    Description: In this study Amplified Fragment Length Polymorphism (AFLP) was applied to species identification of bester hybrids. Hybrids identification was performed by comparison of electrophoresis profiles with parental species. The simultaneous occurrence of diagnostic bands fixed in the parental species, genetic distance and identification and cluster analyses (UPGMA) allow a correct identification. We used 8 primer combinations (Eco+3, Mse+4) and a total of 250 bands (size range 40-1000 bp) were generated. Primer combinations of (E-AAT, M-CGAT) and (E-AAG, M-CGAT) produced diagnostic bands in hybrids and parental species. Moreover, the results of genetic identification showed that Bester hybrids are more similar to beluga (Huso huso) (0.68) in comparison with sterlet (Acipenser ruthenus) (0.45).The results suggested that this technique could be suitable for precise identification of species and inter-generic hybrids like bester.

    Description: The ionic contents, pH and osmolality of seminal plasma and their relationships to sperm motility indices (sperm motility duration and percentage of motile sperm) were evaluated in bester. Semen was collected from six bester specimens and their seminal plasma was evaluated. The seminal plasma contained 54.5±2.8 mM sodium, 22.33±2.5 mM chloride, 7.1±1.2 mM potassium, 3.55±0.6 mM calcium and 0.58±0.09 mM magnesium. The result shows that Na+, Cl- and K+ (54.5±2.8, 22.33±2.5 and 7.1±1.2 mM, respectively) were predominant ions in bester seminal plasma like other sturgeons. However the concentrations of K+, Ca2+ and Cl- in bester seminal plasma were higher than those reported in beluga and sterlet as parental species. The mean pH and osmolality of seminal plasma were 8.09±0.34 and 127.6±20.8 mOsm kg-1, respectively. A significant negative regression was observed between osmolality and percentage motility (r=- 0.893).

    Description: The objective of this study was to analyse the population genetic structure of the Persian sturgeon (Acipenser persicus) in Sefidrud and Gorganrud rivers watershed based on the characterization of microsatellite markers during 2006 – 2008. 100 samples of Persian sturgeon were collected from two regions. Four microsatellite loci ( Ls 68 , Spl 168 , Spl 173 and Afu 68 ) were analyzed for the molecular characterization of this species which resulted in polymorphic patterns. DNA bands were analysed using Biocapt and GenAlex software package. A total of 109 alleles were observed of which the maximum number of alleles (17) were found in Spl168 locus which belonged to sturgeons from Sefidrud river,s watershed and the minimum number of alleles (10) in Ls68 locus belonging to the sturgeons from Gorganrud river,s watershed. Results of microsatellite analysis revealed that the differences between samples of two regions were not statistically significant (p〉0.05), neither for the average number of alleles per locus nor for observed heterozygosities. The calculated Fst and Rst between two regions was 0.07 and 0.17 showing that the genetic difference was significant (p〈 0.01). Samples from Sefidrud river,s watershed in Spl173, Afu68 and Spl168 loci and samples of other regions in Afu68 and Spl168 loci were at Hardy-Weinberg equation. The genetic distance was calculated as 0.4 which represents a significant genetic difference between samples of two studied areas. I n conclusion, this study suggests that the Persian sturgeons in two regions of the southern part of the Caspian Sea are genetically differentiated, therefore fisheries management of these unique stocks for restocking and conservation of gene pools is highly recommended.

    Description: The relationship between morphometric characteristics, motile spermatozoan percentage and sperm motility duration among male breeders of Rutilus frisii kutum in four different rivers (Shirud, Khoshkrud, Sefidrud and Chalevand) of the southwestern part of the Caspian Sea and water osmolarity, pH and temperature in four region of each river (estuary, river entrance to sea, artificial reproduction site and upstream) were considered. The mean motility duration time was 37.2~c4.2, 41.4~c3.9, 52.9~c2.1 and 47.8~c3.4 in the four rivers respectively. Also, the percentage of motile cells was found to be 49~c4.1, 58.2~c6.8, 74~c4.8 and 68.1~c5.9 % for the rivers respectively. The mean osmolarity, temperature and pH were 71.7~c66, 62.5~c85.5, 40.2~c34.2, 36.7~c34.4 mOsml/l, 14, 15, 13.6~c0.2 , 14 degree C and 7.6~c0.1, 7.2~c0.1, 7.4~c0.1 and 7.2~c0.1 respectively. Our findings showed that there is a positive relationship between motile spermatozoan percentage and duration of motility. With the decrease in river water osmolarity from estuaries up to the river upstream, the motile spermatozoan percentage and their duration increased. According to anadromous behaviour of the fish, with increasing distance from artificial breeding sites toward river upstream, quantity of spermatozoa and spermatozoan activity increased enhancing success in fertiliza-tion attempts.

