    Notes: The lingual epithelium is innervated by special sensory (taste) and general sensory (trigeminal) nerves that transmit information about chemical stimuli introduced into the mouth to the higher brain centers. Understanding the cellular mechanisms involved in eliciting responses from these nerves requires a detailed understanding of the contributions of both the paracellular and transcellular pathways. In this paper we focus on the contribution of these 2 pathways to the responses of salts containing sodium and various organic anions in the presence and absence of amiloride. Electrophysiological recordings from trigeminal nerves, chorda tympani nerves, and isolated lingual epithelia were combined with morphological studies investigating the location (and permeability) of tight junctions, the localization of amiloride-inhibitable channels, and Na-K-ATPase in taste and epithelial cells. Based on these measurements, we conclude that diffusion across tight junctions can modulate chorda tympani and trigeminal responses to sodiumcontaining salts and rationalize the enhancement of taste responses to saccharides by NaCl. © 1993 Wiley-Liss, Inc.

    Notes: We have studied the induction of an amiloride-sensitive sodium influx into C6 glioma, NIE, and NB2A neuroblastoma cell lines. In late log phase, cells grown continuously in the presence of 10% fetal calf serum showed Na+ influxes of approximately 25-30 nmol/mg protein min; 〈 5% of this flux was inhibited by amiloride. Removal of serum for 24 h caused a decrease in the total Na+ influx to 15-20 nmol/mg protein/min. Upon readdition of serum to the incubation medium, there was an increase in total Na+ influx, depending on the cell type, of 20-400% within 2 min. This increment in Na+ influx represented an increase in amiloride-sensitive Na+ transport with an apparent K′, of 0.4 mM. By adding serum back at various times after serum deprivation, it was determined that 4 h was required to observe a detectable increase in the amiloride-sensitive Na+ flux. Thus, serum removal results in the induction of the amiloride transport system which, however, remains latent until the reintroduction of serum to the medium. Addition of 5 μg/ml of cycloheximide blocked the increase in Na+ transport, indicating that de novo protein synthesis mediated this serum deprivation-induced increase in Na+ transport. Moreover, inhibition of de novo lipid synthesis by 0.1 mM fenfluramine also blocked the induction of this transport activity, suggesting that a coordinated synthesis of lipid and protein is required for the expression of this sodium transport site. We have also found that this serum stimulated Na+ influx did not saturate with Na+ concentration, up to 140 mM. Also, among commonly used inhibitors of passive Na+ entry into epithelial tissues, only amiloride was capable of inhibiting this transport system in these neural cell lines.

    Description: This paper deals with the development of multiprocessor simulations from a serial set of ordinary differential equations describing a physical system. The identification of computational parallelism within the model equations is discussed. A technique is presented for identifying this parallelism and for partitioning the equations for parallel solution on a multiprocessor. Next, an algorithm which packs the equations into a minimum number of processors is described. The results of applying the packing algorithm to a turboshaft engine model are presented.

    Description: The NASA Lewis Research Center is actively involved in the development of expert system technology to assist users in applying parallel processing to computational fluid and structural dynamic analysis. The goal of this effort is to eliminate the necessity for the physical scientist to become a computer scientist in order to effectively use the computer as a research tool. Programming and operating software utilities have previously been developed to solve systems of ordinary nonlinear differential equations on parallel scalar processors. Current efforts are aimed at extending these capabilities to systems of partial differential equations, that describe the complex behavior of fluids and structures within aerospace propulsion systems. This paper presents some important considerations in the redesign, in particular, the need for algorithms and software utilities that can automatically identify data flow patterns in the application program and partition and allocate calculations to the parallel processors. A library-oriented multiprocessing concept for integrating the hardware and software functions is described.

    Description: An approach to automate the programming and operation of tree-structured networks of multiprocessor systems is discussed. A conceptual, knowledge-based operating environment is presented, and requirements for two major technology elements are identified as follows: (1) An intelligent information translator is proposed for implementating information transfer between dissimilar hardware and software, thereby enabling independent and modular development of future systems and promoting a language-independence of codes and information; (2) A resident system activity manager, which recognizes the systems capabilities and monitors the status of all systems within the environment, is proposed for integrating dissimilar systems into effective parallel processing resources to optimally meet user needs. Finally, key computational capabilities which must be provided before the environment can be realized are identified.
