    Notizen: Chemiluminescence (CL) detection based on the horseradish peroxidase (HRP) catalyzed reaction of luminol with peroxide was investigated as a post-separation detection scheme for microchip-based capillary electrophoresis. An integrated injector, separator and post-separation reactor was fabricated on planar glass wafers. The fluorescein conjugate of HRP (HRP-F1) was used as a sample for optimization of the CL detector response. In devices etched 10 μm deep, with an aluminum mirror integrated onto the backside of the detection zone to enhance collection efficiency, the detection limit, estimated at 3 standard deviations (SD) above background noise, for 1 nL injected sample plugs was 35 nM in HRP-F1. In devices etched 40 μm deep, 8 nL plugs gave a detection limit of 7 nM. Separation and CL detection of the products of an immunological reaction of a F(ab')2 fragment of the HRP conjugate of goat anti-mouse immunoglobulin G (IgG) with mouse IgG was performed on-chip. A linear calibration curve was obtained for the decrease in peak height of the HRP conjugate (53 μg/mL) with increasing mouse IgG (0-60 μg/mL). When microperoxidase was used as an internal standard, the R2 value of a linear least-squares fit was 0.9867, and the relative errors in the slope and intercept were ± 5.8 and ± 1.3%, respectively.

    Notizen: The affinity constant of a monoclonal antibody to fluorescently labeled bovine serum albumin (BSA*) was measured in diluted mouse ascites fluid using a microfluidic chip to perform affinity capillary electrophoresis. Borofloat glass-based devices could be used repeatedly with samples for many months. On-chip separations were performed in less than 60 s, and 30-60 s was required for manual sample exchange. The change in peak height for BSA* with increasing BSA*/anti-BSA concentration ratio was used to determine concentration changes in bound and free BSA*. A Scatchard plot analysis gave an affinity constant (more exactly the intrinsic association constant) of 3.5 ± 0.6 × 107 M-1 for a 1:1 stoichiometric ratio. Two affinity complexes were separated. One complex was identified by the Scatchard method as having a 1:1 stoichiometric ratio. The other complex is proposed to have a stoichiometry with an excess of anti-BSA to BSA*, most likely (anti-BSA)2-BSA*, on the basis of a faster migration time than the 1:1 complex, a decrease in the amount of this complex with increasing [BSA*], and predictions of theoretical models for multi-valent antigens. Potential applications of microchip-based devices in affinity measurements are discussed.

    Notizen: Clinical interest in the use of capillary electrophoresis (CE) has recently been extended to the microchip environment. Clinical analyses demand careful handling of complex samples that are often limited in quantity and in concentration. The integrated sample handling and analysis capabilities of microchip substrates thus seem ideally suited to clinical applications. This review surveys the development of sample handling (injection, mixing, and reaction) and separation elements on-chip. The integration of these elements to create a variety of clinical analyzers has been demonstrated. The application of microchip CE systems to human serum protein analysis, immunoassay, and DNA studies is reviewed, along with various other clinical applications. In addition, the clinical potential of the lab-on-a-chip concept is discussed.

    Beschreibung: Dogs were used to study the effects of varying the differentiator cutoff frequency on the recorded peak first derivative of left ventricular pressure with respect to time (dP/dt), using high-precision solid-state pressure transducers and recording equipment. In canine hearts with a basic periodicity of 1 to 3 Hz, the differentiator frequency response required to record an accurate peak dP/dt is found to be influenced by the value of peak dP/dt. At peak dP/dt ranging from 1500 to 9000 mm Hg/sec (200 and 1200 kPa/sec), a differentiator cutoff frequency of at least 90 Hz was required to record accurately peak dP/dt.

    Beschreibung: A transducer and frequency discriminator circuit is described including a four-terminal circulating diode bridge, a first pair of capacitors connected in series across two terminals of the bridge, and a second pair of capacitors, or other impedance elements, connected in series across the other two terminals of the bridge. A source of balanced alternating electrical energy for energizing the circuit is coupled between the commonly connected plates of the first pair of capacitors and the commonly connected plates of the second pair of capacitors. Due to the operation of the diode bridge, the sum of the resultant charges developed on the first pair of capacitors is proportional to the relationship between the respective capacitors of the second pair, and consequently, an output voltage taken across the first pair of capacitors will be proportional to that relationship.

Considerations for acquiring new medical technology are discussed in some detail. Acquisition of new technology should depend on whether the equipment provides more and relevant clinical data, is for research or for limited use, is properly engineered for patient use, presents information in easily storable and retrievable form, is tested and validated clinically, is fabricated by a reliable manufacturer, is cost effective, and may be readily replaced by a new technology.
