    Description: Breeding has moved beyond the point of simply propagating random stock. Genetic potential can be improved by establishing and crossing genetically defined strains, crossing species and manipulating gametes. Some of these activities require cryopreservation of semen. Cryopreserved sperm could also be a means of exchange of genetic material between locations and populations when transportation of live fish is restricted. In this study we had a survey on rainbow trout sperm quality in Genetic and Breeding Center for Coldwatwr Fishes, Shahid Motahari, Yasouj. Then Cryopreservation trials on rainbow trout (Oncorhynchus mykiss) sperm were carried out using six basic extenders. Egg batches of 25 g were inseminated with semen frozen in five 0.5-ml straws. Thowing temperature of cryopreserved sperm also was tested. In anather trial we investigate different cryoprotectant. The result showd that the best period for sperm collection in this center was from Novamber to January. The best extender based on sperm motility parameters were extender 2 and 1 with 57.2 and 56.9% fertilisatin rates, respectively. So this two extender were used in the further experiments. The best thowing temperathre was 25°C in 30s. Testing different cryoprotectant, adding 10% methanol to extender 1 gave the highest fertilization rate (64.6%) among cryoprotectant that used with this extender. In the other extender (2) adding 5% DMSO and mixture of 5% DMSO and 1% glycerol gave the highest fertilization rates (64.8% and 67.0%, respectively). In conclusion using extender 1 and 2 with mentioned cryoprotectant and thowing rate of 25°C in 30 s was recammended for rainbow trout sperm cryopreservation.

    Description: In the present study, hydrolysed protein of viscera and head of Silver carp (Hypophthalmichthys molitrix) was compared using Alcalase enzyme and internal tissue enzymes at 2 and 4 hours. The result indicated that product by Alcalase (Treatment 1) had significantly higher protein and rate of hydrolysates than that produced by internal tissue enzymes (Treatment 2). So, the highest mean (±SD) protein (68.10±1.33) was related to treatment 1-head (with Alcalase enzyme) after 4 hours and the highest rate of hydrolysates (29.36±1.35) was related to treatment 1-head (with Alcalase enzyme) after 4 hours. The result indicated that rate of hydrolysates raised as time of hydrolysates increased. However the intensity and rate of hydrolysates is reduced. The highest rate of hydrolysates occurred at 120 minutes in the first. This mode was similar for two treatments. The result can be considered as the Alcalase was preferred to internal enzyme.

    Description: The changes in chemical, microbial and quality sensory of fillets for silver carp fish treated with black cumin extract during refrigerated storage were investigated. The fish were divided into two groups. First group was dipped in black cumin extract with concentration of 1% and received the highest score in sensory evaluation. Then the fishes were wrapped in polyethylene package. The second group, as the control samples, were wrapped in polyethylene package after dipping in distilled water. All treatments were stored at refrigerator temperature (1±4°C). The microbial tests including total viable count, psychrotrophic count and chemical tests including peroxide index, thiobarbituric acid, free fatty acid value and total volatile basic nitrogen, with sensory evaluation, were done at 4°C over a period of 15 days. The results showed that the black cumin extract delayed lipid oxidation and protein analyses significantly in treated fishes. Psycrotrophic bacteria and total viable count of samples being treated with black cumin extract were maintained lower than the proposed acceptable limit (7 log cfu/g). In comparison to the control samples, microbial spoilage significantly decreased in treated samples. Furthermore, according to sensory analysis, the treatment with black cumin extract led to high quality during storage.The findings indicated that black cumin exerts had strong antioxidant and antibacterial impacts on silver carp fish, such that the shelf life of fillets being treated with black cumin were 2.5 times more than that of control samples during storage in refrigerator.

    Description: In this study, rainbow trout diet was supplemented with different levels of annatto (Bixa orellana) seed powder and then some blood parameters (i.e. red and white blood cells count, hemoglobin concentration and hematocrit percentage), growth performance and the fillets color was evaluated. The experimental diets consisted of 0 (control), 0.25, 0.5 and 0.75% annatto powder with three replicates. A total of 360 juvenile rainbow trout (100±5 g) were stocked completely randomly in 12 tanks and reared for 40 days. The maximum amount of all measured blood parameters except white blood cells were obtained in supplemented diet with 0.75% annatto seed powder and the minimum amount of the blood indices were observed in control group (p〈0.05). Also, the highest body weight (235 g) was related to 0.75% annatto seed powder and the lowest (108.2 g) was related to control one (p〈0.05). Spectrum of red and yellow colors in the fish fillets were increased by increasing in annatto seed powder level in the diet and the highest values were recorded in the fish fillet fed with 0.75% annatto seed powder in comparison with others (p〈0.05). Therefore, the addition of annatto seed powder at 0.75% in rainbow trout diet has a positive effect on the blood parameters, and on the other hand, by increasing the growth rate and fillet coloring, probably the cost of culturing will reduce and the customer's satisfaction with the fish meat will increase.

