    Notes: Capillary column isobutane chemical ionization was found to be an excellent method for the mass spectral characterization of mustard, other sulfur vesicants and related compounds. Interpretation of [M + H]+ and fragmentation ion information afforded by this technique enabled the identification of many previously unreported mustard impurities. The developed methodology was applied to the analysis of an Iran/Iraq soil sample suspected to have been contaminated with mustard. Mustard and 17 other mustard related impurities were identified and characterized in this sample under electron impact and isobutane chemical ionization conditions.

    Notes: HP 5890 Series II Gas Chromatographs are equipped with prototype hardware enabling electronic pressure programming of all detector and carrier gases. A method of using electronic pressure control (EPC) to optimize detector performance (sensitivity, selectivity, and baseline stability) is demonstrated. With EPC, sample introduction, column separation, and detector performance can be optimized simultaneously without the classical trade-offs in performance. An example is also presented of the use of a new automated system which enables cool on-column injection into 250 or 320 μm columns without the need for a retention gap.

    Notes: A method that combines gas chromatography and mass spectrometry (GC/MS) for the determination of N-0923 in human and monkey plasma is detailed. This novel drug holds promise for the treatment of Parkinson's disease. Clinical use of N-0923 will result both in low peak plasma concentrations and in a short half-life; consequently, a highly sensitive method will be required. Previous methods using high-performance liquid chromatography with electrochemical detection have given reliable data in high dose studies in animals to 0.3-1.0 ng/mL in plasma but are not considered adequate for studies at clinical levels in humans. The data in this paper will show that GC/MS can offer a new direction in the determination of N-0923 in human and monkey samples, with reliable data to 25 pg/mL in plasma. This method is presented as a novel approach to the determination of N-0923, and as a starting point for further development.

    Notes: The feasibility of using capillary electrophoresis/mass Spectrometry, with negative-ion electrospray ionization, for applications of relevance to the dyeing and textile industries has been shown. Structurally similar examples of solubilized vat dyes of different halogen content have been separated and identified with a view to investigating the photodegradation processes which occur when the dyes are subjected to UV light. Separations of the various species formed when a mordant dye is chrome treated are also presented, indicating the ability to study complex formation arising from the different chrome dyeing methods.

    Notes: We report the electrospray (ESI) mass spectra of four fluoroquinolones, enoxacin, norfloxacin, ofloxacin and ciprofloxacin, a group of highly potent antibiotics that have not been previously characterized by this ionization technique. At low sampling cone voltage settings, 50 and 100 V, the electrospray mass spectra contained primarily molecular adduct ions. At a sampling cone voltage setting of 150 V, collisionally activated dissociation mass spectra containing structurally significant product ions were produced and accurately mass measured at a magnetic sector resolution of 3000. Gradient separation and characterization of these compounds was demonstrated by analysis using packed capillary column liquid chromatography with UV and ESI-MS detection. UV detection limits in the 1 to 2 ng range ((signal-to-noise ratio S/N) = 10:1) were estimated and interpretable mass spectra with S/N ratios in excess of 100:1 were observed for 1.5 ng of analyte during electrospray mass spectrometric analysis.

    Notes: Capillary column ammonia chemical ionization mass spectrometry was investigated as a technique for the characterization of a number of organophosphorus chemical warfare agents and simulants, since electron impact ionization provides little or no molecular ion information for these compounds. The effects of source temperature, source pressure, electron energy and electron emission were investigated in order to optimize ammonia chemical ionization conditions. Source temperature was found to have a dramatic effect on the chemical ionization mass spectra acquired. Abundant [M + NH4]+ and/or [M + H]+ quasimolecular ions were usually observed at lower source temperatures for the selected compounds under positive ion chemical ionization conditions, along with a number of characteristic fragmentation ions. Full-scan mass spectra were obtained for 1--2 ng of analyte during the capillary column ammonia chemical ionization operation. The sensitivity, molecular ion information afforded and lack of background interference ions associated with this technique make it a good candidate for the confirmation of organophosphorus chemical warfare agents and simulants.

    Notes: The strength of adhesion and dynamics of detachment of murine 3T3 fibroblasts from self-assembled monolayers were measured in a radial-flow chamber (RFC) by applying models for fluid mechanics, adhesion strength probability distributions, and detachment kinetics. Four models for predicting fluid mechanics in a RFC were compared to evaluate the accuracy of each model and the significance of inlet effects. Analysis of these models indicated an outer region at large radial positions consistent with creeping flow, an intermediate region influenced by inertial dampening, and an inner region dominated by entrance effects from the axially-oriented inlet. In accompanying experiments patterns of the fraction of cells resisting detachment were constructed for individual surfaces as a function of the applied shear stress and evaluated by comparison with integrals of both a normal and a log-normal distribution function. The two functions were equally appropriate, yielding similar estimates of the mean strength of adhesion. Further, varying the Reynolds number in the inlet, Red, between 630 and 1480 (corresponding to volumetric flow rates between 0.9 and 2.1 mL/s) did not affect the mean strength of adhesion. For these same experiments, analysis of the dynamics of detachment revealed three temporal phases: 1) rapid detachment of cells at the onset of flow, consistent with a first-order homogeneous kinetic model; 2) time-dependent rate of detachment during the first 30 sec. of exposure to hydrodynamic shear, consistent with the first-order heterogeneous kinetic model proposed by Dickinson and Cooper (1995); and 3) negligible detachment, indicative of pseudo-steady state after 60 sec. of flow. Our results provide rigorous guidelines for the measurement of adhesive interactions between mammalian cells and prospective biomaterial surfaces using a RFC. © 1997 John Wiley & Sons, Inc.     Notes: The conversion of the enzymatic hydrolysate of shellfish chitin waste to single-cell protein was investigated as part of a comprehensive waste treatment program. Forty-two yeasts were screened for ability to assimilate the monomer of chitin, N-acetylglucosamine, which has been shown to be the sole product of enzymatic hydrolysis of chitin. The Yeast Pichia Kudriavzevii was selected for study, based on ability to grow at high temperature (37°C and above), low pH (4.0 ± 0.5), and in a nutritionally simple medium. Growth rates of P. kudriavzevii were similar on N-acetylglucosamine and on the chitin hydrolysate. Dependencies of specific growth rate on temperature, pH, medium composition, and oxygen tension were studied. The variations of yield, protein content, and total nucleic acid content with the specific growth rate were evaluated. The amino acid distribution of the protein of P. kudriavzevii was obtained.

    Notes: As part of the development of a comprehensive mathematical model for chitinase production by Serratia marcescens QMB 1466 growing on chitin, the different mass transport and kinetic steps involved during chitin hydrolysis were studied. The experimental results for the hydrolysis of chitin by a crude preparation of chitinase show a system kinetically limited by the overall rate of chitin hydrolysis. This rate is linearly related to the concentration of enzyme adsorbed on the chitin particle. Adsorbed and bulk enzyme concentration were found to be related through a Langmuir type of isotherm.
