    The chromosomal distribution and organization of sheep satellite I and II centromeric DNA using characterized sheep-hamster somatic cell hybrids (1996)

    Notes: Abstract A panel of sheep-hamster somatic cell hybrids containing single sheep chromosomes was used to study the chromosomal distribution and organization of two families of sheep centromeric satellite DNA. This study shows that the centromeres of the sheep metacentric chromosomes 1,2 and 3 differ in their organization and relative quantities of sheep satellite I DNA. The results, when correlated with the proposed formation of these metacentric chromosomes by ancient Robertsonian translocations, suggest a loss or replacement of satellite I centromeric DNA from the centromeres of these sheep chromosomes. Using Southern blot analysis and fluorescencein situ hybridization, this study shows that the recent centric fusion chromosome t2 (rob 9;10) contains little satellite II DNA. Together these resuults suggest the possibility of substantial reorganization of sheep centromeric DNA families after Robertsonian translocations.

    Notes: Abstract One hundred and six microsatellite repeat-containing loci, including 59 CA-containing repeats from the CEPH/Genethon collection, were regionally assigned on human chromosome 3 using a somatic cell hybrid mapping panel, dividing the chromosome into 14 intervals. The others were dinucleotide and tetranucleotide repeat-containing loci newly developed for human chromosome 3, of which 26 were also localized by means of genetic linkage analysis against selected CEPH microsatellites. The regional assignment of these two marker sets in a common mapping panel facilitates their integration. Incorporation of these highly polymorphic loci into the developing physical and genetic maps should provide useful information for studies of various diseases involving chromosome 3.

    Notes: Abstract High-resolution bivariate flow karyotypes were obtained using fibroblast cell lines from a sheep with a normal karyotype (2n=54), from sheep carrying Robertsonian translocation chromosomes and from sheep—hamster somatic cell hybrids. By taking advantage of the presence of chromosome polymorphisms, translocation chromosomesand sheep—hamster somatic cell hybrids, all sheep chromosomes were isolated by flow sorting. Chromosome-specific paints were generated from each sorted peak using degenerate oligonucleotide-primed polymerase chain reaction (DOP-PCR). The sheep chromosome present in each peak was identified by chromosome-specific microsatellite analysis of the DOP-PCR products and fluorescence in situ hybridization (FISH) onto DAPI-banded sheep metaphase chromosomes. The chromosome-specific DNA obtained in this study can be used for the production of genomic libraries and as a resource for mapping randomly cloned DNA sequences that will greatly aid the construction of genetic and physical maps in the sheep. The chromosome-specific paints will facilitate chromosome identification and contribute to the study of karyotype evolution in the sheep and related species.
