Description: The BioEnergy Science Center (BESC) is undertaking large experimental campaigns to understand the biosynthesis and biodegradation of biomass and to develop biofuel solutions. BESC is generating large volumes of diverse data, including genome sequences, omics data and assay results. The purpose of the BESC Knowledgebase is to serve as a centralized repository for experimentally generated data and to provide an integrated, interactive and user-friendly analysis framework. The Portal makes available tools for visualization, integration and analysis of data either produced by BESC or obtained from external resources. Description: Methylmercury is a potent neurotoxin produced in natural environments from inorganic mercury by anaerobic bacteria. However, until now the genes and proteins involved have remained unidentified. Here, we report a two-gene cluster, hgcA and hgcB, required for mercury methylation by Desulfovibrio desulfuricans ND132 and Geobacter sulfurreducens PCA. In either bacterium, deletion of hgcA, hgcB, or both genes abolishes mercury methylation. The genes encode a putative corrinoid protein, HgcA, and a 2[4Fe-4S] ferredoxin, HgcB, consistent with roles as a methyl carrier and an electron donor required for corrinoid cofactor reduction, respectively. Among bacteria and archaea with sequenced genomes, gene orthologs are present in confirmed methylators but absent in nonmethylators, suggesting a common mercury methylation pathway in all methylating bacteria and archaea sequenced to date.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Parks, Jerry M -- Johs, Alexander -- Podar, Mircea -- Bridou, Romain -- Hurt, Richard A Jr -- Smith, Steven D -- Tomanicek, Stephen J -- Qian, Yun -- Brown, Steven D -- Brandt, Craig C -- Palumbo, Anthony V -- Smith, Jeremy C -- Wall, Judy D -- Elias, Dwayne A -- Liang, Liyuan -- New York, N.Y. -- Science. 2013 Mar 15;339(6125):1332-5. doi: 10.1126/science.1230667. Description: Experimental removal of helpers from a communal bird population decreased the reproductive success of the reduced units. By controlling for variables that are potentially correlated with both reproductive success and unit size, this experiment establishes that helpers-contribute significantly to their indirect fitness.〈br /〉〈span class="detail_caption"〉Notes: 〈/span〉Brown, J L -- Brown, E R -- Brown, S D -- Dow, D D -- New York, N.Y. -- Science. 1982 Jan 22;215(4531):421-2.〈br /〉〈span class="detail_caption"〉Record origin:〈/span〉 〈a href="http://www.ncbi.nlm.nih.gov/pubmed/17814957" target="_blank"〉PubMed〈/a〉

Notes: Calcium aluminate bars were formulated to study the effect of real and simulated biological environments on strength. Static fatigue tests revealed that the strength of these potential internal bone prostheses decreased to ca. 60% of their initial strength after less than 100 min while being held at constant stress in solution. Calcium aluminate bars aged for 12 weeks in water or Ringer solution were also found to exhibit large losses in strength, while bars implanted in vivo for the same period of time weakened somewhat less. Semi-quantitative analysis of the solutions in which the calcium aluminate bars were aged revealed that Al3+ and Ca2+ were being leached from the bars in both the saline and water baths, while the concentration of Mg2+ was decreasing in the Ringer solution.Scanning electron microscope (SEM) studies revealed extensive microstructural changes in specimens aged in water or saline solution for 1 and 12 weeks. Similar but far less extensive changes were seen in bars aged in vivo in rabbits for 12 weeks.

Notes: Aging studies were done on calcia stabilized zirconia rods of 72% theoretical density to determine the effect of actual and simulated biological environments on their strength. They were aged without stress in vitro in Ringer's solution for 1, 2 and 4 weeks or in vivo in rabbits for 12 weeks. Rods aged in vitro showed mean losses in bending strength of 16, 17 and 19% respectively after 1, 2 and 4 weeks of immersion, while those aged in vivo showed a mean loss of 25%. It was concluded that the material tested would be unsatisfactory as an orthopedic replacement because of the rapid decrease in strength which occurred when exposed to actual or simulated biological media.
