    Notes: TSH is a heterodimeric glycoprotein hormone, whose dissociated subunits are without biological activity. This has precluded the assessment of the relative contribution of each subunit to hormone action. We have raised anti-idiotypes to monoclonal antibodies specific, respectively, for the α and β hTSH subunits. The anti-β anti-idiotype inhibited l25I-hTSH binding to the β subunit-specific monoclonal quantitatively, whereas 125I-hTSH binding to the α subunit-specific monoclonal was not inhibited by anti-α anti-idiotypes, suggesting that only the former is an “internal image” anti-idiotype. Neither of the two anti-idiotypes nor equimolar mixtures thereof inhibited 125I-bTSH binding to thyroid membranes, even though radiolabelled anti-idiotypes showed saturable binding to thyroid plasma membrane which was inhibited 41-65% by bTSH. Each anti-idiotype alone caused 9% inhibition (compared to 50% by NRIgG) of thyroid plasma membrane adenylate cyclase. Equimolar mixtures (125 μg/ml IgG of each anti-idiotype) induced enzyme activity equivalent to 85% of that of 250 mU/ml of TSH. The TSH-like action of the two anti-idiotypes was also reflected in their capacity to increase (450% by 250 μg/ml IgG compared to normal rabbit IgG) the uptake of 131I into isolated thyrocytes and to promote the organization of such cells into follicular structures. At 250 μg/ml, anti-β anti-idiotype promoted the organization of small follicles and only at a concentration of 500 μg/ml did it enhance 131I uptake. Anti α idiotype was without effect in both assays. Lastly, mixture of anti-idiotypes bound to the ∼ 197,000 Mr band (TSH holoreceptor) on protein blots of thyroid plasma membranes resolved on NaDSO4-polyacrylamide gel electrophoresis under non-reducing conditions. Individual anti-idiotypes were without effect.The TSH α and β subunits apparently deliver two cooperative signals to the receptor and that specificity is associated with the β subunit, while the α subunit is important in enhancing receptor affinity for the heterodimer and in stablizing TSH-receptor complex.

    Notes: We raised an antihuman thyrotropin anti-idiotypic antibody and showed that it was active at the thyrotropin receptor. Thus this antibody inhibited 125I b-TSH binding to thyroid plasma membranes, stimulated adenylate cyclase activity through a guanyl nucleotide-dependent mechanism, increased radioiodide entry rate into isolated porcine thyroid follicular cells, and induced such cultured cells to organize into follicles. All these parameters are typical of thyrotropin action. This work raises the possibility that thyroid stimulating antibodies that cause the hyperthyroidism of Graves disease may be, at least in some patients, anti-thyrotropin anti-idiotypic antibodies. It also offers a novel method whereby antireceptor antibodies used in the isolation and characterization of the receptor may be raised from ligands.

    Notes: A mechanical model consisting of three elements (elastic, viscoelastic, and plastic) is proposed to describe the deformational behavior of a polymeric membrane operating in a pressure-driven process. The changes in the membrane thickness are proposed to be calculated from the experimentally obtained flux data.     Notes: Twelve different formulations were developed with urethane acrylate oligomer in combination with a number of functional monomers in the presence of different additives, fillers, and plasticizers. Hessian cloth (jute product) was coated with these formulated solutions and cured under UV radiation. Enhancement of tensile strength and elongation was determined. Loss of these properties due to simulating weathering was also measured. The treated Hessian cloths were buried in soil, water, and mud for a long period and it was found that the samples put in mud were completely degradable while those treated in water and soil retained their tensile properties. © 1995 John Wiley & Sons, Inc.

    Notes: The polyethylene membranes are hydrophilized under the action of chlorosulfonic acid. Such a hydrophilized membrane shows much better water permeability in comparison with the unmodified one. The ion-exchange capacity of the sulfonated membrane is taken as a measure for the hydrophilicity. The influences of the concentration of the sulfonating agent, temperature, and reaction time, on the ion-exchange capacity and water permeability, are investigated. It is shown that the increase in the degree of sulfonation does not always lead to an increase in the water permeability of the membrane. A lower concentration of the sulfonating agent and a lower temperature are to be chosen, so that only the surface and the inner walls of the pores are sulfonated. Sulfonation in the bulk leads to the deterioration of mechanical properties of the membrane without having any contribution to the increase in the water permeability of the membrane.

    Notes: Penbutolol is a β-adrenoceptor blocking agent, and it contains the clinically relevant (-)-S-enantiomer. It was reported that the (+)-R-enantiomer of penbutolol is pharmacologically 50 times less active than the (-)-S-isomer in β-sympatholysis and without intrinsic sympathomimetic activity and refractory period in the heart muscle. Furthermore, the (+)-R-enantiomer does possess mutagenic activity.A high-performance liquid chromatographic (HPLC) method is described for direct identification, stereochemical separation, and quantitation of (+)-R-enantiomer in the clinically used (-)-S-isomer. The method involves the use of cellulose tris-3,5-dimethylphenyl carbamate chiral stationary phase coated on silica gel (OD-Chiralcel column). The capacity factors (k′) for the first eluted enantiomer and stereochemical separation factor (α) obtained were 1.32 and 1.98, respectively. The maximum stereochemical resolution factor (R) was 5.05. The method could be applied for optical purity determination of (-)-(S)-penbutolol in pharmaceutical formulation to detect for the presence of the undesirable (+)-R-enantiomer.

    Notes: The deformational behavior of the porous polymeric tube in the operational period has been studied. The tubes are prepared from a polymeric composition consisting of low-density polyethylene and vulcanized rubber particles. It is established that during operation the tubes undergo only radial elastic deformation. A threshold pressure is necessary to initiate the radial deformation. At a constant pressure, the radial deformation increases with an increase in the polyethylene content in the tube material. The stress necessary to break the tubes during operation is much lower than the tensile strength.

    Notes: The pore-formation mechanism in filled polyethylene film during the process of thermomechanical deformation has been studied. The film material consists of a high density polyethylene and SiO2. The rate of deformation as well as the initial thickness of the polyethylene film play a vital role in the formation of the porous structure. With an increase in the deformation rate, the contribution of crazes in the film decreases. Keeping all the parameters constant, with the increase in the initial film thickness, the porosity of the membranes decreases. The mechanism of pore formation is observed by scanning electron microscopy. It is established that the formation of crazes in the deformation process starts from the zones with lower adhesion of the filler with polymer and in the course of the process propagates throughout the mass. The final pore-size distributions as well as the permeability data provide additional indirect evidence in favor of the observed mechanism.

    Notes: The environmentally sensitive properties of polyethylene membranes filled with SiO2 have been studied. The porous characteristics of the membranes is investigated with the aqueous solutions of pH 1-8 and aqueous electrolyte solutions with concentrations of 10-5-10-1 mol/L. The experimental results are explained by the concept of coagulation-swelling behavior of the filler under different environmental conditions. The relaxation of membrane resistance with a change in pH is also studied.