    Description: Hybrid Bester sturgeon (female Beluga x male Sterlet) was produced for the first time in Iran using sperm from a male Acipenser ruthenus and eggs of a female Huso huso in Shahid Marjani Sturgeon Porpagation Center (Agh Ghala, Golestan province). Bester fries and those of Beluga as control treatment weighed 490mg and 377mg respectively after about one month and were transported to International Sturgeon Research Institute (Rasht). All fishes were fed by artificial concentrated food (48-50% protein and 1547% fat) after a period of feeding on Artemia and Daphnia. Sorting was carried out according to weight increase for both fishes. Results showed that the weight increase in warmer months (water temperature 18-28°C) was higher than the colder months and ceased at 6-12°C. The trend in weight increase was of the same pattern for both fishes. There were no significant differences between fish length up to 5 months of rearing but afterwards, Bester showed a higher rate of length increase compared to that of the Beluga. The length increase rate for both fishes was sharper during the first year than the second year. Fish length increase was ceased at 6.841.4°C which coincided with the increase in fish weight.

    Description: Motility of spermatozoa was studied on 12 and 7 specimens of Acipenser persicus and A. stellatus, respectively. The density measured to be 2.22±0.65x109ml-1 in A. persicus and 2.21±0.55x109ml-1 in A. stellatus. Semen samples were diluted with two extenders containing tris 118mM, sucrose 23.4mM, pH=8, egg yolk (20%), dimethyl sulfoxide (15%) and penicillin potassium (500IU/ml) and biociphus extender containing glycerol as a cryoprotectant at a ratio of 1:1 and then transferred to 0.5ml straws and frozen in a computer controlled low temperature apparatus and stored in liquid nitrogen for one week. To study fertilizing ability, the spermatozoa were then used to inseminate eggs after thawing. Mean sperm motility in fresh spermatozoa was 86.6% in A. persicus and 73.75% in A. stellatus which decreased to 32.2% (P〈0.001) and 37.5% (P〈0.001) in frozen spermatozoa, respectively. Also mean fertilization rate decreased from 90.4% to 30.7% in A. persicus and from 72% to 36.8% in A. stellatus.

    Description: Short term preservation of spermatozoa of salmonid fishes for practical use, increasing of reproduction potentials and reducing of expenses, has an important role in hatcheries. In this study spermatozoan motility and fertilization rate of 29 commercially cultured rainbow trout were examined. Semen was collected by abdominal stripping at the volume of 4.49±1 (mean±SD) ml and sperm concentration was determined by heamocytometer following 1:600 dilution with saline solution (0.9% NaCl). To find the effects of preservation time on fertilization capacity, samples were diluted with extender (Glucose 54gL-1, Tris 0.5gL-1, DMSO 9% , Egg yolk 20%) and held at 3 different cool conditions : room temperature (+15°C), refrigerator (+5°C) and crashed ice (+ 1-2°C) and sperm motility followed up each 30 mins until complete ceasation. Fertilization, hatching and larval stage rate were determined after 24, 48 and 72 hours. Results showed that the motility of sperm samples held at 15 °C and 5°C were completely ceased after 2 and 24 hours respectively. However it lasted up to 144 hours at1-2°C.Fertilization, hatching and larval stage rates of sperm samples hold at 1- 2°C were 68.45% , 62.1% and 58.2% (after 24 hours), 53.2% , 43.2% and 40.9% (after 48 hours) and 36.6% , 31.6% and 23.7% after 72 hours, respectively, and all showed a significant decrease compared with control (90.32%, 83.27% and 81.12% respectively). Comparing the results showed that holding samples in test tubes at 1-2°C is the best method for short term storage of rainbow trout spermatozoa.
