    Description / Table of Contents: Summary Metabolically healthy subjects were given an intravenous injection of 1,13 mg14C-labelled glibenclamide (HB 419). The plasma level, renal elimination of the radioactivity and metabolism of the substance were investigated. Two minutes after administration the HB 419 is virtually present only in the blood and at the end of the distribution period mostly in the extracellular space. 53% of the radioactivity is excreted via the kidneys in the form of metabolites. If glibenclamide is administrated in the same way to the same subjects after pretreatment with phenylbutazone there are no differences in the course of the plasma levels or the rate of elimination from the blood. There is, however, a significant difference in the excretion of the radioactivity in the urine. In the presence of phenylbutazone significantly less HB 419 metabolite is excreted renally. In view of the known alternative route of elimination it is suggested that the amount not excreted in the urine is in compensation eliminated via the bile. There was no difference in the metabolism of glibenclamide between the control and phenylbutazone treated groups. The potentiation by phenylbutazone of HB 419 action, and probably also that of other antidiabetic sulphonylureas, must therefore be due predominantly to other causes (Communication III).
    Notes: Zusammenfassung Stoffwechselgesunde Versuchspersonen erhielten14C-markiertes Glibenclamid in einer Dosis von 1.13 mg/Vpn i.v. gespritzt. Plasmaspiegelverläufe, renale Elimination der Radioaktivität und die Metabolisierung der Substanz wurden untersucht. Zwei Minuten nach der Applikation ist HB 419 praktisch nur im Blutund nach Abschluß der Verteilung weitgehend im Extracellulärraum vorhanden. 53% der Radioaktivität werden über die Nieren in Form von Metaboliten ausgeschieden. Wird den gleichen Probanden nach Prämedikation mit Phenylbutazon Glibenclamid in gleicher Weise verabfolgt, ergibt sich kein Unterschied hinsichtlich der Plasmaspiegelverläufe und der Eliminationsgeschwindigkeit aus dem Blut. Ein signifikanter Unterschied besteht jedoch in der Ausscheidung der Radioaktivität in den Harn (26.3%). In Gegenwart von Phenylbutazon wird ein signifikant geringerer Anteil von HB 419-Metaboliten renal eliminiert. Aufgrund des bekannten zweiten Ausscheidungsweges wird vermutet, daß der fehlende Anteil kompensatorisch über die Galle eliminiert wird. Die Metabolisierung von Glibenclamid weist keine Differenzen zwischen Phenylbutazon-und Kontroll-Gruppe auf. Die Wirkungspotenzierung von HB 419 — wahrscheinlich auch diejenige anderer antidiabetisch wirksamer Sulfonylharnstoffe — durch Phenylbutazon dürfte demnach überweigend andere Ursachen haben.     Notes: Nascent high-molecular-weight (UHMW) polyethylene (PE) samples of different origins show a rather high crystallinity of about 70-75% and contain both a major portion of orthorhombic extended chain crystallites and a minor portion of triclinic crystallites. The triclinic content is greater the higher the molecular weight of the sample and the higher the activity of the used catalyser. A melting / recooling treatment results in a reduction of crystallinity by about 15-25% and disappearance of the triclinic phase. Further, an irreversible conversion of nascent orthorhombic extended chain crystallites to orthorhombic folded chain crystallites of increased lateral dimensions and crystalline perfections takes place during the melting / recooling treatment. The results are compared to those obtained for lower-molecular-weight PE samples and for high-strength / modulus PE fibers of different origins.

    Description / Table of Contents: The supramolecular structure of compact polyamide-6 was investigated by electron microscopy with ultrathin sections using different methods of solidification and contrasting. Besides usual transmission electron microscopy the high voltage electron microscopy was applied which allows to investigate thick sections. Comparative investigations were performed with thin films. The changes in morphology caused by high pressure in the crystallization of polyamide-6 are demonstrated by fracture surface replication as well as by scanning electron microscopy.
    Notes: Zur Charakterisierung der übermolekularen Struktur von kompaktem Polyamid-6 wurden elektronenmikroskopische Untersuchungen an Ultradünnschnitten unter Anwendung verschiedener Härtungs-und Kontrastierungsmethoden durchgeführt. Neben der konventionellen Transmissionselektronenmikroskopie wurde die Höchstspannungselektronenmikroskopie eingesetzt, die es ermöglicht, dickere Schnitte zu untersuchen. Vergleichende Untersuchungen wurden an Dünnfilmen vorgenommen. Die durch den Einfluß hohen Druckes bei der Kristallisation von PA-6 verursachten morphologischen Veränderungen wurden sowohl anhand von Bruchflächenabdrücken als auch rasterelektronenmikroskpisch nachgewiesen.

    Description / Table of Contents: With respect to the particular properties of polymers the specimen preparation and observation in electron microscopic investigations require specific techniques and great attention in their application. By representative examples the scope and problems of structural investigations of polymers as well as the possibilities and efficiency of electron microscopic techniques are reviewed. Particularly, the different structures revealed by transmission electron microscopy with various polymer materials are described and the problems of sample preparation, observation techniques and information obtained are considered.
    Notes: Bei der elektronenmikroskopischen Untersuchung von organischen polymeren Stoffen erfordert die Präparations-und Untersuchungstechnik auf Grund der spezifischen Eigenschaften der Polymere besondere Aufmerksamkeit. In Form einer Übersicht werden anhand von repräsentativen Beispielen einerseits die Mannigfaltigkeit von Polymerstrukturen und die Problematik bei ihrer Untersuchung und andererseits die Möglichkeiten und das Leistungsvermögen elektronenmikroskopischer Untersuchungen von Polymersystemen dargelegt. Im einzelnen beschäftigt sich der Beitrag mit verschiedensten Polymerstrukturen, die im wesentlichen mit der Transmissionselektronenmikroskopie an unterschiedlichen polymeren Materialien erhalten wurden, und zeigt dabei polymerspezifische Probleme der Probenpräparation und Untersuchungsmethodik sowie den möglichen Informationsgehalt auf.

    Description / Table of Contents: The film formation, annealing and drawing properties of thin films of PA-6, PETP and PA-6/PETP have been investigated by scanning electron microscopy. The film formation of unipolymer systems was found to be essentially affected by the substrate. Contrarily, in the film formation from multicomponent polymer systems the intermolecular forces of the components play the most important part and superimpose the effect of the substrate on the morphological structure being formed. Annealing of the films causes rearrangement of the morphological structure and affects the deformation behaviour.
    Notes: Es wird über rasterelektronenmikroskopische Untersuchungen zum Filmbildungs-, Temperungs- und Reckverhalten von dünnen Polyamid-6-, Polyethylenterephthalat- und Polyamid-6/Polyethylenterephthalat-Filmen berichtet. Die Untersuchungen zeigten, daß die Filmbildung unipolymerer Systeme im wesentlichen durch das Substrat beeinflußt wird. Im Gegensatz dazu spielen bei der Filmbildung aus polymeren Mehrkomponentensystemen die zwischenmolekularen Kräfte der einzelnen Polymerkomponenten die entscheidende Rolle und überlagern den Einfluß des Substrates auf die sich ausbildende morphologische Struktur. Durch Temperbehandlungen der Filme wird eine Reorganisation der morphologischen Struktur bewirkt und das Deformationsverhalten der untersuchten Dünnfilme mitgeprägt.
