    Notizen: SYNOPSIS. Incorporation of 14C-labeled biopterin into Crithidia fasciculata was inhibited by 1 mM colchicine or lumicolchicine. These substances do not penetrate the cell membrane, hence they cannot interact with the subpellicular microtubules. In view of this, interference of colchicine with biopterin transport must occur on the outer surface of the cell membrane. Binding of colchicine to Crithidia was not temperature-dependent and did not exhibit saturation kinetics. These facts exclude a binding as in the case of tubulin, or similar proteins which may be present in the membrane. The results suggest an inhibition reflecting steric hindrance of the biopterin carrier system.

    Notizen: Summary The role of Juvenile Hormone III in phonotaxis, mating behavior and egg production of cricket females from Gryllus campestris, Gryllus bimaculatus, Teleogryllus commodus, and Acheta domesticus was investigated. 1. Phonotaxis of crickets was measured on a Kramer spherical treadmill by means of an infrared system which allowed free walking at any speed and direction. Crickets were allatectomized during the adult or last nymphal stage, and the controls sham-operated. On each cricket, 2 to 9 phonotactic tests were performed during a period of 12 to 51 days; the allatectomized nymphs were tested as adults. 2. All females deprived of their corpora allata during the adult stage continued to exhibit phonotaxis, whereas females operated as nymphs developed and maintained phonotaxis (Tables 1–4). Some of these females allatectomized or sham-operated during the adult or nymphal stage exhibited correct phonotactic behavior as well as undirected walking. 3. At the end of the test series, all allatectomized females were found to be devoid of Juvenile Hormone, whereas the controls varied according to age from 0.2 to 2.0 pmol/10 μl. The individual ecdysteroid levels differed unpredictably in allatectomized and control females (0.02 to 6.4 pmol/10 μl), and no correlation was found between the number of eggs produced by allatectomized females and their ecdysteroid titer. 4. All allatectomized females displayed sexual receptivity. 5. Adults allatectomized during the last nymphal instar were still able as adults to develop single eggs in G. campestris and G. bimaculatus, while females of T. commodus did produce and store moderate numbers. However, in A. domesticus no eggs were produced, and terminal oocytes had reached 0.5 to 0.7 mm in length, which is the previtellogenic size of terminal oocytes in normal females at the age of one to two days.

    Notizen: Abstract FemaleH. armigera moths are highly attracted by a steam distillate from pigeonpea plants, one of their main hosts. A mixture of six compounds, all sesquiterpenes (β-caryophyllene,α-humulene,α-guajene,α-muurolene,γ-muurolene, andα-bulnesene), mixed in the proportions as found in the steam distillate, elicited the same behavioral responses (oriented upwind flights and contacts with the odor source) as the steam distillate. Onlyα-bulnesene was attractive by itself, but still less than the whole mixture. In addition, the sesquiterpene mixture acts as an oviposition stimulant. Both behavioral responses, orientation and oviposition, are concentration dependent. Electrophysiological recordings from female and male antennae (EAG) showed the same qualitative and quantitative responses to each of the compounds of the sesquiterpene mixture. The EAG responses to the original steam distillate were higher and similar to chickpea kairomonal components, which were also tested. The pigeonpea sesquiterpene mixture and its individual components elicited weak EAG responses only. The response of the male antenna to female-produced pheromone components was in the same range as the pigeonpea steam distillate.

    Notizen: Summary The neurosecretory activity of azadirachtin A-treated Locusta migratoria that failed to mature was compared with that of rapidly maturing control females by means of histological techniques. High resolution drymount autoradiography was used to localize [22,23-3H2]dihydroazadirachtin A in the brain and corpus cardiacum (CC). The neurosecretory system of the locusts treated with this insecticide was accompanied by an unusually high accumulation of paraldehyde fuchsin (PAF)-stainable neurosecretory material (NSM) in the brain fibers and in the storage lobes of the CC. The control females never had such an intense accumulation of NSM. The accumulation of NSM, resulting from its poor release, may influence its synthesis which is controlled by feed-back regulation. [22,23-3H2]Dihydroazadirachtin A is localized mainly at the peripheral blood-brain barrier and does not penetrate into the brain effectively, whereas it completely covers the CC. It is concluded that azadirachtin A affects the endocrine activity of the brain in an indirect way.

The effects on the nucleus were the most noticeable and included: (1) crenulation of nuclear shape, (2) clumping of heterochromatin, and (3) pyknosis. The degenerative changes to the nuclei appeared to precede changes within the cytoplasm. It was concluded that the degeneration of all three endocrine glands within the ring complex would contribute to a generalised disruption of neuroendocrine function.

    Notizen: Durch chromatographische Trennung an phosphorylierter Cellulose wurden Biopterin und 7-Biopterin erstmals in reiner Form erhalten. Im Gegensatz zum Biopterin ist 7-Biopterin im Wachstumstest bei Crithidia fasciculata inaktiv. Die Ausbeute ist bei der Biopterinsynthese stark von den Reaktionsbedingungen abhängig und liefert unter optimalen Bedingungen 7% Biopterin und 26% 7-Biopterin, bezogen auf eingesetztes 2.4.5-Triamino-6-hydroxy-pyrimidin. Die Radiosynthese der beiden pterine, ausgehend von K14CN, wird beschrieben. Ferner werden die Eigenschaften des 2-Amino-4-hydroxy-6-bzw. 7-[L-erythro-1.2.3-trihydroxy-propyl]-pteridins mitgeteilt.

    Notizen: Bei der Kondensation von 2.4-Diamino-6-hydroxy-pyrimidin (Ia) mit D-Glucose, D-Mannose, D-Galaktose und D-Ribose entstehen in wäßrigem äthanol unter saurer Katalyse N-Acetale vom Typ Va. Diese Glykosylamine geben keine Amadori-Umlagerung und spalten bei der Einführung einer Stickstoff- Funktion (z. B. durch Nitrosieren) den Zuckerrest fast völlig ab. In geringer Menge konnte das 5-Nitroso-4-galaktosylamino-pyrimidin gefaßt werden. D-Glucose und D-D-Mannose sowie eine Reihe von Aldehyden (Formaldehyd, Malondialdehyd u. ä.) reagieren mit dem Aminopyrimidin Ia unter C-Alkylierung in Stellung 5 des Pyrimidinringes. Die Fähigkeit von weiteren substituierten Pyrimidinen (VI-XVI) zur C-Alkylierung und zur N- Acetalbildung bei der Umsetzung mit Aldosen wird untersucht. Nur bei Kondensation des 2-Dimethylamino-pyrimidins IX und des 4-Benzylamino-pyrimidins VIII mit Aldosen entstehen isolierbare Reaktionsprodukte.

    Notizen: D-Ribose, L-Arabinose, D-Xylose und L-Xylose werden durch Reduktion ihrer Methyl-pentosefuranosid-5-tosylate mit Lithiumalanat und milde Hydrolyse der Glykoside in einer Gesamtausbeute von 26-34% in die entsprechenden 5-Desoxy-pentosen umgewandelt. Nach Kondensation mit 2.4.5-Triamino-6-oxo-dihydropyrimidin werden daraus Biopterin und 7-Biopterin sowie deren mögliche optische Isomere hergestellt. Der Vergleich mit natürlichem Biopterin bestätigt die L-erythro-Konfiguration seiner 6-Dihydroxypropyl-Seitenkette. Die optischen Isomeren zeigen im mikrobiologischen Test etwa 10% der Wachstumswirkung des L-erythro-Biopterins.
