    Description: When the Mg2+ ion in the catalytic center of Escherichia coli RNA polymerase (RNAP) is replaced with Fe2+, hydroxyl radicals are generated. In the promoter complex, such radicals cleave template DNA near the transcription start site, whereas the beta' subunit is cleaved at a conserved motif NADFDGD (Asn-Ala-Asp-Phe-Asp-Gly-Asp). Substitution of the three aspartate residues with alanine creates a dominant lethal mutation. The mutant RNAP is catalytically inactive but can bind promoters and form an open complex. The mutant fails to support Fe2+-induced cleavage of DNA or protein.     Description / Table of Contents: Zusammenfassung Es werden vergleichende Bestimmungen des Kaliumgehaltes in Feldspäten mit den Methoden Flammen-Atomemissionsspektrometrie mit und ohne Leitelement, Flammen-Atomabsorptionsspektrometrie und massenspektrometrische Isotopenverdünnungsanalyse bei Probeneinwaagen von etwa 50 mg durchgeführt. Die massenspektrometrische Meßtechnik des Kaliums bei thermischer Oberflächenionisation wird beschrieben. Der Kaliumgehalt zweier Kalifeldspäte wird bei Anwendung der verschiedenen Verfahren mit relativen Standardabweichungen von 0,3–1,8% bestimmt.
    Notes: Summary The potassium content in feldspars is determined by the methods of flame atomic emission spectrometry with and without internal standard, flame atomic absorption spectrometry and mass spectrometric isotope dilution analysis with sample weights of about 50 mg. The mass spectrometric measurements of potassium by the thermal ionization technique is described. The potassium content of two potassium feldspars is determined by the different methods with relative standard deviations of 0.3–1.8%.
