K. The results indicated that the exchangeable K content increased significantly due to earthworm activity but nitric acid-extractable K did not change significantly. It is inferred that earthworms increase the availability of K by shifting the equilibrium among the forms of K from relatively un-available forms to more available forms in the soil chosen for the study.

    Notizen: Abstract An incubation experiment was conducted to study the changes that occur in potassium availability and other soil properties with ingestion of soil by earthworms. Two soils were used. Raumai soil with high non-exchangeable K and Milson soil with low non-exchangeable K were incubated with two species of earthworm, Aporrectodea caliginosa and Lumbricus rubellus, for 8 weeks. The casts and soil samples were analysed for exchangeable K, Ca, Mg, Na, and H, pH, organic C, and texture. The results indicated that in Raumai soil, the exchangeable K levels of the casts of both earthworm species were significantly higher than for the control soil, the effect being more marked for L. rubellus than for A. caliginosa. In Milson soil, the exchangeable K levels were significantly lower in the casts of both types of earthworm than in the control soil. The nitric acid-extractable K of the soil and casts was not markedly different for either soil type, but available non-exchangeable K values were significantly higher for the casts of L. rubellus from Milson soil than for the noningested Milson soil. In Raumai soil, the exchangeable Ca was higher in the casts of L. rubellus, exchangeable Mg and H were reduced, and exchangeable Na did not change markedly in the cast compared to the control soil. For Milson soil, the casts contained lower exchangeable Ca and H but higher Na and Mg than the control. The casts of both species of earthworm had significantly higher pH values for both soil types. There was no marked difference in the organic C content of the control soil and cast samples for Milson but a reduction in the casts of A. caliginosa for the Raumai soil. Finer fractions increased in the casts of both earthworm species in both soil types.

    Notizen: Conclusions Extraction of ATP from microorganisms in desert soil is best carried out using the n-butanol-octanol method. Using this method, one obtains an average ATP-recovery of 68% under widely varying soil moisture and textural conditions. We have also shown that the half-life of non-microbial ATP in soil is not more than one hour and thus the possibility of a build-up of non-microbial ATP in desert soil is unlikely. This method can be used for rapid survey of the microbial content in soil environments with a minimum of soil disruption. It is also a valuable tool for investigating the effect of various environmental factors on fluctuations in soil microflora content.

    Notizen: Summary The respiratory activity of a cropland soil which received between 100 and 120 tonnes (dry) of municipal sludge ha−1 annually over a five year period, was examined manometrically. The soil contained an average of 34 mg Cd kg−1. Compared to soil from conventionally managed fields, the biological consumption of gaseous oxygen by sludge-treated soil was four times greater than its untreated conventionally fertilized counterpart. This enhanced level of biological activity was restricted to the surface 25 cm of soil at the disposal site. There was no indication that increased soil cadmium levels had impaired the overall respiratory activity of the sludge treated soil.

A major distinction between the two organisms was observed to be the inability of the non-nodulating strain to grow in the root-tip region ofT. pratense. A small quantitative difference in antigen composition but no significant qualitative or quantitative differences in the extracellular polysaccharides of the two bacteria were noted. The non-nodulating rhizobium could not be induced to nodulate seedlings ofT. pratense either in the presence of nodulating rhizobia or after ultraviolet treatment of the bacteria.

    Notizen: Abstract Discoid granules were isolated from the tegument of adult Schistosoma mansoni by differential centrifugation of freeze-thaw extracts. Comparison of proteins and glycoproteins from discoid granules, surface membranes and the soluble fraction implied that discoid granules and the surface membrane were functionally linked. The results are discussed in terms of discoid granule function and surface membrane organisation.
