    Notes: We report the first direct demonstration that the cytoplasmic transport of organelles and vesicles (collectively called particles) takes place along microtubules. Living keratocytes from the corneal stroma of the frog, Rana pipiens, were observed with Allen video-enhanced constrast, differential interference constrast (AVEC-DIC) microscopy [Allen et al, 1981]. In sufficiently thin regions of these cells a network of linear elements was visible. When particles were observed in motion, they always moved along these linear elements. The linear elements remained intact and in focus on the microscope when lysed in a cell lysis solution that stabilized microtubules. Preparations were then fixed in formaldehyde, washed with phosphate-buffered saline (PBS), incubated with rabbit antitubulin, washed with PBS, stained with rhodamine-conjugated goat antirabbit, and washed with PBS. The extracted cells continued to remain in place and in focus on the microscope throughout these procedures. The same cells were then observed using epifluorescence optics and a silicon-intensified target (SIT) video camera. A network of fluorescent linear elements was seen to correspond in number, form, and position to the linear elements seen in the live AVEC-DIC image. Taken together, the AVEC-DIC and fluorescence microscopy observations prove that the linear elements along which particles move are microtubules (MTLEs). The observed particle speeds, pause times, and distances moved varied widely, even for the same particle on the same microtubule. Particles were also observed to switch from one microtubule to another as they were transported. The polarity of the microtubules did not seem to affect the particle direction, since particles were observed to move in both directions on the same MTLE. When not in motion these particles behaved as if anchored to the microtubules since they showed negligible Brownian motion. Finally, it was observed that an elongate particle could move onto two intersecting linear elements such that it was deformed into an inverted “Y” shape. This indicates that there may be more than a single site of attachment between the force generator and the particle.

    Notes: An optimized potential function for base-stacking interaction is constructed. Stacking energies between the complementary pairs of a dimer are calculated as a function of the rotational angle and separation distance. Using several different sets of atomic charges, the electrostatic component in the monopole-monopole approximation (MMA) is compared to the more refined segmented multipole-multipole representation (SMMA); the general features of the stacking minima are found to be correctly reproduced with IEHT or CNDO atomic charges. The electrostatic component is observed to control the location of stacking minima. The MMA, in general, is not a reliable approximation of the SMMA in regions away from minima; however, the MMA is reliable in predicting the location and nature of stacking minima. The attractive part of the Lennard-Jones 6-12 potential is compared to and parameterized against the expression for the second-order interaction terms composed of multipole-bond polarizability for the polarization energy and transition-dipole bond polarizabilities for approximation of the dispersion energy. The repulsive part of the Lennard-Jones potential is compared to a Kitaygorodski-type repulsive function; changing the exponent from its usual value of 12 to 11.7 gives significantly better agreement with the more refined repulsive function. Stacking minima calculated with the optimized potential method are compared with various perturbation-type treatments. The optimized potential method yields results that compare as well with melting data as do any of the more recent and expensive perturbation methods.

    Notes: Absolute rate constants for the addition of the highly nucleophilic 2-hydroxy-2-propyl radical to eight fast-reacting 1- and 1,1-disubstituted alkenes in MeOH at room temperature have been determined by laser flash photolysis. Also the absorption spectra of the 2-hydroxy-2-propyl and the benzylic and alkyl-type adduct radicals are presented. The rate constants were obtained using various methods for the analysis of the kinetic traces and support earlier findings.

Die Polymerisation weist während der Bestrahlung im allgemeinen eine S-förmige Umsatzkurve auf, mit hohen Geschwindigkeiten und quantitativem Umsatz bei Temperaturen von 10°C unterhalb des Schmelzpunktes. Die Geschwindigkeiten waren nach der Bestrahlung niedriger und zeigten eine typische Annäherung an einen Grenzumsatz. Die Polymerisationsgeschwindigkeiten nahmen mit der Temperatur und der Bestrahlungsdosis sowie mit der Länge der Alkylkette zu. N-Methylmaleimid zeigte eine hohe Geschwindigkeit bei der Polymerisation in Substanz, die der Flüchtigkeit des Monomers zugeschrieben wurde.
    Notes: N-alkylmaleimides from the N-methyl to the N-octadecyl derivative were polymerized in the solid state by γ-radiation. Polymerization during irradiation generally showed sigmoidal conversion curves with high rates and complete conversion 10°C below the melting point. Post-irradiation rates were lower and showed the typical approach to a “limiting” conversion. The polymerization rates increased with temperature and radiation dose and with the length of the alkyl chain. N-methylmaleimide had a high rate of in-source polymerization which was attributed to the volatility of the monomer.

