    Notizen: A broad empirical study of nine independent sets of data on fluidized-bed heat transfer is presented, with correlation of the data in two groups. A wide range of the many variables is covered, and some data on commercial units are included. Data for external (i.e., walls of the fluidizing vessel) and internal (i.e., tubes in the bed) heat transfer surfaces are correlated graphically. The correlations indicate the importance of heat transport by the mobile particles and of unsteady state conduction in the gas.

    Notizen: The fusion in vitro of embryonic myoblasts to form multinucleated myotubes requires the addition of serum to a basal nutrient medium. The serum requirement for fusion can be satisfied by insulin with somatotropin potentiating its effect. Myotubes formed under these conditions fail to differentiate to cross-striated, spontaneously contractile muscle fibers. This arrest of development is reversible if serum is restored to the medium.Development of the enzyme glycogen synthetase was studied as an additional indicator of muscle differentiation. In cultures developing in the presence of serum, this enzyme was demonstrated by autoradiography to be highly concentrated in myotubes as compared to mononuclear cells. The activity of the enzyme remains low in (1) cultures formed in response to insulin and somatotropin in the absence of serum, as well as (2) in cultures formed in unsupplemented basal medium which are virtually lacking in myotubes. The addition of serum to (1) restores the development of this enzyme. Serum which has been extensively digested with the proteolytic preparation, pronase, and subjected to boiling temperature, when combined with insulin and somatotropin is also capable of promoting the development of glycogen synthetase to a specific activity which exceeds the control. The serum factor is not lost on exhaustive dialysis, nor can enzyme promoting activity be liberated by heat denaturation of serum proteins.

    Notizen: Selenium-deficient male mice were obtained by feeding a selenium-deficient diet for three successive generations to Swiss-Webster mice. Examination of epididymal sperm by transmission electron microscopy revealed progressively increasing alterations in the shape and arrangement of mitochondria within the midpiece. Other midpiece anomalies included acute bends, disorientation of the axoneme and dense fibers, and cytoplasmic masses at atypical locations. Some cross sections showed both the principal piece and midpiece within the same plasma membrane. Negatively stained whole mounts of mitochondrial ghosts prepared from epididymal sperm of normal and first-generation selenium-deficient mice and rats indicated that the selenium-deficient ghosts were smaller, less curved, and more fragile than those of normal sperm mitochondria. Thus, selenium appears to be required for the normal development or stabilization of mitochondria1 shape during spermiogenesis in these rodents.

    Notizen: Three successive generations of mice were fed a Torula yeast based Se-deficient diet with or without 0.1 ppm Se in the drinking water. The Se-deficient mice, in the course of three generations, showed a decrease in body weight, testis weight, epididymal weight, and sperm production. The percentage of morphologically abnormal sperm increased in successive generations. The majority of sperm defects were found in the midpiece region of the tail. Many of these aberrant sperm were motile. A progressive decrease in fertility was noted during the first two generations of Se deficiency. This system confirms the essential role of Se in spermatogenesis and provides a model for the evaluation of the primary effect of Se deprivation on the structural development of sperm.

    Notizen: The keratinous capsules surrounding rat sperm mitochondria were isolated 24 days after intratesticular injections of [75Se] selenite or [35S] cysteine. Dodecyl sulfate-polyacrylamide gel electrophoresis of purified, doubly labeled mitochondrial capsules revealed only a single 75Se-labeled component, whose molecular weight was 17,000, in agreement with previously reported observations obtained with cruder sperm fractions. Most of the 35S label and the major zone of stained protein on the gels coincided with the position of 75Se, suggesting that selenium is associated with a cysteine-rich structural protein. The level of selenium in rat sperm, 195 ± 3.2 ng/108 sperm (approximately 30 ppm), determined by hydride generation and atomic absorption spectrophotometry, is consistent with a structural function for this trace element in the sperm.