    Description: Considering the importance of rainbow trout (Oncorhynchus mykiss) in supplying required protein of people and effort to increase the efficiency of these fish reproduction, some related factors such as sperm quality and potential fertility of male are necessary. The aim of this study was to find out the effects of different dosages of Arginine on the biochemical parameters (including LDH, AST, ALT, ALP, iron, magnesium, phosphorous, chloride, calcium, sodium, Potassium, cholesterol, uric acid, urea, fructose, glucose, total protein and pH) of rainbow trout seminal plasma. For this purpose, five practical diets (each consisting of 3 triplicates) were supplemented with Arginine at 0.00 (Control), 0.50, 1.00, 1.50 and 2.00%. Broodstock feed last for 90 days. At the end of the feeding period one fish was captured from each replicate in order to collect their semen. Results indicate that there were no significant differences in LDH, ALP, Fe^2+ and P content among different treatments. The lowest level of AST and ALT and the highest level of Ca^2+ and Mg^2+ ions were obsereved in the treatment fed with 1.50% of Arginine which showed significant differences with other treatments (p〈0.05). Moreover, the amount of Cl- , Na^+ and K^+ ions were significantly increased in the seminal plasma in fish which were fed on diets containing arginine in comparison to control. As the amount of Arginine were increased, the levels of uric acid stepped up significantly in contrast to the urea and glucose. The highest amounts of cholesterol, fructose and total protein were observed in the treatments fed on 2.00, 0.50 and 1.00% of Arginine, respectively, that showed significant differences with other treatments (p〈0.05). The highest pH value was assayed in the 1.50% of Arginine treatment. The results of the Pearson correlation coefficient showed a significant positive correlation between Ca^2+ and Mg^2+ ions and Na^+ with K^+ and Clions (r=0.750, r=0.769 and r= 0.938, p〈0.01), respectively. On the other hand, a significant negative correlation between cholesterol with Cl- , Na^+ , K^+ , ALT and LDH (r=- 0.764, r=-0.724 and r=-0.728, p〈0.01) and (r=-0.531 and r=-0.560, p〈0.05) and also a significant positive correlation between K^+ and Clions (r=0.836, p〈0.01) were observed. Finally, it can be expressed that the levels of most of the ions were increased and there was a reduction in the levels of enzymes in seminal plasma of fish which were fed with practical diet including 1.5% of Arginine. So it can be recommended that adding this value of Arginine to the diets of rainbow trout broodstock, would improve the sperm quality which results in the enhancement of efficiency in rainbow trout reproduction.

    Description: The effect of essential fatty acids (EFA) and vitamin C-enriched Artemia nauplii on growth, survival, and resistance to temperature (high) stress in rainbow trout larvae reared in tanks were investigated. The larvae (average weight 120.43mg±13.5) were fed 6 times daily starting at the onset of exogenous feeding for 1 week. Triplicate groups of fish were offered one of four treatments (1) commercial starter food for rainbow trout، (2) newly hatched Artemia nauplii (unenriched) ،(3) highly unsaturated fatty acid (HUFA) + vitamin C-enriched Artemia nauplii and (4) combination of 10 % HUFA+ vitamin C enriched nauplii and commercial starter food. After 1 week, all groups of fish were switched to the commercial diet for an additional period of 3 weeks. Statistical analysis of growth after the first week and at the end of the experiment, showed that growth of larvae in various treatments was significantly different (P〈0.05). After 4 weeks, the larvae in treatment 3 with the average weight of 657.50 ± 57.93 mg had the highest body weight (P〈0.05). The highest percentage of survival (96%) was observed in treatment 3 (P〈0.05). Proximate compositions of trout larvae after one week feeding with experimental diets showed that the protein in the larvae of treatments 3 and 4 was significantly different compared to other treatments (P〈0.05). The best result of resistance to temperature (up to 24ºC) was observed in larvae reared on treatment 3 with 91.34 ± 1.52 percent (P〈0.05).

