    Notes: Various multisurface mass-scale tissue culture propagators ranging in capacity up to 200 liters have been developed at Abbott Laboratories. These patented units consist, of an enclosed vessel containing a multiplicity of separated glass plates or disks on which cells may attach and proliferate. Means for mixing and aeration of the medium are provided. Sample ports facilitate the addition of cultures and media, the withdrawal of samples, the washing of cell monolayers, and the harvesting of cells and cell products. The large cell growth surface per occupied volume, the provision for separating tissue cells from media simply and easily, and the minimization of the amount of labor required per cell growth area are some of the many advantages of the multisurface tissue propagator that are described.

    Notes: A process is described in which the baboon endogenous virus (BaEV) is produced under optimum conditions in cell culture, and concentrated by hollow-fiber ultrafiltration technology under conditions of large-scale production. This system has advantages over conventional systems in that the flow rate is increased 2.5-fold during concentration. Thermal inactivation of BaEV was retarded by the addition of lactose glutamate to the harvested tissue culture fluid. After concentration, at least 91% of the virus RNA-directed DNA polymerase is recovered with a concomitant increase in infectious virus. Materials needed for modifying described systems may be obtained from commercial sources.

    Notes: A new concept of tissue culture equipment and procedures was developed for the mass-scale growth of several types of animal tissue cells in monolayers on multiple glass surfaces. Continuous, cell lines, primary and diploid cell strains were grown in this equipment. Cells studied include primary bovine kidney, human diploid WI-38, human foreskin, and mouse CCL1 cells. Photomicrographic comparisons of cells grown by these techniques indicate they are morphologically identical to tissue culture cells grown in glass bottles or tubes. The growth of the tissue culture cells in the propagator was monitored by carbohydrate Utilization and acid production. Large-scale production of viruses and biochemicals on cells grown in the multiple-plate tissue culture propagator was accomplished. Virus titers were equal to those obtained from conventional bottle or tube cultures for several strains of influenza, parainfluenza, and respiratory syneytial viruses. High-titred mouse interferon was also produced in this system. In addition to tissue culture cell production, Eaton agent, Mycoplasma pneumoniae was grown on the multiple glass surfaces on a mass scale.

    Notes: Various biomass (X), product (P), and substrate (S) rate equations are considered in order to synthesize a general xanthan fermentation model from literature data. Analytical forms that provide reasonable descriptions for the X, P, and S behaviors reported by Moraine and Rogovin are shown to be the logistic equation, the Luedeking-Piret equation a modified Luedeking-Piret equation, respectively. The autonomous logistic equation allows the serial evaluation of parameters for all thee equations, rather than a simultaneous determination required by nonautonomous models.

    Notes: A simple and reproducible quantitative method for measuring dissolved oxygen (DO) in uninfected and baculovirus infected cells in culture is described. To establish this method, an industrial DO measuring system for fermentation was employed. During this process the physical characteristics of the cell culture vessel were taken into account permitting a direct readout of DO in microliters per vessel. During these studies, it was experimentally documented that insect cells, particularly baculovirus infected cells, in culture from 1 to 14 days utilize an appreciable level of DO.

    Notes: In order to identify and characterize the wheat grain allergens involved in bakers' asthma, proteins were sequentially extracted from whole-meal flour. The polypeptide composition of the individual solubility fractions (albumin/globulin, gliadin and glutenin) was analyzed by sodium dodecyl sulfate-gel electrophoresis (SDS-PAGE), and high-resolution two-dimensional gel electrophoresis with immobilized pH gradient 4-9 in the first dimension (IPG-Dalt). The resolved polypeptides were transferred onto an immobilizing polyvinylidene difluoride membrane and incubated with a pooled serum from four asthmatic bakers. Bound IgE was demonstrated by autoradiography using 125I-labeled anti-human IgE. Our study demonstrated that the serum of the bakers allergic to flour contained IgE antibodies which bound to numerous polypeptides of all three solubility fractions. The highest percentage of IgE binding was observed with certain albumin and/or globulin polypeptides, whereas the gliadins and glutenins exhibited considerably less allergenicity. SDS-PAGE revealed that the protein which bound the highest percentage of IgE from the sera of the allergic bakers is a 27 kDa albumin. More detailed investigations using IPG-Dalt demonstrated that this allergen is not a single polypeptide but consists of several polypeptide spots that differ in their isoelectric points. Quantitative studies using computer-assisted laser densitometry revealed that the amount of patients' IgE bound by these particular polypeptides differed considerably between the seven wheat cultivars examined, ranging from 13% to 53% of the total radioactive uptake.

    Notes: A low fluorescence background polypropylene (PP) membrane has been developed for ultimate use as an electroblotting membrane in DNA sequencing based on fluorescence detection. The DNA binding capacity of this membrane is improved by a surface modification using radio frequency plasma discharge (RFPD) in ammonia gas. The RFPD operational parameters are evaluated both in terms of membrane nitrogen content and in terms of the product's capacity for binding radioisotope-labeled DNA fragments. The surface morphologies of the derivatized membranes are examined by scanning electron microscopy; their mechanical and electrical properties, which are important for the subsequent sequencing procedures, are likewise established. Due to the goal of developing a membrane suitable for multiplex processing, in which the electroblotted DNA must withstand dozens of hybridization/stripping cycles, special attention is given the covalent attachment of DNA to the membrane. The modified PP membrane is evaluated in a multiplex sequencing application using radioisotope-labeled DNA probes, and found to yield somewhat better binding of a given amount of electroblotted DNA than the commonly used GeneScreen membrane. A tenfold repetition of the probing indicates little loss of signal; the membrane-bound DNA is stable upon storage and shows no detectable loss in probing efficiency after one month.

    Notes: Measurements of the shape of electrophoretic bands of phycoerythrin and conalbumin have been made at regular intervals during migration in agarose gels. Analysis of the peak shapes suggests the existence of a significant degree of asymmetry. This is to be contrasted with the symmetry around the peak associated with the generally assumed Gaussian band. The degree of asymmetry of the bands decreased as a function of time and increased with agarose concentration. A similar experiment on DNA indicated constancy of the degree of asymmetry as a function of time. These results can be interpreted as, but do not prove the validity of, a nonlocal diffusion equation which generalizes a theory originally put forth by Giddings and Eyring (J. Am. Chem. Soc. 1955, 59, 416-420). The results may be significant in framing a measure of the resolvability of electrophoretic peaks.
