The molecular weight of kallikrein inactivator (KI) is determined in the ultracentrifuge, by the ARCHIBALD and TRAUTMANN methods. The molecular weight of highly purified KI is found to be 6500 ± 200 and is independent on the pH values of the solutions as well as on the KI concentration.It follows from the sedimentation, diffusion and viscosity measurements that the molecules of KI have non-spherical forms.The KI molecules associate not only with enzyms, but also with low molecular substances such as purines, pyrimidines, ADP, ATP. High valent anions (e.g. citric acid, pyro- and metaphosphoric acids) have little effect on such association. The bivalent cations, calcium, magnesium, and zinc, do not cause the dimerisation.The interaction between KI and the low molecular substances follows the law of mass action.Equations for the dependence of molecular weight and of the rate of association on the concentrations of protein and the complex formed are derived. The equilibrium constants of KI molecules associated in the presence of adenin and thymin are experimentally determined.The molecular weight distribution curye of KI is obtained from TRAUTMANN diagram at its isoelectrical point (pH 10.5). The 1% solution of KI used in the earlier experimental work contains mostly the molecular weight of 6500 and less of 13000.Fractionation and partial separation of these components are possible by means of dialysis and gelfiltration.A trace of foreign substance having a similar UV-spectrum as adenosin is found in the substance used in our earlier experimental work. Although present in KI only in traces, it causes association. The molecular weight of this KI depends therefore on the pH of the solution as well as on the KI concentrations. In acidic and alkaline solution it is found to have a minimum molecular weight; while in neutral solution a maximum value is obtained.By diluting a neutral solution of this substance, the molecular weight decreases at KI concentration smaller than 0.2% and approaches 6500. The molecular weight in solution of pH 12.5 increases with time and reaches a constant value of 13000 after standing for several days. The change in KI molecular weight caused by differing pH values of the solutions are fully reversible for solutions having pH values between 1.1 and 10.5. For pH greater than 12, the molecular weights are partly irreversible.

Das Molekulargewicht des Kallikrein-Inaktivators (KI) wurde mittels der Ultrazentrifugenmethode nach ARCHIBALD und TRAUTMANN bestimmt. Das Molekulargewicht der hoch gereinigten Präparate ist vom pH der Lösung und von der Konzentration unabhängig und beträgt 6500 ± 200.Aus Sedimentations-, Diffusions- und Viskositätsmessungen ergibt sich, daß das Molekül des KI eine merklich von der Kugelform abweichende Gestalt besitzt.Der KI bildet nicht nur mit Enzymen, sondern auch mit niedermolekularen Stoffen Assoziate; solche Stoffe sind z. B.: Purine, Pyrimidine, ADP, ATP und in geringem Maßemehrwertige Anionen (z. B. Citronensäure, Pyro- und Metaphosphorsäure). Die zweiwertigen Kationen des Calciums, Magnesiums und Zinks bewirken dagegen keine Dimerisierung.Die Wechselwirkung des KI mit den niedermolekularen Stoffen ist eine Gleichgewichts-reaktion und folgt dem Massenwirkungsgesetz.Gleichungen für die Abhängigkeit des mittleren Molekulargewichts und des Assoziationsgrades von der Eiweißkonzentration und der Konzentration des Komplexbildners werden aufges ellt. Die Gleichgewichtskonstante der Assoziationsreaktion wird für Adenin und Thymin ermittelt.Aus TRAUTMANN-Diagrammen wurde beim isoelektrischen Punkt des KI (pH 10, 5) die Molekulargewichtsverteilung ermittelt. Die früher untersuchten Präparate enthalten in 1-proz. wäßriger Lösung vor allen Moleküle vom Molekulargewicht 6500, in geringer Menge ferner Assoziate vom Molekulargewicht 13000.Durch Dialyse und Gelfiltration ist eine Fraktionierung und partielle Trennung der Komponenten möglich.In den bereits früher untersuchten Präparaten wurde ein Stoff mit einem adenosinähnlichen UV-Spektrum gefunden, der in sehr kleiner Konzentration eine Assoziation des KI bewirkt. Das Molekulargewicht dieser unreinen Präparate ist daher sowohl vom pH der Lösung als auch von der Konzentration abhängig. In saurer und alkalischer Lösung besitzen diese Präparate ein Mindest-Molekulargewicht; in neutraler Lösung erreicht das Molekulargewicht einen maximalen Wert.Beim Verdünnen einer neutralen Lösung dieser Präparate verringert sich das Molekulargewicht bei Konzentrationen unter 0,2% und nähert sich dem Wert 6500. In alkalischer Lösung bei pH 12,5 nimmt das Molekulargewicht mit der Zeit zu und erreicht nach einigen Tagen einen konstanten maximalen Wert von 13000. Die durch pH-Änderungen verursachten Molekulargewichtsänderungen sind im pH-Bereich von 1,l bis 10,5 vollständig reversibel, über pH 12 dagegen teilweise irreversibel.
