Um die Wirkung von Salzlösungen auf die Konformation der Helix von Amylose und Amylopektin zu untersuchen, wurden die spezifischen Drehungen [α]D verschiedener Polysaccharide bestimmt. Die Fähigkeit von Anionen und Kationen, den Wert von [α]D,s herabzusetzen, steigt gemiäß der Reihenfolge Cl- 〉 Br- 〉 SCN- und Li+ 〉 G+. Deshalb liegt der stzirkste Abfall von [α]D,s in konzentrierten Guanidinrhodanid(GSCN)-Losungen vor, was auf Grund der Eignung zu1 Zerstörung der Struktur des Wassers durch diese Ionen z u erwarten war (D±, 〉 D1, für G+- und SCN--Ionen).Die Verkleinerung von [α]D,s durch Salze erfolgt also ähnlich der Verkleinerung, die sich aus der Temperatursteigerung einer wälßrigen Lösung von Amylose oder Amylopektin ergibt. Die Wirkung der Ionen besteht also darin, daß ganz allgemein die Zahl der Cluster und Wasserstoffbrückenbindungen verringert wird. Wie man jedoch aus einem Vergleich der Dispersionskraft mit der Änderung des [α]D,s erkennen kann, zeigt die geringe Änderung von -13 für [α]D,s fur das wirksamste Lösungsmittel an, daß die Helix in diesen Salzlösungen zumeist nur teilweise zerstört ist und daß die Hauptursache fuuml;r die Herabsetzung von [α]D,s, in einer Steigerung der Wärme-oder statistischen Bewegung des Polymeren und seiner Atome liegt. Die größte Änderung von [α]D,s bei dem Übergang von Wasser zu Salzlösungen erfolgt durch die Anteile mit niederem Molekulargewicht wie Glucose, Methyl-α-D-glucosid und Maltose, weil diese Moleküle nicht so sehr in ihren statistischen Bewegungen behindert sind und daher entsprechend mehr auf die Steigerung der Fluiditat des Wassers ansprechen. Eine entgegengesetzte Wirkung hat indessen das Sulfation in MgSO4, oder G2SO4, bei Maltose und Polyglucose, nicht aber bei Glucose oder Methyl-α-D-glucosid. Offensichtlich stabilisiert das Sulfation die Wasserstoffbrückenbindung zwischen den Hydroxylgruppen von C-2 und C′-3, und dabei verringert sieh dadurch die statistische Bewegung der Moleküle mit kleinerem Molekulargewicht. Der hohe Wert von D+ für Mg++ steigert die Änderung von [α]D,s für Glycogen. Untersuchungen der Lichtstreuung und der Viskositat zeigen, daß Stärke in diesen Salzlösungen auch während 12 Monaten nicht abgebaut wird; d. h., daß die Polysaccharide in diesen Lösungeu offensichtlich vor Oxydationswirkungen geschützt sind.

The specific rotations [α]D of various polysaccharides were examined in different salt solutions in order to determine the effect of such solutions on the conformation of the helix of amylose and amylopectin. The ability of anions and cations to decrease the value of [α]D,s increases according to the sequences Cl- 〈 Br- 〈 SCN- and Li+ 〈 G+. Hence, the largest decrease in [α]D,s occurs in concentrated guanidinium thiocyanate (GSCN) solutions which is to be expected on the bases of the ability of these ions to destroy the sturcture of water (D± 〉 D1 for G+ and SCN- ions). The reduction in [α]D,s, is therefore similar to that reduction resulting from an increase in the temperature of an aqueous amylose or amylopectin solution. The effect of the ions is therefore to reduce the number of water clusters and hydrogen bonds in general. However, as seen from a comparison of the dispersing power with a change in [α]D,s, the small change of -13 for [α]D,s for the most effective solvent indicates that the helix is at most only partially destroyed in these salt solution and that the major effect in decreasing the value of [α]D,s is the increase in the thermal or random motion of the polymer and its atoms. The greatest change in [α]D,s, in going from water to salt solutions occurs with the lower molecular weight species such as glucose, methyl α-D-glucoside, and maltose because these molecules are not as restricted in their random motions and therefore can take a greater advantage of the increase in fluidity of the water. However, the sulfate ion in either MgSO4 or G2SO4 gives the Opposite effect for maltose and polyglucose but not for glucose or Methyl alpha;-D-glucoside. Apparently the sulfate ion stabilizes the hydrogen bond between C-2 and C′-3 hydroxyl groups and thereby decreases the random motion of the lower molecular weight species. The high value of D+ for the Mg++ increases the change in [α]D,s for glycogen. Light scattering and viscosity studies show that these salt solutions do not degrade the starch even after 12 months and thereby must protect the polysaccharides from oxidation effects.
