    Notes: Abstract. Purified and crude phosphoenolpyruvate carboxylase from the CAM plant Kalanchoë daigremontiana Hamet et Perrier (Bryophyllum diagremontianum) was assayed at temperatures between 10 and 45° C. The optimum temperature of the enzyme activity changed with substrate availability and effector concentration in the assay. l-malate inhibited the enzyme activity and lowered the optimum temperature. Glucose-6-phosphate raised the optimum temperature to 43°C. Km values for phosphoenolpyruvate increased with assay temperature from 0.12 mol m-3 at 15° C to 0.36 molm−3 at 35° C. Inhibition by malate increased with temperature and acidity of the assay. In the crude enzyme 50% of control activity was inhibited by 1.65 mol m-3 malate at 15° C and by 0.5 mol m-3 at 35° C (at pH 7.0). With purification malate sensitivity was lost (Ki values for malate at least 10 times higher). The shift in optimum temperatures for PEP-carboxylase activity thus results from changes in the kinetic parameters with temperature and allosteric effectors. The often low optimum temperatures for CO2 fixation observed in nature may thus be the result of substrate and effector concentrations in the cytoplasm and the antagonistic effect of temperature on substrate affinity and effector efficiency on phosphoenolpyruvate carboxylase.

    Notes: Summary 75.4 MHz 13C NMR CP/MAS spectra allowed the quantification of the secondary structure of (γOMe-L-Glu)n and (γOBzl-L-Glu)n. (OMe-L-Glu)n contains α-helix and β-sheet structures the ratio of which varies considerably with the average degree of polymerization ( $$\overline {{\text{DP}}} $$ ) of the samples and with the nature of the reaction medium. At $$\overline {{\text{DP}}} $$ ≥20 (γOBzl-L-Glu)n contains more than 95% α-helix structure regardless of $$\overline {{\text{DP}}} $$ and reaction medium. The difference between both polyglutamates is explained by different molecular weight distributions resulting from aggregation and precipitation of oligo (γOMe-L-Glu)n in the ß-sheet form.

    Notes: Summary Various copolypeptides with a two block sequence are prepared by batchwise copolymerizations of amino acid N-carboxyanhydrides (NCAs) using primary amines as initiators. An α-helix forming amino acid and a ß-sheet forming amino acid were combined in the two blocks and the mutual influence of the two secondary structures was investigated by means of 13C nmr crosspolarization magic-angle spinning (CP/MAS) spectra. It was found that the second (succeeding) block adopts partially the secondary structure of the first block.

    Notes: Summary 50.3 MHz 13C NMR cross-polymerization/magic angle spinning spectra of the raw silk of the mulberry silk moth (bombyx mori) and of crystalline silk fibroin were compared with those of synthetic copolypeptides of alanine and glycine. In crude silk, but not in crystalline fibroin , tyrosine, valine,aspartic acid and glutamic acid were detected in addition to alanine, glycine and serine. Furthermore, crude silk obviously contains ca. 10 % of an α-helix fraction which mainly consists of Ala-units. A small α-helix fraction (ca. 5 %) was also detected in the raw silk of the tussah silk moth (antheraea mylitta).

    Notes: Summary 75.46 MHz 13C-NMR CP/MAS spectra of solid poly(1-alanines) with various degrees of polymerization were measured. The signals of all three carbons are split into two peaks. One peak presents the α-helix structure and one the pleated sheet form. Equimolar mixtures of helical poly(L-alanine) with poly(glycine) and poly(L-valine) were measured under various conditions. It was found that the peak areas are proportional to the mole ratios of the different secondary structures and neither a mismatch of the Hartmann-Hahn conditions nor variations of the mixing time or the pulse interval (relaxation delay) have a significant influence on the intensity ratios. Hence, a quantitative evaluation of the 13C NMR spectra with respect to the secondary structure is feasible.
