    Notes: A simple isotachophoretic (ITP) preconcentration column for subsequent online capillary zone electrophoresis is described, and its use for enhancing detectability in capillary zone electrophoresis (CZE) is evaluated. Sample preconcentration using ITP is shown to enhance detectability in CZE by several orders of magnitude. Experimental verification of the detectability gain is demonstrated for cationic and anionic separations; an increase in detectability of more than 200-fold was achieved. The practical applicability of this method is demonstrated using a direct analysis of thiamine in human blood.

    Notes: An isotachophoretic method was developed and optimized for simultaneous determination of pyruvate, acetoacetate, lactate, and 3-hydroxybutyrate in 1-10 μL of untreated heparin plasma of patients with diabetes mellitus. A commercial instrument with a column switching system was used. The preseparation capillary was filled with the leading electrolyte of pH 4.2 and the analytical capillary was filled with leading electrolyte of pH 3.0. The concentrations of individual ketone bodies and lactate found in human plasma varied within 0.01-1.4 mmol/L and 0.8-11.8 mmol/L, respectively. Analysis time was about 30 min.

    Notes: Seven representatives of the serogroup B Staphylococcus aureus bacteriophages, 29, 53, 55, 83A, 85, φ11 and 80α, were examined by capillary electrophoresis (CE) for genomic homology using DNA restriction analysis. Genomic DNA of individual bacteriophages was cleaved by HindIII restriction endonuclease, and the resulting restriction fragments were separated by standard horizontal agarose slab gel electrophoresis (SGE) as well as by CE in low-melting-point agarose solutions. The number and size of restriction fragments identified by both methods were compared. The high separation power of CE makes it possible to extend the restriction fragment patterns. In most of the restriction patterns, some additional restriction fragments as small as 150 bp, not identified by SGE, were detected. With respect to speed, high separation efficiency, low sample consumption and automation, CE offers a simple procedure for processing of multiple samples cost-effectively in a reasonable time. The comparison of the complemented restriction patterns of the different phage strains and the subsequent identification of their common fragments leads to a deeper understanding of their phylogenetic relationships. The genome homologies expressed for individual phage pairs in terms of coefficient F values ranged from 15 to 69%. These values are in good accordance with the degree of DNA homology of these phages as determined by DNA hybridization studies and thermal denaturation analysis of DNA by other authors. The total size of each phage genome was estimated by adding the sizes of individual restriction fragments.

    Notes: Capillary zone electrophoresis was optimized for the separation of thiouracil, methylthiouracil and propylthiouracil. Methylthiouracil could be determined in various types of urine (human, bovine, horse), either without any pretreatment or in ethyl acetate extracts, within 15 min. For identification, the simultaneous detection at three UV wavelengths (216, 245 and 278 nm) was advantageously used while for quantification the wavelength of the absorbance maximum at 278 nm was preferred. Under optimized conditions a linear response of the detector in the concentration range 0.1-100 ppm was obtained. On analysis of untreated urine, a detection limit of 0.5 ppm was found; for urine extracts the detection limit was 0.1 ppm. Univocal peak identification, based on absorption at three wavelength, was only possible above 2 ppm. Relative standard deviation for standard solutions of methylthiouracil, diluted in the background electrolyte, was 1%; for methylthiouracil in extracts dissolved in the background electrolyte it was 4.5%, and for methylthiouracil in untreated urine, 12.7%.

    Notes: A new preparative electrophoretic method in free solution is described, consisting of three consecutive steps: (i) filling the separation chamber with a suitable electrolyte system and sample in parallel streams by laminar hydrodynamic flow, (ii) applying the voltage across the chamber with isotachophoretic separation for a definite time interval operating in the direction perpendicular to that of filling, (iii) reapplying a hydrodynamic flow (without voltage) and collecting the separated species via an array of outlets. This approach completely eliminates the main drawback of the continuous flow electrophoresis (CFE), i.e., the electrohydrodynamic distortion of zones. This method utilizes the instrumentation devised for CFE and enables the isolation of large amounts of individual sample species comparable to that of CFE, with a resolution comparable to that of capillary isotachophoresis. The precise timing of the consecutive steps in the procedure as well as the stability of the operational parameters are of key importance for reproducibility. By using cationic isotachophoresis with 3 synthetic pI markers as model sample species, the reproducibility, stability and the separation power of the newly presented method are demonstrated. The sample throughput corresponds to micromoles per hour.

    Notes: A method for the determination of amprolium in feed stuff by analytical capillary isotachophoresis with column coupling is described. Up to 20 μl of the raw water extract of amprolium from a feed stuff can be analysed directly without any pretreatment in 15 min. The detection limit reached the level of ppm, the reproducibility of analyses in the followed range of 0.2-1.2 nmoles was 3 rel. %.
