    Notizen: Summary Experiments were performed to determine whether β-glycerophosphate (β-GP) promoted mineralization in vitro by modulating bone cell metabolic activity and/or serving as a local source of inorganic phosphate ions (Pi). Using MC3T3-E1, ROS 17/2.8, and chick osteoblast-like cells in the presence of β-GP or Pi, we examined mineral formation, lactate generation, alkaline phosphatase (AP) activity, and protein and phospholipid synthesis. Neither β-GP nor Pi modulated any of the major biosynthetic activities of the bone cells. Thus, we found no change in the levels of phospholipids, and the total protein concentration remained constant. Measurement of lactate synthesis showed that β-GP did not effect the rate of anaerobic glycolysis. Evaluation of medium Pi levels clearly indicated that β-GP was hydrolyzed by bone cells; within 24 hours, almost 80% of 10 mM β-GP was hydrolyzed. It is likely that this local increase in medium Pi concentration promoted rapid mineral deposition. Chemical, energy dispersive X-ray, and Fourier transform infrared analysis of the mineral formed in the presence of β-GP showed that it was nonapatitic; moreover, mineral particles were also seen in the culture medium itself. Experiments performed with a cell-free system indicated that mineral particles formed spontaneously in the presence of AP and β-GP and were deposited into a collagen matrix. We conclude that medium supplementation with β-GP or Pi should not exceed 2 mM. If this value is exceeded, then there will be nonphysiological mineral deposition in the bone cell culture.

    Notizen: Summary 14C-Formic acid and other14C-organic compounds are formed on surface materials when mixtures of14CO,12CO2 or N2 and water vapor are irradiated with ultraviolet light (UV) ofλ 〉 250 nm. The rate of organic formation is roughly proportional to the quantity of substratum irradiated. The available evidence suggests that14CO adsorbed to or in contact with the substratum is excited by the long wavelength UV and reacts with adsorbed H2O or surface hydroxyl groups yielding the organic products. Photodestruction of the14C-organics yields14CO2 and14CO. A steady state is attained when organic products reach a concentration such that the rate of photodestruction is equal to the rate of synthesis. The product accumulation is greater and the photodestruction is slower when N2 is used as diluent gas. Differences in the rates of synthesis, rates of photodestruction and amounts of product accumulation are observed with different silica and alumina substrata. The substrata with large surface areas are most effective for synthesis while maximum photoprotection of organics is afforded by substrata containing high concentrations of surface hydroxyl groups. The observation of the synthesis on a variety of substrata using realistic simulations of atmospheres and solar energies strengthens previous proposals that this process may occur on Mars and may have been important on the primitive Earth.

    Notizen: Summary Intestinal calcium-binding protein (CaBP) levels of rats fed a high (1.5%) Ca diet were the same whether the animals were parathyroidectomized (PTX), sham-operated controls pair-fed with the PTX animals, or sham-operated controls fed ad libitum. Consequently, a given base level of CaBP seems to be parathyroid hormone independent and not closely related to feed intake. On the other hand, whereas the ad libitum fed controls more than doubled their intestinal CaBP in response to a 2-day low-calcium (0.02%) regimen, neither the parathyroidectomized animals nor the pair-fed sham-operated controls were able to do so. Since the latter two groups consumed less feed and therefore less vitamin D than the ad libitum fed animals, the inability to increase CaBP in response to a low-calcium diet may have been caused by a restricted vitamin D intake rather than by the absence of parathyroid hormone.

    Notizen: Abstract The objective of this study was to examine the nucleotides of chick growth-plate cartilage and to measure the concentration of adenine nucleotides in the pre-mineralizing and mineralizing zones. Nucleotides were isolated from the two regions using a rapid-freezing technique and the concentration of individual components was ascertained by HPLC. The actual values of ATP, ADP, and other nucleotides in cartilage was low. The lowest values were recorded in the mineralized zone. In this latter zone the energy charge ratio and the ATP/ADP ratio were depressed. This was probably due to 02-related inhibition of mitochondrial oxidative activity . Additionally, the percentage of octanoate, a short-chain fatty acid that accumulates when aerobic metabolism is disturbed, was found to have increased in the calcifying zone. These findings suggest that calcification of cartilage is associated with hypoxia-related modulation of chondrocyte metabolism.

    Notizen: Summary Catenaria allomycis Couch (Blastocladiales) is an endobiotic fungal parasite primarily of species of the genusAllomyces. The life cycle ofC. allomycis contains both sexual and asexual phases. Synaptonemal complexes have been found in young developing resistant sporangia (RS) suggesting that meiosis occurs within the thick walled RS prior to syngamy. Ultrastructural evidence suggests that meiosis proceeds through pachytene in the developing RS and is arrested in diplotene of prophase I until the sporangia are induced to germinate at which time the meiotic process is completed. Quantitative nuclear counts in developing RS support the ultrastructural observations. Meiotic nuclei are characterized by polar fenestrae in the nuclear envelope and intranuclear plaque-like microtubule organizing centers (MTOC).

    Notizen: Summary The tppB locus of Salmonella typhimurium encodes the anaerobically-induced tripeptide permease. We have demonstrated that expression of tppB requires the function of the ompR and envZ gene products, originally identified as positive regulatory proteins required for the osmotic regulation of porin expression. Significantly, tppB expression is not osmotically regulated. We have also identified three additional genes whose expression depends on OmpR. Thus OmpR and EnvZ serve a more general regulatory role than has previously been supposed. This study provides the first detailed genetic analysis of the ompB locus of S. typhimurium.

    Notizen: Prior studies have shown that vitamin D regulation of protein kinase C activity (PKC) in the cell layer of chondrocyte cultures is cell maturation-dependent. In the present study, we examined the membrane distribution of PKC and whether 1α,25-(OH)2D3 and 24R,25-(OH)2D3 can directly regulate enzyme activity in isolated plasma membranes and extracellular matrix vesicles. Matrix vesicle PKC was activated by bryostatin-1 and inhibited by a PKC-specific pseudosubstrate inhibitor peptide. Depletion of membrane PKC activity using isoform-specific anti-PKC antibodies suggested that PKCα is the major isoform in cell layer lysates as well as in plasma membranes isolated from both cell types; PKCζ is the predominant form in matrix vesicles. This was confirmed in Western blots of immunoprecipitates as well as in studies using control peptides to block binding of the isoform specific antibody to the enzyme and using a PKCζ-specific pseudosubstrate inhibitor peptide. The presence of PKCζ in matrix vesicles was further verified by immunoelectron microscopy. Enzyme activity in the matrix vesicle was insensitive to exogenous lipid, whereas that in the plasma membrane required lipid for full activity. 1,25-(OH)2D3 and 24,25-(OH)2D3 inhibited matrix vesicle PKC, but stimulated plasma membrane PKC when added directly to the isolated membrane fractions. PKC activity in the matrix vesicle was calcium-independent, whereas that in the plasma membrane required calcium. Moreover, the vitamin D-sensitive PKC in matrix vesicles was not dependent on calcium, whereas the vitamin D-sensitive enzyme in plasma membranes was calcium-dependent. It is concluded that PKC isoforms are differentially distributed between matrix vesicles and plasma membranes and that enzyme activity is regulated in a membrane-specific manner. This suggests the existence of a nongenomic mechanism whereby the effects of 1,25-(OH)2D3 and 24,25-(OH)2D3 may be mediated via PKC. Further, PKCζ may be important in nongenomic, autocrine signal transduction at sites distal from the cell. © 1996 Wiley-Liss, Inc.
