    Notes: Enzyme-purified elastin from bovine ligamentum nuchae was digested with elastase in the presence of sodium dodecyl sulfate. Chromatographic fractionation of the digest, after removal of the detergent, resulted in the high-yield isolation of two peptide fractions (F2 and F3) that differed in size and composition. The larger, F2, which accounted for about 55% of the starting material, was subjected to sedimentation-equilibrium analysis in three chaotropic solvents. Comparison of the distribution of point-average molecular weights (Mw and Mz) with protein concentration in the three systems lead to the conclusion that significant self-association of peptides occurred in the absence of 6M guanidinium hydrochloride. In this solvent, the molecular-weight distribution was between 25,000 and 34,000, a range of values in agreement with an intrinisic viscosity of 13.1 cc g-1 determined in the same solvent. Assessment of chain weight by N- and C-terminal analysis was consistent with F2 being a multichain molecule comprising four polypeptide chains linked by three polyfunctional amino acids. Results are interpreted in terms of an anisotropic ultrastructural model of the protein, in which four polypeptide chains constitute the primary filament visualized by electron microscopy in the intact fiber.

    Notes: Debarking wastewaters of the forest industry contain high concentrations of tannins that are inhibitory to methane bacteria. The tannins can be polymerized to nontoxic colored compounds by the applications of an autoxidation pretreatment, enabling the anaerobic treatment of easily biodegradable components in the wastewater. The continuous anaerobic treatment of untreated and autoxidized pine bark extract was studied in laboratory-scale columns packed with a granular sludge bed. The autoxidation doubled the conversion efficiency of bark extract COD to methane (from 19 to 40%). After 5 months of operation, anaerobic treatment of the autoxidized extracts was feasible at high influent concentrations (14 g COD/L) and loading rates (26 g biodegradable COD/L · d) with 98% elimination of the biodegradable fraction. The detoxification pretreatment polymerized the toxic tannins to poorly biodegradable high molecular weight tannins and humic compounds which were not eliminated during anaerobic treatment. Although the original tannins of the untreated extract were eliminated by 60%, they were not biodegraded to volatile fatty acids and methane but instead were transformed to phenolic degradation intermediates (phenol, p-cresol, 3-phenyl-propionate, and carboxycyclohexane). Therefore, the autoxidation pretreatment did not decrease the content of readily biodegradable substrates which accounted for 53% of the extract COD. The recalcitrant COD expected in the effluents of reactors treating autoxidized debarking waste-water can be effectively separated by calcium precipitation prior to anaerobic treatment.

    Notes: Infrared absorption spectra were used to study the structural changes resulting from oxidation in a number of unvulcanized, purified polymers under a variety of experimental conditions. The results showed that as oxidation progressed, the structural changes which were detected were qualitatively the same for all of the polymers under all of these experimental conditions. Correlation of volumetric measurements with spectral data indicated that considerably more oxygen was absorbed by the polymer samples before structural changes could be detected than the minimum amount required for the initial detection of the hydroxyl and carbonyl structures. From the interpretation of information from the absorption spectrum, it is believed that the structural evidence is in substantial agreement with current theories on autoxidation. Although the initial hydroperoxide formation is not observed in the spectrum, it is quite probable that this reaction takes place with almost immediate decomposition which results in a net hydroperoxide concentration so low as to make infrared methods insensitive for its detection. Correlation of physical properties with structural studies indicate that little change in these properties take place until the hydroxyl and carbonyl structures become evident in the infrared spectrum.

    Notes: Three complementary two-dimensional systems for the analysis of cereal prolamins are described. These are electrophoresis at pH 3.1 followed by electrophoresis at pH 9.2, isoelectric focusing (IEF) followed by sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) and SDS-PAGE under non-reducing conditions followed by SDS-PAGE with reduction. They together give information on the pIs, Mrs and charge properties of the individual prolamin components, and on their interactions to form oligomers stabilized by inter-chain disulphide bonds. The three systems are then compared for their effectiveness in analysing unreduced prolamin I fractions from wheat and rye, and the IEF/SDS-PAGE system for analysing reduced and pyridylethylated prolamin fractions from all the major cereals. Finally, applications of the systems in biochemical and genetic studies are discussed and illustrated with three examples: analysis of the structural relationships of the S-rich prolamins (B hordeins and γ-type hordeins) of barley, determination of the interactions of wheat and rye prolamin subunits in a 2RS/2BL translocation line, and the mapping of genes for α-type prolamins in the wild grass Haynaldia villosa.

    Notes: A photographic study, at microsecond framing rates, has been made of the impact behaviour of thin layer samples of emulsion explosives. The experiments were performed using a drop-weight apparatus with transparent anvils. The effects of incorporating aluminium granules (∼100 μm in size) and glass microspheres (50 μm to 150 μm diameter) in the emulsion matrix were also studied. It was possible to record the flow of the emulsion both with and without additives. Of particular interest were observations of the onset and development of crystallization in the layer. In no case was there evidence for initiation of explosion.

    Description: Studies of global change and other regional issues depend on ecological data collected at multiple study areas or sites. An information system model is proposed for compiling diverse data from dispersed sources so that the data are consistent, complete, and readily available. The model includes investigators who collect and analyze field measurements, science teams that synthesize data, a project information system that collates data, a data archive center that distributes data to secondary users, and a master data directory that provides broader searching opportunities. Special attention to format consistency is required, such as units of measure, spatial coordinates, dates, and notation for missing values. Often data may need to be enhanced by estimating missing values, aggregating to common temporal units, or adding other related data such as climatic and soils data. Full documentation, an efficient data distribution mechanism, and an equitable way to acknowledge the original source of data are also required.
