Zusammenfassung Es werden Versuche beschrieben, bei denen die Zellcyclusdauer normaler und trisomer Zellen verglichen wird. Verwendet wurden Fibroblastenkulturen aus Hautbiopsien Neugeborener vom Karyotyp 46XY, 47XY21+ und 47XY18+. Die ersten Versuche waren dafür vorgesehen, das Verhalten normaler und trisomer Zellen unter den gegebenen Kulturbedingungen zu testen. Die Zeitdauer der log-Phase des Wachstums und die Fähigkeit, 3H-Thymidin einzubauen, waren für alle Zelltypen sehr ähnlich. Die Dauer der intermitotischen Periode eines jeden Zelltyps wurde bestimmt durch Zählung markierter Mitosen nach einem 30minutigen Puls-Labeling. Die experimentellen Ergebnisse lassen vermuten, daß G 2 etwa 3 Std länger in trisomen Zellen als in normalen Kontrollzellen ist.

Summary Experiments are described in which the cell cycle times of normal and trisomic cells were compared. The cells used were human fibroblast cultures established from skin biopsies from new-borns with karyotypes of 46XY, 47XY21+ and 47XY18+. Preliminary experiments were designed to test the behaviour of normal and trisomic cells under the experimental culture conditions. The timing of the log-phase of growth and the ability to utilise tritiated thymidine were very similar for all cell types. The intervals of the inter-mitotic period of each cell type were compared using the technique of scoring labelled mitoses after exposure to a 30-minute pulse of tritiated thymidine. The experimental results indicate that in trisomic cells G 2 may be approximately 3 hrs longer than that of the normal controls.
