    Notizen: Structures of the trifunctional crosslink molecules, pyridinoline, deoxypyridinoline and glucosylgalactosyl-pyridinoline, have been characterized by positive-ion liquid secondary-ion mass spectrometry (LSIMS) and collision-induced dissociation with scanning-array four-sector tandem mass spectrometry. It has been found that: (i) the naturally occurring cations (Cat+) are readily desorbed by positive-ion LSIMS utilizing glycerol + thioglycerol (1:1) as liquid matrix and dominate the spectra, facilitating accurate mass measurement; (ii) changing the matrix to 3-nitrobenzyl alcohol leaves little evidence of the cation, and (iii) collision-induced dissociation of the cations (Cat+) of pyridinoline, deoxypyridinoline and glucosylgalatosyl-pyridinoline gives a wealth of fragmentation for structure elucidation. These fundamental studies of tandem mass spectra of the known trifunctional crosslink molecules provided valuable structure-related information, which help in the identification of the tissue-specific peptides containing these crosslinks. Artefact ions in these tandem mass spectra are remarkably abundant.

    Notizen: Fast atom bombardment mass spectra of a series of phospholipids are reported. Abundant molecular ions are noted together with fragment ions which provide structural information about the acyl and phosphate ester groupings. The application of fast atom bombardment mass spectrometry to the analysis of complex phospholipid(phosphatidylcholine) mixtures is discussed.

    Notizen: The behaviour of glucosinolates and desulphoglucosinolates under fast atom bombardment conditions has been investigated. Relatively little fragmentation was noted in the mass spectra of these compounds. The positive ion fast atom bombardment mass spectra of glucosinolates exhibited abundant cationized and protonated molecula ions, thus complementing the previously reported electron impact and chemical ionization mass spectra of these compounds. Desulphoglucosinolates yield only the protonated molecular ion. Under negative ion conditions the spectra of glucosinolates were dominated by the molecular anion and this has been used as a means of analysing a crude plant extract containing a complex mixture of glucosinolates.

    Notizen: UV absorbance is a convenient detection method for monitoring a wide variety of different components separated by capillary electrophoresis (CE). However, a significant disadvantage of UV detection is its low sensitivity and this is a major factor limiting the application of CE as an analytical technique. In order to improve sensitivity, several methods of on-line sample preconcentration by electrofocusing, including “stacking” and isotachophoresis, have been investigated and the potential of on-line solid phase preconcentrators for this purpose has also been demonstrated. We have developed methods for the separation of isoforms of metallothionein (MT), a low Mr metal-binding protein which functions as a detoxification system and cellular buffer for heavy metals, primarily in liver and kidney. While a variety of modifications to the capillary surface and electrolyte chemistry have been used to resolve many MT isoforms in liver samples from a variety of species, there are few available alternatives to UV detection and thus sensitivity limits the application of these methods to samples with low MT levels, such as urine or plasma. In this study we have investigated the use of an inexpensive and easy to assemble C18 concentrator which was fitted to the capillary inlet to extend the detection limits for MT isoform analysis. Samples were pressure loaded onto the concentrator for up to 5 min, eluted using 33% acetonitrile and then subjected to electrophoresis in borate or phosphate buffer. The purified isoforms MT-1 and MT-2 from rabbit liver were best resolved using an acidic acetonitrile eluent and a phosphate buffer electrolyte at pH 7. Sensitivity enhancement was 〉700-fold and the relationship between loading time and isoform absorbance peak area was linear when only a small proportion of concentrator capacity was used. Analysis of sheep liver extracts demonstrated that both sensitivity and specificity were enhanced. The potential of other solid-phase concentrators is discussed.

    Beschreibung: Cerium oxide nanoparticles have oxygen defects in their lattice structure that enables them to act as a regenerative free radical scavenger in a physiological environment. We performed a comprehensive in vivo analysis of the biological distribution and antioxidant capabilities of nanoceria administered to mice perorally (PO), intravenously (IV), or intraperitoneally (IP) by dosing animals weekly for 2 or 5 weeks with 0.5 mg kg −1 nanoceria. Next, we examined if nanoceria administration would decrease ROS production in mice treated with carbon tetrachloride (CCl 4 ). Our results showed that the most extensive and cumulative nano-deposition was via IV and IP administered while PO administration showed mice excreted greater than 95% of their nanoceria within 24 h. Organ deposition for IV and IP mice was greatest in the spleen followed by the liver, lungs, and kidneys. Elimination for all administration routes was through feces. Nanoceria administration showed no overt toxicity, however, WBC counts were elevated with IV and IP administration. Our in vivo studies show that nanoceria administration to mice with induced liver toxicity (by CCl 4 ) showed similar findings to mice treated with N -acetyl cystine (NAC), a common therapeutic to reduce oxidative stress. Taken together, our studies show that nanoceria remains deposited in tissues and may decrease ROS, thereby suggesting that cerium oxide nanoparticles may be a useful antioxidant treatment for oxidative stress. © 2011 Wiley Periodicals, Inc.
