    Description: 〈p〉Soils underpin terrestrial ecosystem functions, but they face numerous anthropogenic pressures. Despite their crucial ecological role, we know little about how soils react to more than two environmental factors at a time. Here, we show experimentally that increasing the number of simultaneous global change factors (up to 10) caused increasing directional changes in soil properties, soil processes, and microbial communities, though there was greater uncertainty in predicting the magnitude of change. Our study provides a blueprint for addressing multifactor change with an efficient, broadly applicable experimental design for studying the impacts of global environmental change.〈/p〉

    Description: The minor outer capsid protein P2 of Rice dwarf virus (RDV), a member of the genus Phytoreovirus in the family Reoviridae , is essential for viral cell entry. Here, we clarified the structure of P2 and the interactions to host insect cells. Negative stain electron microscopy (EM) showed that P2 proteins are monomeric and flexible L-shaped filamentous structures of ~20 nm in length. Cryo-EM structure revealed the spatial arrangement of P2 in the capsid, which was prescribed by the characteristic virion structure. The P2 proteins were visualized as partial rod-shaped structures of ~10 nm in length in the cryo-EM map and accommodated in crevasses on the viral surface around icosahedral 5-fold axes with hydrophobic interactions. The remaining disordered region of P2 assumed to be extended to the radial direction towards exterior. Electron tomography clearly showed that RDV particles were away from the cellular membrane at a uniform distance and several spike-like densities, probably corresponding to P2, connecting a viral particle to the host cellular membrane during cell entry. By combining the in vitro and in vivo structural information, we could gain new insights into the detailed mechanism of the cell entry of RDV.

    Description: Mass concentrations of black carbon (BC) in air ( M BC ) and rainwater ( C BC ) in the East China Sea were measured at Hedo on Okinawa Island, Japan from April 2010 to March 2013. The monthly averaged M BC and C BC showed marked seasonal variations, being highest in spring (0.32 ± 0.13 µg m −3 and 92 ± 76 µg L −1 , respectively), and lowest in summer (0.06 ± 0.03 µg m −3 and 8.0 ± 4.1 µg L −1 , respectively). The high M BC and C BC in spring were associated with transport of air masses from the Asian continent by northwesterly winds. The BC wet deposition flux ( F BC ), estimated as the product of C BC and precipitation amount, also showed a distinct seasonal variation. The monthly average F BC during the four spring seasons (16.8 ± 6.7 mg m −2  month −1 ) was about three times higher than the annual average F BC (5.5 ± 9.9 mg m −2  month −1 ) owing to the high C BC and precipitation amount in spring. As a result, about 76% of the annual BC deposition occurred in spring on average. The F BC in spring is comparable to the average BC net flux in North China, indicating the importance of precipitation over the East China Sea as a sink of BC transported from North China. In summer, C BC values were correlated with M BC for rain events associated with local convective activity, as identified by the convective available potential energy. A one-dimensional thermodynamic model successfully explained the relation between C BC and M BC .

    Description: CD4(+) T helper cells are well known for their role in providing critical signals during priming of cytotoxic CD8(+) T lymphocyte (CTL) responses in vivo. T-cell help is required for the generation of primary CTL responses as well as in promoting protective CD8(+) memory T-cell development. However, the role of CD4 help in the control of CTL responses at the effector stage is unknown. Here we show that fully helped effector CTLs are themselves not self-sufficient for entry into the infected tissue, but rely on the CD4(+) T cells to provide the necessary cue. CD4(+) T helper cells control the migration of CTL indirectly through the secretion of IFN-gamma and induction of local chemokine secretion in the infected tissue.     Description: It has been thought that the lunar highland crust was formed by the crystallization and floatation of plagioclase from a global magma ocean, although the actual generation mechanisms are still debated. The composition of the lunar highland crust is therefore important for understanding the formation of such a magma ocean and the subsequent evolution of the Moon. The Multiband Imager on the Selenological and Engineering Explorer (SELENE) has a high spatial resolution of optimized spectral coverage, which should allow a clear view of the composition of the lunar crust. Here we report the global distribution of rocks of high plagioclase abundance (approaching 100 vol.%), using an unambiguous plagioclase absorption band recorded by the SELENE Multiband Imager.     Description: Plasmacytoid dendritic cells (pDCs) detect viruses in the acidified endosomes by means of Toll-like receptors (TLRs). Yet, pDC responses to certain single-stranded RNA (ssRNA) viruses occur only after live viral infection. We present evidence here that the recognition of such viruses by TLR7 requires transport of cytosolic viral replication intermediates into the lysosome by the process of autophagy. In addition, autophagy was found to be required for the production of interferon-alpha by pDCs.     Description: The inside of Shackleton Crater at the lunar south pole is permanently shadowed; it has been inferred to hold water-ice deposits. The Terrain Camera (TC), a 10-meter-resolution stereo camera onboard the Selenological and Engineering Explorer (SELENE) spacecraft, succeeded in imaging the inside of the crater, which was faintly lit by sunlight scattered from the upper inner wall near the rim. The estimated temperature of the crater floor, based on the crater shape model derived from the TC data, is less than approximately 90 kelvin, cold enough to hold water-ice. However, at the TC's spatial resolution, the derived albedo indicates that exposed relatively pure water-ice deposits are not on the crater floor.     Description: Twenty years after the proposal that pattern recognition receptors detect invasion by microbial pathogens, the field of immunology has witnessed several discoveries that have elucidated receptors and signaling pathways of microbial recognition systems and how they control the generation of T and B lymphocyte-mediated immune responses. However, there are still many fundamental questions that remain poorly understood, even though sometimes the answers are assumed to be known.     Description: We have developed a focusing system for extreme ultraviolet light produced by high-order harmonic generation. An ellipsoidal mirror with a precise surface shape was fabricated and installed into the focusing system. A rigid mirror manipulator and a beam profiler were employed to perform precise and stable mirror alignment. As a demonstration of the focusing performance, high-order harmonics in the wavelength range of 13.5–19.5 nm were successfully focused into a 2.4 × 2.3 μ m 2 spot.
