    Notes: The fluorescence behavior of the 4-phenoxymethylstilbene chromophore in the free state and when bound to a polymer backbone was studied using the model compound 4-(4-ethylphenoxymethyl)stilbene, the homopolymer poly[4-(4-stilbenylmethoxy)styrene] and two random copolymers of styrene and 4-(4-stilbenylmethoxy)styrene in which the styrene/chromophore mole ratios were 1/1 and 2/1, respectively. A strong dependence on concentration and excitation wavelength was observed for the fluorescence behavior of the free and bound chromophores, both in solution and in the solid state. The overall results lead to a proposed mechanism for the emission in which the chromophore forms a fluorescent dimer in the ground state. By studying the effect of solvent power variations it was concluded also that the association is intramacromolecular and occurs between nearest and/or non-nearest neighbors in the chain. The excitation spectra are consistent with these assumptions, and the corresponding electroluminescence spectra show that the associated chromophores are the active species.

    Description: Although genomic instability, epigenetic abnormality, and gene expression dysregulation are hallmarks of colorectal cancer, these features have not been simultaneously analyzed at single-cell resolution. Using optimized single-cell multiomics sequencing together with multiregional sampling of the primary tumor and lymphatic and distant metastases, we developed insights beyond intratumoral heterogeneity. Genome-wide DNA methylation levels were relatively consistent within a single genetic sublineage. The genome-wide DNA demethylation patterns of cancer cells were consistent in all 10 patients whose DNA we sequenced. The cancer cells’ DNA demethylation degrees clearly correlated with the densities of the heterochromatin-associated histone modification H3K9me3 of normal tissue and those of repetitive element long interspersed nuclear element 1. Our work demonstrates the feasibility of reconstructing genetic lineages and tracing their epigenomic and transcriptomic dynamics with single-cell multiomics sequencing.

    Description: Huang and Xu argue that the cyclo -N 5 – ion in (N 5 ) 6 (H 3 O) 3 (NH 4 ) 4 Cl we described in our report is theoretically unfavorable and is instead protonated. Their conclusion is invalid, as they use an improper method to assess the proton transfer in a solid crystal structure. We present an in-depth experimental and theoretical analysis of (N 5 ) 6 (H 3 O) 3 (NH 4 ) 4 Cl that supports the results in the original paper.
