    Notes: Abstract The organophosphonates are biogenic and xenobiotic compounds characterized by the presence of a stable carbon to phosphorus (C-P) bond. The C-P bond imparts upon these molecules a relative resistance to (bio)degradation and fears have been expressed over their environmental recalcitrance and possible ecotoxicity, as more than 20×103 tonnes of these compounds enter the environment annually in the U.S.A. and western Europe alone (Egli, 1988). Biodegradation of organophosphonates is generally accepted to be dependent upon the phosphate status of the cell, with biodegradation occurring only under conditions of phosphate limitation. In recent years, however, several novel bacteria capable of completely mineralizing both natural and man-made organophosphonates have been isolated. These organisms represent a departure, both at a physiological and genetic level, from the accepted consensus that organophosphonates are utilized only phosphorus sources. This review covers all aspects of our knowledge of organophosphonate metabolism over the last 50 years, concentrating on the advances made in the last 10 years.

    Notes: We present an approach for determining in vitro the means and distributions of a set of phenomenological parameters, including cell speed and persistence time, which can be used to evaluate the effect of isotropic variations in the extracellular environment on the motility of human tissue cells. Using time-lapse videomicroscopy and semi-automated image analysis, we tracked the paths traveled by slow-moving, isolated human vascular smooth muscle cells over 48 hours on surfaces of petri dishes coated with 10 μg/mL of the adhesive extracellular matrix proteins type IV collagen, fibronectin or laminin. By applying a persistent random walk model to experimental data for mean-squared displacement as a function of time for these cells, we rigorously distinguished individual cells with different motile characteristics not obvious based on qualitative comparisons between the structures of individual cell paths. We also positively identified the presence of immotile cells. Based on the behavior of 34 to 77 cells on each substrate, we found mean cell speeds and persistence times on the order of 10 micron/h and 3 hours, respectively, on all three ECM substrates, while the fraction of motile cells varied from 65% on laminin to 78% on collagen. On all three surfaces experimental number distributions of speed and persistence time could be described by normal and exponential waiting time distributions, respectively. Our approach provides a framework for addressing questions concerning the mechanistic relationship between cellular and environmental properties and cell motility.

    Notes: The motility of a population of swimming bacteria can be characterized by a random motility coefficient, μ, the operational equivalent of a diffusion coefficient at the macroscopic level and in the absence of interacting chemical gradients. At the microscopic level, random motility is related to the single-cell parameters: speed, tumbling probability, and index of directional persistence (related to the angle a cell's path assumes following a change in direction). Various mathematical models have been proposed for relating the macroscopic random motility coefficient to these microscopic single-cell parameters. In separate experiments, we have measured motility at both the cell-population and single-cell levels for Escherichia coli. The agreement of these results shows that the macroscopic transport behavior of a population of motile bacteria can be predicted from straightforward microscopic observations on single cells.

    Notes: The facilitated transport of carbon monoxide through thin, immobilized films of cuprous chloride solutions is investigated, using a tracer C14O technique. Steady-state tracer flux measurements are used to determine the second-order forward rate constant and equilibrium constant for the reversible reaction of CO with cuprous ion. The enchanced results at low CO partial pressures can be interpreted in terms of a consecutive reaction model. The cuprous ion is remarkably effective as a carrier for CO. The CO permeation rate in the presence of cuprous ion can be increased by two orders of magnitude over the nonfacilitated flux. As such, the cuprous ion is an attractive facilitating agent for use in a membrane separation system.
