    Beschreibung: Human pathogens impact patient health through a complex interplay with the host, but models to study the role of host genetics in this process are limited. Human induced pluripotent stem cells (iPSCs) offer the ability to produce host-specific differentiated cells and thus have the potential to transform the study of infectious disease; however, no iPSC models of infectious disease have been described. Here we report that hepatocyte-like cells derived from iPSCs support the entire life cycle of hepatitis C virus, including inflammatory responses to infection, enabling studies of how host genetics impact viral pathogenesis.

    Beschreibung: The expression of human immunodeficiency virus (HIV) after T cell activation is regulated by NF-kappa B, an inducible DNA-binding protein that stimulates transcription. Proteins encoded by a variety of DNA viruses are also able to activate expression from the HIV enhancer. To determine how this activation occurs, specific genes from herpes simplex virus type 1 and adenovirus that activate HIV in T lymphoma cells have been identified. The cis-acting regulatory sequences in the HIV enhancer that mediate their effect have also been characterized. The relevant genes are those for ICP0-an immediate-early product of herpes simplex virus type 1-and the form of E1A encoded by the 13S messenger RNA of adenovirus. Activation of HIV by adenovirus E1A was found to depend on the TATA box, whereas herpesvirus ICP0 did not work through a single defined cis-acting element.     Beschreibung: The liquid-vapor interface of a bismuth-gallium mixture (0.2 percent bismuth and 99.8 percent gallium) at 36 degrees C has been studied by grazing incidence x-ray diffraction. The data show, in agreement with thermodynamic arguments, that bismuth is heavily concentrated in the liquid-vapor interface. The x-ray diffraction data are interpreted with the assistance of a simple model that represents the interface as a partial monolayer of bismuth.     Notizen: Abstract Using numerical methods, the initial rates of oxygen uptake by the red blood cell have been computed. The methods accommodate both a water layer and membrane which may act as diffusive impedance to gas influx. The differential solubilities of the gas in these two layers have also been incorporated in the methods. The presence of a 0.50–0.65 μm deoxygenated water layer has been calculated to simulate the experimental results by Roughton (1959). Experimental studies of CO and NO uptake by the red cell could also be simulated. Although a membrane-only model with given parameters can also account for the observed rates of oxygenation of the red cell (Weingardenet al., submitted for publication), the additional incorporation of differential solubilities of oxygen in the different layers of the RBC yields results that indicate a three layer model to be more plausible. Using a thin layer-red cell oxygenation system, the rates of oxygenation were determined for red cells surrounded by a 4.2 μm deoxygenated water layer. The rates were found to compare favorably to the results of the theoretical model.
