    Notes: A model for the hydration behavior of human stratum corneum has been developed from measurements on in vitro samples isolated in a manner which minimized tissue treatment and trauma. Water sorption/desorption rate measurements as a function of water activity, temperature, and tissue integrity are reported. These data, together with thermodynamic data (infrared and nmr results reported earlier) are consistent with a model in which rapidly sorbed/desorbed water (ca. 0.5 mg water/mg stratum corneum) is associated with (“bound” by) the tissue, while slowly sorbed/desorbed “free” water (up to 12 mg water/mg stratum corneum) is kinetically restricted and probably intracellular in location. Both equilibrium water binding and desorption kinetic data suggest structural changes of this cellular water barrier upon hydration. Evidence for hysteresis in water sorption isotherms (reported by others) could not be reproduced.

    Notes: Culturing of Aspergillus flavus was conducted in static flask cultures and 4 in. × 5 ft columns (containing 7-8 kg corn) to measure the effects of moisture, temperature, and air flow upon growth and the production of aflatoxin. Aflatoxin levels as high as 6200 ppb (dry basis) in 10 days were observed. Conditions were selected (ca. 20% moisture, 0.008 liter air/kg corn/min air flow with 1.5 liter/kg/min recirculated) for production of aflatoxin in 1200 bushels of corn in a 18-ft diam corrugated steel Butler storage bin for preparation of contaminated corn for animal feeding trials and for testing of an ammoniation process for decontamination of aflatoxin in corn. A target level of 1000 ppb aflatoxin was attained.

    Notes: Bacillus subtilis NRRL B-2612, isolated at this laboratory, was used to produce polyglutamic acid from devitalized wheat gluten. Conditions for production of the polypeptide were investigated. When NRRL B-2612 was grown on wheat gluten in shaken flasks with phsophate buffer and salts at pH 6.5, yields of polymer reflected a 25-40% incorporation of glutamic acid into polyglutamic acid. Repeated alcoholic precipitation yielded peptides containing between 45 and 90% D-glutamic acid. Physical and chemical properties of a typical purified polyglutamic acid indicated that the product was a gamma-linked polypeptide composed of 75% D-glutamic acid and 25% L-glutamic acid with a molecular weight of approximately 200,000.

    Notes: The improvement of the biocatalytic reduction of 2-allyl-carboethoxy-cyclopentanone (2) to the corresponding cyclopentanol derivative (+)-(1R,2R)-(1) was accomplished employing baker's yeast in organic media. This chiral cyclopentanol derivative (1), analyzed by high resolution gas chromatography performed over β-cyclodextrin stationary phase, was obtained in 38% yield (〉99% e.e.).     Notes: An investigation into the shifts induced in the NMR spectra of some derivatives of substituted phenols by tris(dipivalomethanato)europium and tris(dipivalomethanato)praseodymium is reported. The phenol derivatives were studied to ascertain the possible utility of the shift reagents as an aid in the analysis of phenolformaldehyde resins by NMR spectroscopy, and to gain information on induced shifts with molecules having more than one possible co-ordination site.

    Notes: Adiabatic reactive rheometry involves the simultaneous measurement of viscosity and temperature changes during adiabatic polymerization. Using the adiabatic reactor method to relate temperature to fractional conversion results in a useful rheokinetic tool ideally suited for fastreacting reaction injection molding (RIM) systems for which the mold-filling step is nearly adiabatic. In this work, a small laboratory RIM machine is used to mix the reactants and deliver them to a constant stress rheometer retrofitted with a wide-gap Couette geometry and two thermocouples. Measurements on two polyurethane systems are reported. A simple cross-linking system is used to verify the adiabatic rheokinetic method through comparison to a known gel conversion. Subsequent measurements on a phase-separating RIM system show that increases in catalyst level, hard segment content, and initial reactant temperature result in a decreased gel time and an increased gel conversion. The viscosity rise profiles aid our understanding of the onset and development of phase separation. They are also essential for mold-filling models and establishing moldability criteria for these RIM systems.
