Notes: Cosmetic creams were analyzed for their content of jojoba wax and shea fat using anticircular HPTLC, a fast, precise, and reliable method. After optimization of the solvent system (petrol ether/diethyl ether) for the separation of jojoba wax and shea fat, the spots were visualized with phosphomolybdic acid by the “dip-in” technique, subsequently heated, and finally recorded by scanning with a densitometer at 546 nm. Results showed that one particular concentration range gives an optimum correlation between calibration curves and sample evaluations.

Notes: A preparative anticircular chromatography (PACH) device was constructed to elucidate different applications of separation problems as well as to compare different sample collection techniques in this particular PLC mode. An anticircular PC apparatus had to be established which allowed the utilization of 20 X 20 cm thin-layer plates as well as sampling positioning at any given radius. Typical solvent migration times were 8-12 minutes and ca. 8-10 ml of solvent was needed for an entire 20 cm plate. It was shown that peak broadening effects due to diffusion during long migration could be avoided in the same way as for analytical anticircular PLC on 10 X 10 plates. On the other hand, longer migration distances allow increased resolution, in turn allowing higher sample loads, particularly in the case of trace compound enrichment. A full description of the apparatus is given, including the sampling device.Different sample collection techniques were tried and evaluated, including the through-flow technique by on-line center collection for plate washing as well as classical scraping-off of interesting zones. Further evaluation of the method included quantification and evaluation of consistency by UV/VIS scanning after spot-visualization. Several application examples are given to illustrate the efficiency of the preparative anticircular PLC-system.

Notes: Enantiopure 5,5-disubstituted t-butyl 2-t-butyl-3-methyl-4-oxoimidazolidine-1-carboxylates readily available by diastereoselective double alkylation of the parent compound (Boc-BMI) can be converted to α-branched α-amino acids with two bulky substituents (PhCH2/Et, PhCH2/i-Pr, PhCH2/CH2C6H11) in four simple steps: hydrolysis to the amino acid amides, N-benzoylation, cleavage of the amino acid amide group, and debenzoylation (Scheme 3, products 6-10, overall yields 〉50%). This procedure constitutes an extension of the scope of amino acid synthesis from (R)- and (S)-Boc-BMI.

Abstract: A thick layer of a sol of vanadium pentoxide with thread-like particles has been put on a glass slide. The electron-microscopic examination by means of the polystyrenesilica-replica-method shows the single threads in the surface of the dried layer. Only this method reproduces form and size of the threads somewhat distorted.

Notes: The Cu2+ complex of 1, having a non-coordinating carboxylic group, can be reacted under the typical conditions of peptide formation with amines such as 2-methylpropylamine or (pyrid-2-yl)methylamine to give, after removal of Cu2+ with CN-, the amides 4 and 5. The Cu2+ ion is of paramount importance since it protects the four amino groups of the macrocycle so that the amide condensation can specifically be done with the exogenous amine It is also shown that the Cu2+ complex of 1 can be covalently attached to bovine serum albumin (BSA), thus opening the possibility to use this compound as a labelling agent for proteins and antibodies.

Notes: Both enantiomers of tert-butyl 2-(tert-butyl)-5,5-dimethyl-4-oxoimidazolidine-1-carboxylate (11; Bbdmoic) were prepared from L-alanine (Schemes 1 and 2). The parent heterocycle, 2-tert-butyl-5,5-dimethylimidazolidin-4-one (12; from 2-aminoisobutyramide, H-Aib-NH2, and pivalaldehyde) was also available in both enantiomeric forms by resolution with O,O′-dibenzoyltartaric acid. The compound (R)- or (S)-11 was used as an auxiliary, but also as a chiral Aib building block in a dipeptide synthesis. The 3-propanoyl derivative 13 of (R)-11 was used for the preparation of enantiomerically pure 2-methyl-3-phenylpropanoic acid (enantiomer ratio (e.r.) 99.5:0.5), by benzylation of the Zn-enolate (→ 14; Scheme 3). Oxidative coupling of the bis-enolate derived from heptanedioic acid and (S)-11 (→ 23) and methanolysis of the auxiliary gave dimethyl trans-cyclopentane-1,2-dicarboxylate (26) with an e.r. of 93:7 (Scheme 5, Fig. 5). The 3-(Boc-Gly)-Bbdmoic derivative 29 was doubly deprotonated and, after addition of ZnBr2 alkylated with alkyl, benzyl, or allyl halides to give the higher amino-acid derivatives with excellent selectivities (e.r. 〉 99.5:0.5, Schemes 6 and 7). Michael additions of cuprates to [(E)-MeCH=CHCO]-Bbdmoic 36 occurred in high yields, but high diastereoselectivities were only observed with aryl cuprates (diastereoisomer ratio (d.r.) 99:1 for R = Ph, Scheme 8). Finally, 3-(Boc-CH2)-Bbdmoic 17 was alkylated through the ester Li-enolate with primary and secondary alkyl, allyl, and benzyl halides with diastereoselectivities (ds) ranging from 91 to 98%, giving acetals of Boc-Aib-Xxx-O(t-Bu) dipeptides (Scheme 4). The effectiveness of Bbdmoic is compared with that of other chiral auxiliaries previously used for the same types of transformations.