    Description: The present study evaluated the effect of Lactobacillus plantarum as a probiotic on the growth performance and haematological parameters of juvenile rainbow trout (Oncorhynchus mykiss) weighing 29.6±1.8 g immunized with streptococcus/lactococcus bivalent vaccine for 60 days at 16±1.5ºC. Fish were randomly allocated to 4 equal groups: probiotic (g kg^-1 feed equal to 10^8 cells g ^-1 ) supplemented diet group (group P), vaccinated fish fed with a normal diet (group V), vaccinated fish fed with probiotic (group P+V) and unvaccinated fish fed normal diet (group C). Results showed that weight gain, final weight, condition factor and thermalunit growth coefficient were improved in P+V group compared with other groups but was significantly different from groups V and C (p〈0.05). Also, feed conversion ratio in the P+V group was lower than other trails (p〈0.05). In addition, white blood cell count (WBC) in groups V and P+V were significantly higher than in both P and C groups (p〈0.05). Also, PCV was significantly increased (p〈0.05) in both P and P+V groups compared to group V and the control group. No significant difference (p〉0.05) was found in RBC, Hb, MCH, MCHC and MCV levels among the different experimental groups. These data show that application of L. plantarum could be a benefit of vaccinated trout, enhancing fish growth and haematological parameters.

    Description: 8-week feeding trial was conducted for juvenile rainbow trout (Oncorhynchus mykiss) to determine the optimal amount of Tenebrio molitor meal (TM) that could replace fishmeal in feeds without reducing growth performance, hematological indices and non-specific immune responses. Five isonitrogenous and isolipidic diets were formulated with TM replacing 0% (Control), 25% (TM25), 50% (TM50), 75% (TM75) and 100% (TM100) fishmeal respectively. Each diet was randomly divided into triplicate groups with 20 juveniles (initial weight 10±0.5 g). At the end of the experiment, the mean of the final weight of the fish fed diets with control (41.7±0.7) and diet containing 25% TM powder (43.1±2.1) was significantly higher than other treatments (p〈0.05). The lowest feed conversion ratio was observed in control (0.76±0.1) which had a significant difference compared to TM75 (0.93±0.06) (p〈0.05). Weight gain (WG) was affected by dietary TM inclusion and decreased with increasing TM. Red blood cells was not affected by treatment, while hemoglobin content and hematocrit decreased with increasing TM inclusion levels (p〈0.05). Plasma complement activity, the IgM level and cortisol were not affected by dietary TM. Dietary treatments had no significant effects on plasma glucose and triglyceride of fish compared to the control (p〈0.05). Plasma total protein, albumin and globulin were significantly affected by dietary TM (p〈0.05). Plasma cortisol levels and alternative complement (ACH50) pathway activity did not show any significant difference among treatments (p〉0.05). According to a broken-line model, the recommended substituted ratio of TM for fish meal was estimated to be 25% for rainbow trout obtaining maximum weight gain.

    Description: Due to lack of access to comprehensive and up-to-date information regarding equipment, personnel, laboratory services tariffs and chemicals in the laboratories of Iran Fisheries Research Institute and felt the necessity of having this information for optimal management of the laboratories, it was decided with regard to all information needs in this area and using previous experiences in this regard as well as the experiences of other institutions and related organizations at the national (and even international) level, the system would be designed. The main purpose of implementing this plan was to establish and implement a software system based on web technology to create a comprehensive database of the Fisheries Research Institute's laboratories, so that in addition to organizing information, using periodic reports on various related areas a more appropriate monitoring and management system could be applied. Fortunately, after the design and implementation stages, the system in early January 2011 was fully applicable. The main parts of this system include portal; database; advanced data entry, search and reporting capabilities; direct tests order system and discussion forums.

    Description: Some biological aspects of yellowfin and skipjack tuna according to catch data of traditional gillnet fisheries were studied in the north of Oman Sea during 1997-99. Based on Gonadosomatic Index (GSI) results, the spawning period or yellowfin begins in spring and reaches to the peak level in May-June. For skipjack tuna, in addition to the above mentioned spawning period, the other observed period was found in autumn-winter with a peak in January-February. The first length or maturity (LM50%) of yellowfin and skipjack tuna females were estimated 76 and 61 cm, respectively. Observation of stomach contents showed that squid was the major components of diet for yellowfin and fish for skipjack tuna. A comparison between mean length of different years and landing sites using two-way analysis of variance indicated a significant differences, The length-weight equations showed an algometric growth in tuna and isometric growth in skipjack.
