A hypodiploid strain of Tetrahymena thermophila has been obtained that shows arrest at the stage of condensed nuclei, corresponding to metaphase I of normal conjugates and induced arrest at meiotic metaphase I (i.e. at the stage of condensed, bivalent chromosomes) in its wt partner mate. The metaphase I arrested conjugants retained their old macronuclei and most of them underwent cell fusion, instead of separation of exconjugants. The doublets were viable and cortically integrated. When the arrest inducing strain was crossed to the haploid tester strain, the haploid micronuclei were arrested in the meiotic metaphase I as the diploid ones had been; the monovalent, chromosomes were condensed, the arms of sister chromatids were not separated, and they were not segregated. Separation of the arms of sister chromatids and disjunction of bivalent chromosomes were not prerequisite for the formation of micro-tubular spindles in those cells that were arrested in meiotic metaphase I. After re-feeding, the doublet cells resumed cell divisions, segregating two macronuclei and micronuclei at random. One macronucleus was derived from the arrest inducing strain and the other from the tester strain. Heterokaryon strains with macronuclei derived from the parental arrest inducing strain and with the micronucleus derived from the parental wt tester strain were obtained. Surprisingly, these heterokaryons did not induce meiotic arrest. Thus, the arrest in the meiotic metaphase I was induced by the micronucleus and not by the macronucleus of the arrest inducing strain.

    Notizen: Fenestrin is a cytoskeletal protein, which participates in conjugation and cell cycle regulation of Tetrahymena by formation of specific structures during these processes. In cell cycle it appears at the anterior poles of future proter and opisth cells. By 2-D electrophoresis, we found that in starved T. thermophila cells fenestrin appears in four isoforms that have the pH in the range of 5.2–5.5. The differences of pH of about 0.1–0.3 between isoforms could suggest that they differ by the phosphorylation state. By mass spectrometry, we identified some of the peptides derived form the enzyme digestion of fenestrin. These peptides were used for searching through T. thermophila genome, available at the site . A nucleotide sequence coding one of the peptides was initially found. Then, by extension, the sequence of all fenestrin gene appeared, containing the other peptides identified by spectrometry. By RT-PCR we characterized one intron in this gene. The analysis of the aminoacid sequence of the exons revealed that fenestrin is composed from at least two domains: N-terminal leucine-rich domain and C-terminal glutamic acid-rich domain. The BLAST search on general databases showed the existence of human and mouse proteins homologous to fenestrin in sequence, molecular mass and domain organization. Thus, fenestrin appears to be a protein correlated to cell polarity with an evolutionarily conserved structure. Its function should be cleared by mutational analysis and gene knock out.

    Notizen: . Conjugant pairs of Tetrahymena thermophila were mechanically separated by vigorous pipetting at the early stages of meiotic prophase. The complete sequence of conjugational nuclear events including the appearance of pronuclei, development of the new macronuclei (postzygotic development), and resorption of the old macronuclei was observed in the separated cells, without pronuclear exchange. The pronuclei in the separated cells were recognised by the presence of components of the extranuclear cytoskeleton, which were labelled with anti-tubulin and anti-fenestrin antibodies in the same way as in undisturbed conjugants. The apical region of the separated conjugants (the post-junction area), corresponding to the junction area of conjugants was labelled with anti-fenestrin antibody and maintained the properties required for the nuclear development. The results of the genetic study were consistent with a hypothesis that cytogamy (pronuclear fusion) was induced in the separated conjugants. Therefore, the lasting cell contact is not necessary for the successful completion of conjugational nuclear events.

    Notizen: Summary The importance of charged groups during phagocytic recognition of filamentous Cyanobacteria (Oscillatoria formosa andAnabaena spp.) by the stenophagic ciliatePseudomicrothorax dubius has been studied. Anionic and cationic domains are evenly and randomly distributed over the cyanobacterial surface, as demonstrated with scanning electron microscopy following labeling with colloidal gold (−) and colloidal gold coupled with poly-L-lysine (+). The phagocytosis ofOscillatoria was inhibited when filaments were treated with cationic reagents such as poly-L-lysine (pLL), FeCl3 and carbodiimide. In contrast elimination of cationic charges on theOscillatoria surface by treatment with poly-L-glutamic acid (pLGa) or colloidal gold did not affect phagocytosis. The effects of sequential treatment with pLL and pLGa demonstrated that pLL reduced phagocytosis of pLGa-pretreatedOscillatoria, whereas the pLGa restored phagocytosis of pLL-pretreated filaments. Scanning electron microscopy showed that pLL- or pLGa- treated filaments can still adsorb the oppositely charged colloidal gold particles on their surface. However, the treatment of filaments with pLL followed by pLGa prevented subsequent labeling with gold as well as with pLL-gold particles. Filaments ofAnabaena spp., which are not normally ingested byPseudomicrothorax, were also treated individually or sequentially with pLL and pLGa. None of these treatments, however, provoked phagocytosis ofAnabaena byPseudomicrothorax. We suggest that the surface charge alone does not play a crucial role in phagocytic recognition inPseudomicrothorax and that phagocytosis-specific molecules are implicated.