    Notes: Whereas the radiation-induced polymerization of crystalline barium methacrylate monohydrate has been well established there have been conflicting reports concerning barium methacrylate anhydrate. We have now shown that the yields of high molecular weight polymer are negligible if adequate precautions are taken to prevent polymerization occurring during the separation of polymer from residual monomer. However, monomer losses of up to 80% have been observed by titration of C=C bonds with bromine and confirmed by decreases in the appropriate infrared absorptions. X-ray diffraction studies showed that the crystalline lattice was largely destroyed during the reaction. ESR spectra indicate that initial trapped radicals are converted into propagating radicals with a different conformation than has been reported for the monohydrate.

    Notes: The combined techniques of light microscopy, scanning (SEM) and transmission (TEM) electron microscopy were used for the first time to study the structure of unicameral lungs of a Tegu lizard (Tupinambis nigropunctatus). The lungs are prolate spheroid bags with blood supplied by superficial branches of a dorsal pulmonary artery and returned by diffuse, more deeply located veins. The primary bronchus enters the medial aspect near the apex of the lung. The lung wall is composed of trabeculae: (1) arranged in a faviform pattern, (2) forming individual faveoli (gas exchange chambers) which appear deepest in the cranial one-half of the lung, (3) all of which have a smooth muscle core overlain by either a ciliated or nonciliated epithelium. A ciliated epithelium lines the luminal surfaces of the large primary trabeculae and parts of smaller secondary trabeculae; it is composed of cone-shaped cells with ciliated-microvillous surfaces, and of columnar serous secreting cells. Nonciliated epithelium covers the luminal surface of portions of some secondary trabeculae, abluminal surfaces of primary and secondary trabeculae and all surfaces of the small tertiary trabeculae forming the faveoli. The nonciliated epithelium overlies an extensive superficial capillary network. The blood-gas barrier (0.7-1.0 μm thick) is composed of a thin cytoplasmic flange of Type I pneumonocytes, a thick homogeneous basal lamina and an attenuated endothelial cytoplasm. Numerous surfactant-producing Type II pneumonocytes are closely associated with the Type I pneumonocytes. The nonrespiratory ciliated epithelium may function in humidification of air and clearing of the lungs.

    Notes: Ultrastructural observations and glyoxilic acid-induced fluorescence of catecholamines indicate that tracts of axons lie at the base of the ciliary bands and run throughout their length in bipinnaria and brachiolaria larvae of Pisaster ochraceus. Two types of nerve cells occur at regular intervals within the ciliary bands. Type I nerve cells are associated with the axonal tracts, and type II nerve cells, which are ciliated, occur along the edge of the ciliary bands. Two prominent ganglia, which appear as accumulations of nerve cells and neuropile, occur on the lower lip of the larval mouth. Smaller ganglia occur irregularly throughout the ciliary band. Synapses were never clearly identified and were assumed to be unspecialized. Nervous tissues were also found associated with the esophageal muscles, the attachment organ, and the larval arms. Organization of the nervous system and its association with effectors suggest it controls swimming and feeding. Several similarities exist between the nervous systems of larval asteroids, larval echinoids, and adult echinoderms.

    Notes: The supporting elements of the avian propatagium were examined in intact birds and as isolated components, using static force-length measurements, calculated models, and airflow observations. The propatagial surface supported between Lig. propatagiale (LP) and brachium-antebrachium is equally resistant to distortion over the range of wing extension used in flight. The lengths LP assumes in flight occur across a nearly linear, low-stiffness portion of the force-length curve of its extensible pars elastica. In an artificial airflow, intact wings automatically extend; their degree of extension is roughly correlated with the airflow velocity. Comparisons between geometric models of the wing and the passive force-length properties of LPs suggest that the stress along LP blances the drag forces acting to extend the elbow. The mechanical properties (stiffness) of the LP vary and appear to be tuned for flight-type characteristics, e.g., changes in wing extension during flight and drag. Lig. limitants cubiti and LP combine to limit elbow extension at its maximum, a safety device in flight preventing hyperextension of the elbow and reduction of the propatagium's cambered flight surface. Calculations using muscle and ligament lengths suggest that M. deltoideus, pars propatagialis, via its insertions onto both the propatagial ligaments, controls and coordinates propatagial deployment, leading edge tenseness, and elbow/wing extension across the range of wing extensions used in flight. The propatagial ligaments and M. deltoideus, pars propatagialis, along with skeleto-ligamentous elbow/carpus apparatus, are integral components of the wing's extension control mechanism. © 1995 Wiley-Liss, Inc.
