    Notes: A paralog (intraspecies homolog) of the Saccharomyces cerevisiae YEF3 gene, encoding elongation factor-3, has been sequenced in the course of the yeast genome project, and identified by database searching; this gene has been designated HEF3. Bioinformatic and Northern blot analysis indicate that the HEF3 gene is not expressed during vegetative growth. Deletion of the HEF3 gene reveals no growth defects, nor any defects in mating or sporulation. A high copy 2μ clone of HEF3 was constructed, and was shown to be unable to complement a null allele of yef3. Finally, an in vitro assay for ribosome-stimulated ATPase activity was performed with isogenic HEF3 and Δhef3 strains; no difference in biochemical activity could be detected in these strains. From these results, we conclude that the HEF3 gene does not encode a functional homolog of YEF3. © 1998 John Wiley & Sons, Ltd.

    Notes: The conversion of the uninucleate amoebal form of Physarum polycephalum to the multi-nucleate plasmodial form is under the control of a genetic region which contains matA (or mt), a determinant of mating specificity. The region is the site of most gad mutations, which give amoebae the ability to produce plasmodia in clones without mating (ie, to self). In the present study, nonselfing revertants were isolated from two matA2-derived gad mutants and two matA3-derived gad mutants. Some revertants were found to have regained exactly, or nearly, the same phenotype as the original matA2 or matA3 strain. Others expressed new mating types, having gained the ability to mate with strains of the parental matA type. The results are compatible with a model in which new mating types arise from forward mutations (gad) and back mutations (npf or no plasmodium formation) occurring successively in a single gene, matA.

    Notes: GABA is a known inhibitory neurotransmitter in the mammalian brain. The site of GABAergic synapses can be determined with immunocytochemical methods that localize either GABA or its synthesizing enzyme, glutamate decarboxylase (GAD). In general, GABAergic axon terminals contain pleomorphic synaptic vesicles and form symmetric synapses. However, a small number of GABAergic axon terminals in selected brain regions (spinal cord, cerebellum, superior colliculus, striatum, globus pallidus, inferior olive, and substantia nigra) form asymmetric synapses. GAD- and GABA-immunoreactive processes that contain synaptic vesicles participate in every known morphological type of chemical synapse. These include axosomatic, axodendritic, axospinous, initial segment, axoaxonic, dendrodendritic, serial, reciprocal, and ribbon synapses. Although GABAergic synapses form a heterogeneous group, they most commonly form axosomatic, axodendritic, and initial segment synapses in the brain and spinal cord. These findings provide helpful guidelines for the identification of GABAergic synapses in future studies.

    Description: A mathematical model based on the Euler-Bernoulli beam theory is proposed for predicting the effective Young's moduli of piece-wise isotropic composite laminates with wavy patterns in the main load-carrying layers. Strains in corrugated layers, in-phase layers, and out-of-phase layers are predicted for various geometries and material configurations by assuming matrix layers as elastic foundations. Experimental results obtained from corrugated aluminum specimens and aluminum/epoxy specimens with in-phase and out-of-phase wavy patterns coincide very well with the predictions. The work represents a preliminary effort toward further generalization of the model for two-dimensional anisotropic laminates containing wavy patterns in the main load-carrying layers.

    Description: NASA is conducting the Airframe Structural Integrity Program in support of the aging commercial transport fleet. This interdisciplinary program is being worked in cooperation with the U.S. airframe manufacturers, airline operators, and the FAA. Advanced analysis methods are under development and an extensive testing program is under way to study fatigue crack growth and fracture in complex built-up shell structures. Innovative nondestructive examination technologies are also being developed to provide large area inspection capability to detect corrosion, disbonds, and cracks. Recent fracture mechanics results applicable to predicting the growth of cracks under monotonic and cyclic loading at rivets in fuselage lap-splice joints are reviewed.

    Description: An interactive cathode ray tube (CRT) terminal application has been developed which automatically generates the job control language (JCL) and submits NASTRAN batch jobs at an IBM mainframe installation. The relevant parameters for each NASTRAN batch job are stored in a table. Once a table entry is defined, the corresponding NASTRAN batch job may be submitted as often as desired with a minimum of keystrokes. For most users, there is no need for any knowledge of JCL.

    Description: NASA is developing a 'tool box' that includes a number of advanced structural analysis computer codes which, taken together, represent the comprehensive fracture mechanics capability required to predict the onset of widespread fatigue damage. These structural analysis tools have complementary and specialized capabilities ranging from a finite-element-based stress-analysis code for two- and three-dimensional built-up structures with cracks to a fatigue and fracture analysis code that uses stress-intensity factors and material-property data found in 'look-up' tables or from equations. NASA is conducting critical experiments necessary to verify the predictive capabilities of the codes, and these tests represent a first step in the technology-validation and industry-acceptance processes. NASA has established cooperative programs with aircraft manufacturers to facilitate the comprehensive transfer of this technology by making these advanced structural analysis codes available to industry.

    Description: Over the past two decades a number of thermomechanical constitutive theories have been proposed for viscoplastic metals. These models are in most cases similar in that they utilize a set of internal state variables which provide locally averaged representations of microphysical phenomena such as dislocation rearrangement and grain boundary sliding. The state of development of several of these models is now at the point where accurate theoretical solutions can be obtained for a wide variety of structural problems at elevated temperatures. The fundamentals of viscoplasticity are briefly reviewed and a general framework is outlined. Several of the more prominent models are reviewed, and predictions from models are compared to experimental results.

    Description: An investigation of thermomechanical testing and deformation behavior of tubular specimens under torsional loading is described. Experimental issues concerning test accuracy and control specific to thermomechanical loadings under a torsional regime are discussed. A series of shear strain-controlled tests involving the nickel-base superalloy Hastelloy X were performed with various temperature excursions and compared to similar thermomechanical uniaxial tests. The concept and use of second invariants of the deviatoric stress and strain tensors as a means of comparing uniaxial and torsional specimens is also briefly presented and discussed in light of previous thermomechanical tests conducted under uniaxial conditions.

    Description: The present cumulative damage model for the prediction of stiffness loss in graphite/epoxy laminates applies a thermomechanical constitutive theory for elastic composites with distributed damage. The model proceeds from a continuum mechanics and thermodynamics approach in which the distributed damage is characterized by a set of second-order tensor-valued internal state variables. A set of damage-dependent laminated plate equations is obtained; this is developed by modifying classical Kirchhoff plate theory.
