    Notes: A comparative analysis of the sperm-whale myoglobin structure in the crystal and in solution has been carried out with the technique previously formulated by the authors which uses the large-angle X-ray diffuse scattering for investigating the protein structure in solution. A `modified cube method', correctly taking into account the cavities within the protein molecule accessible to the solvent, is proposed for an accurate estimate of the solvent influence and for calculation of the scattering intensities. A comparison of the theoretical myoglobin scattering curves with the experimental scattering curve of this protein obtained by Stuhrmann shows rather noticeable quantitative divergences, which can be eliminated by a small increase in the distance between the `hairpin' GH and the other part of the protein molecule. It is also shown that several other physically reasonable shifts of helices (or groups of helices) do not lead to an agreement between the experimental and theoretical scattering indicatrices.

    Notes: Two methods of calculation of indicatrices of diffuse X-ray scattering by globular proteins in a medium with a non-zero electron density are described. The methods are used in calculation of curves of scattering by sperm whale native myoglobin in different solvents. The indicatrices obtained are close to the experimental. The nature of the so-called `10 Å, maximum', which is observed on the scattering curves of globular proteins in dense solvents as well as in concentrated protein solutions and protein gels, is discussed.

    Notes: A new method has been developed for solving the integral equation which determines the slit function H(x) used for correcting small-angle scattering curves for the effects of the length of the collimating slits. With this technique the H(x) function has been calculated for the weighting function for the Beeman four-slit collimation system. The slit function has also been evaluated for a Gaussian weighting function and for a weighting function for which the intensity distribution in the detector plane has the form of a trapezoid. The slit functions for all of these weighting functions gave excellent results when employed to compute collimation corrections for several test functions. The slit functions also permitted a study of the sensitivity of the corrected intensity to changes in the form of the weighting function. The tests verified that, as has usually been assumed in the past, the corrected intensity is relatively intensitive to small changes in the weighting function.

    Notes: A method developed earlier [Fedorov & Aleshin (1966) Vysokomol. Soed. 8, 5016; translated into English in Polymer Sci. USSR (1967) 8, 1657] which permits the determination of the radial electron density distribution function of cylinders on the basis of the small-angle scattering curve, is applied to the study of the tobacco mosaic virus structure. The results obtained are in good agreement with the X-ray structural analysis of TMV. Questions concerning the extrapolation of the experimental curve, the estimation of the obtained resolution of electron density as well as the possibilities of this method are also considered.

    Notes: Conclusions 1. The construction of a trestle with crabs of the service gantry cranes on the gated wall of the upper pool made it possible to operate the gates under conditions of incompleted construction, which provided start-up of all units on time. 2. The use of service gates during construction eliminated the construction of additional temporary flashborads and corresponding consumption of metal. 3. The use of structural elements of the concrete-buggy trestle of the Sayano-Shushenskoe hydrostation instead of the earlier specified reinforced-concrete elements of the bridge and crane beams considerably simplified fabrication and shortened the assembly time. 4. The timely design developments for the mechanical equipment in the construction period, correlated to the actual conditions at the construction site, made it possible to accomplish the assembly works in the shortest possible time.

    Notes: An improved cube method has been developed for calculating the intensity of diffuse x-ray scattering of macromolecules in solution using a certain set of their atomic coordinates. The technique is based on the ideas of B. Lee and F. M. Richards [(1971) J. Mol. Biol. 55, 374-400] and Richards [(1977) Annu. Rev. Biophys. Bioeng. 6, 151-176] on the possibility of estimating the molecular and accessible surface of a particle by “rolling” a sphere, simulating a water molecule, on its molecular surface. It is shown that this technique is more advantageous than earlier versions of the cube methods. The improved technique for calculating scattering curves was utilized for several globular proteins, and for the first time, reliable scattering curves were obtained for protein-“bound” water complexes. In the case of globular proteins and tRNA, this technique has permitted a strict evaluation of their accessible surfaces, their volumes, and, apparently for the first time, their complete molecular surfaces.

    Notes: The cross-sectional radius of gyration of the deoxyribonucleoprotein (DNP) threads was measured by small-angle X-ray scattering in a wide range of ionic strengths (from 0.0005 to 2 M NaCl). For DNP in a solution of low ionic strength, this value is 30 Å. The increase of ionic strength results in partial deproteinization of DNP, while the cross-sectional radius of gyration varies from 25 Å for DNP in 0.7 M NaCl to 10 Å for DNP in 2 M NaCl. It is suggested that gradual deproteinization by the increase of NaCl concentration causes conformational changes, which are associated with the alteration of the DNP superstructure. The data are interpreted on the basis of the superhelical model of DNA packing in DNP; however, the coexistence of superhelical and unfolded regions in the DNP structure is also a possibility.
