    Notes: Summary A capillary-scanning system for increasing the total throughput of DNA sequencing has been developed. The DNA sequencing method using the system features a spatial and temporal separation of laser-induced fluorescence detection from capillary gel electrophoresis. Fluorescently labelled adenine base fragments of pBluescript SK(−) were, produced in enzymatic sequencing reactions, and separated by capillary gel electrophoresis using UV-visible transparent capillaries filled with 8% T, 0% C polyacrylamide gel. The capillary containing all bands of the fragments was then scanned longitudinally with a 488 nm argon ion laser beam in 2.6 min. The adenine base sequence of the DNA was determined out to 400 bases by detecting fluorescence signals generated from the bands during the scan. The present scan speed is essentially limited by a slow strip-chart recorder and could be greatly increased by employing a fast data acquisition system.

    Notes: Abstract The Cu/Zn superoxide dismutase (SOD1) catalyzes the dismutation of superoxide radicals produced in the course of biological oxidations. When placed under the control of the rat SOD1 gene promoter and transfected into human HepG2 hepatoma cells, the activity of a chloramphenicol acetyltransferase reporter gene was found to increase three- to four-fold in the presence of heavy metals (cadmium, zinc and copper). Functional analysis of mutant derivatives of the SOD1 gene promoter and the use of a heterologous promoter system confirmed that the induction of the SOD1 gene by metal ions requires a metal-responsive element (MRE) located between positions −273 and −267 (GCGCGCA). It was also shown by gel mobility shift assays that an MRE binding protein is induced by the exposure of the human liver cell line HepG2 to heavy metals. These results suggest that the MRE participates in the induction of the SOD1 gene by heavy metals.

    Notes: Abstract: While the intercalation of C60 and the formation of C 60 - in the supercages of NaX and NaY are confirmed by using 129Xe NMR and ESR, the photophysical properties of C60 and C 60 - are characterized by monitoring transient reflectance spectra and kinetics, fluorescence kinetics, and diffuse reflectance spectra. C 60 - is considerably more abundant in NaY than in NaX. This difference is explained in terms of polarity difference between two zeolites. Both C60 and C 60 - have remarkably elongated excited-state lifetimes due to their collision-free environment in zeolitic nanocavities although C 60 - has much shorter lifetimes than C60. C 60 - , in particular, shows intense absorption and emission due to its reduced symmetry in zeolites.
