    Notes: Using gas chromatography/mass spectrometry we have observed six peaks in the single ion chromatograph at m/z 674 of the alkaline extract of barley straw cell walls. The breakdown pattern of these ions suggests that they could all be isomers of 5,5′-bis-dehydroferulic acid. Since only the 5,5′ linkage is reported to be present in plants this tentatively suggests that these compounds exhibit stable orientational isomerism about this bond. Plausible breakdown mechanisms are presented which allow for the complete assignment of structure to all the isomers. These mechanisms suggest that the two pairs of breakdown ions at m/z 556 and 467, and 265 and 193 are representative of trans and cis double bonds, respectively.

    Notes: The benefits obtained from the addition of small quantities of chlorinated polyolefins to paving grade asphalt binders were investigated. A chlorinated polyethylene plastomer, Tyrin 2552, and a chlorinated olefinic elastomer, Tyrin CM0730, were added to asphalt binders at 3 and 5 wt % and subsequently reactively processed to facilitate compatibilization. The mixtures were analyzed for rheological performance relating to fatigue and rutting as well as low-temperature fracture performance. The addition of small quantities of these polymers to the asphalt binders resulted in significant improvements over conventional modifiers at both the high and low temperature extremes. © 1994 John Wiley & Sons, Inc.

    Notes: Stability of heterologous protein expression during production is critical for regulatory approval of vaccine and therapeutic products. Leishmania GP63, a zinc metalloproteinase that is a potential vaccine candidate, has been expressed on the surface of Chinese hamster ovary (CHO) cells. Flow cytometry was used to follow the stability of GP63 expression. Expression of proteolytically active GP63 (GP63WT) was unstable whether or not methotrexate (MTX) selection was maintained. In contrast, expression of an active site mutant (GP63E265D) was stable under MTX selection. In the absence of selection, the decline in GP63E265D expression was more gradual than the loss of GP63WT expression. Different molecular mechanisms accounted for these losses and resulted in higher growth rate nonproducer populations. A dynamic population model was used to calculate the conversion rates of GP63WT producers to nonproducers. © 1997 John Wiley & Sons, Inc. Biotechnol Bioeng 53: 594-600, 1997.

    Notes: Ordered hexagonal arrays of parallel collagen fibers produce the brilliant green structural color of the fleshy, supraorbital caruncles of male Velvet Asity (Philepitta castanea; Aves: Eurylaimidae). The collagen arrays are organized in larger macrofibrils that are packed irregularly within cone-shaped papillae that cover the surface of the caruncle. The color of the caruncle conforms closely to the wavelengths predicted by applying Bragg's Law of constructive reflection to measurements of the size and spatial organization of the collagen arrays. These observations constitute a novel mechanism of structural color production in animals. These collagen arrays are convergently similar to the smaller, highly structured collagen arrays in the mammalian cornea, which exploit the same physical mechanism to produce optical transparency. © 1994 Wiley-Liss, Inc.

    Notes: Linear viscoelastic properties are found to be a sensitive measure of flow-induced structural changes in a block copolymer. Styrene-butadiene-styrene block copolymer (SBS) with 26% polystyrene (PS) forms a macrostructure in the quiescent state with grains of the order of 1-10 μm. Within each grain, phase separation gives rise to a regular two-phase microstructure with cylindrical PS domains with radius of the order of 200 Å. Large-amplitude oscillatory shear (γ = 4.5) at temperatures between 139 and 181°C was applied to after the grain structure with the objectives of removing the discontinuities at the grain boundaries and of aligning the domains into a continuous ultrastructure. The SBS behaved like a solid (tan δ 〈 1 at low ω) before and like a liquid (tan δ 〉 1) after shear modification. This change expressed itself in the removal of the long relaxation times from the linear viscoelastic spectrum; the intermediate and low relaxation times were not affected by the shear modification. The viscoelastic spectrum slowly recovered during annealing with recovery times of the order of the longest relaxation time of the quiescent structure. Birefringence studies showed that the SBS did not recover into its original grain structure but into a highly oriented domain structure. The discontinuities at the grain boundaries could not be removed completely.

    Notes: The importance of having a rapid method for determining the viable biomass in activated-sludge wastewater treatment plants (WWTP) for process control and development is well recognized. The firefly bioluminescence ATP assay has been proposed for this purpose. Such an assay using partially purified firefly luciferase and synthetic firefly luciferin for the bioluminescence reaction, a liquid scintillation counter in the out-of-coincidence mode as luminescence detector, and a sludge ATP extraction technique involving dimethyl sulfoxide at room temperature is described. Experiments with several pure bacteria cultures were done to demonstrate the feasibility of applying this assay to activated sludge to activated sludge WWTP investigation and control. The ATP content of samples taken from various points in a 350000 gal/day brewery activated-sludge WWTP was monitored for 4.5 months. Good linear correlation between ATP and mixed-liquor suspended solids, return sludge suspended solids, and effluent suspended solids were observed. Percentage viabilities of the various sludge samples were derived from the ATP results.

    Notes: The flow field inside an orifice flowmeter with a beta ratio of 0.50 operating at a Reynolds number of 91,100 has been studied using a three-color, 3-D laser Doppler anemometer system. Mean velocity measurements show large radial velocities leading into the orifice, the fluid separating from the orifice plate at the throat, the presence of a vena contracta farther downstream, flow reattachment to the pipe wall 5.3 pipe radii (R) downstream, the presence of a small upstream recirculation zone, and both a primary and secondary recirculation zone downstream of the orifice plate. The static wall pressure distribution attains a minimum pressure at 1.5 R, which does not coincide with the location of the vena contracta (0.75 R). Distributions of the entire Reynolds stress tensor are presented along with calculated values of turbulence kinetic energy, turbulence kinetic energy production, vorticity, and turbulence induced accelerations. These data are analyzed to interpret the complex turbulent flow field.
