    Notes: Thermotropic polyamides with high molecular weights were synthesized by melt polycondensation of 3,3′-disubstituted-4,4′-biphenylenediacetamides with α,ο-diphenoxyalkane-4,4′-dicarboxylic acids. Methyl, methoxy, and chloro groups were used as 3,3′-substituents. IR measurements revealed that there are hydrogen-bonded carbonyls and free carbonyls the intensities of which depended on the polymer structure and the temperature. The thermotropic liquid crystallinity of the polyamides is assumed to occur by a decrease in intermolecular hydrogen bondings between carbonyls and amide NH's which was caused both with 3,3′-substitutions of the biphenylene moiety and with introduction of long alkylene spacers in the polymer backbone. In addition, mechanical properties of the thermotropic polyamides were measured on the molded dumbbell-type specimen. The 3,3′-dichloro polyamides showed medium tensile strengths and moduli in the range of 500-890 kgf/cm2a nd 19.0 × 103 to 27.0 × 103 kgf/cm2, respectively. © 1996 John Wiley & Sons, Inc.

    Notes: The strong interaction of hexa-histidine fusion proteins with metal chelate adsorbents was utilized to immobilize β-galactosidase with a hexa-histidine peptide at the N-terminus to the Ni2+-nitrilotriacetic acid adsorbent. The fusion protein was cloned and expressed in Escherichia coli. The purified soluble fusion protein showed the same specific activity as the purified β-galactosidase and retained 64 percent of its β-galactosidase activity when bound to the adsorbent. To demonstrate the potential of the immobilized β-galactosidase in organic chemistry, allyl-β-D-galactosidase was synthesized from lactose and allyl alcohol on a gram scale. The same enzyme preparation was reused in three subsequent batches to prepare the model compound with high yield. © 1993 John Wiley & Sons, Inc.

    Notes: Biological phenol degradation in a draft tube gas-liquid-solid fluidized bed (DTFB) bioreactor containing a mixed culture immobilized on spherical activated carbon particles was investigated. The characteristics of biofilms including the biofilm dry density and thickness, the volumetric oxygen mass transfer coefficient, and the phenol removal rates under different operating conditions in the DTFB were evaluated. A phenol degradation rate as high as 18 kg/m3-day with an effluent phenol concentration less than 1 g/m3 was achieved, signifying the high treatment efficiency of using a DTFB.

    Notes: Fed-batch culture of Bacillus thuringiensis in a modified airlift reactor has been developed by using adaptive control of glucose concentration in the reactor. The glucose concentration was estimated via a correlation equation between carbon dioxide production rate and glucose consumption rate. The estimated glucose concentration as the output variable was fed back to computer for calculation of substrate addition. The modified reactor was an airlift reactor with a net draft tube. The airlift reactor had high oxygen transfer rate and low shear stress which were important factors for production of thuringiensin. Fed-batch culture of Bacillus thuringiensis in the modified airlift reactor provided significant improvement of thuringiensin production. © 1995 John Wiley & Sons, Inc.

    Notes: A method of on-line optimal control for fed-batch culture of bakers yeast production is proposed. The feed rate is taken as the control variable. The specific growth rate of the yeast is the output variable and is determined from the balance equation of oxygen. A moving model is obtained by using the data from the feed rate and the specific growth rate. Based on the moving model, an optimal feed rate for fed-batch culture is then achieved.

    Notes: We have performed controlled fed-batch fermentation experiments to compare the production level of hepatitis B surface antigen (HBsAg) by recombinant yeast Saccharomyces cerevisiae strains (YNN27/pYBH-1, YNN27/ p2μ-S11, YNN27/pDCB-S2) containing plasmid vector with alcohol dehydrogenase (ADH1), acid phosphatase (PHO5), and glyceraldehyde-3-phosphate dehydrogenase (GPD) promoter, respectively. Yeast cell concentrations of 15-35 g dry cell weight/L were obtained. By limiting phosphorous concentration, HBsAg expression level for the YNN27/p2μ-S11 strain with inducible PHO5 promoter reached 0.2-0.3 mg/L. By controlling nutrient addition rate and dissolved oxygen concentration, HBsAg concentrations of 3-10 mg/L were achieved in 60-70 h fermentation using the YNN27/pDCB-S2 strain with the constitutive GPD promoter.

    Notes: A kinetic model was developed to describe the rate of reaction between chromium(III) and polyacrylamide (PAAm) that leads to the crosslinking or gelation of the polymer. Cr(III) was added directly to solutions of polyacrylamide which had a degree of hydrolysis of approximately 4%. The unreacted Cr(III) was separated from that which had reacted with the polymer by equilibrium dialysis and measured by atomic absorption.The rate of the reaction was described by an empirical model based on the assumption of invariant polymer group concentration during the reaction. The order of reaction was determined to be 1.32 for Cr(III), 0.8 for the polymer group, and -1.0 for hydrogen ion. Modifications were made to the model to allow for variation during reaction of the concentration of the polyacrylamide reactive group. The modified model accurately described the reaction rate for Cr(III) concentration ranging from 0.19 to 1.2 mM, polyacrylamide concentrations from 6.9 to 69 mM, and pH values from 4.0 to 5.5.

    Notes: The interface of a rubber-toughened epoxy resin was modified by using epoxide end-capped carboxyl-terminated butadiene and acrylonitrile random copolymer (CTBN). The end-capping epoxides were formulated with different ratios of flexible diglycidyl ether of propylene glycol (DER732) and rigid diglycidyl ether of bisphenol-A (Epon 828). The microstructure and the fracture behavior of these rubber-modified epoxy resins were studied by transmission electron microscopy (TEM) and scanning electron microscopy (SEM), respectively. The thermal and mechanical properties were also investigated. With an increase in the amount of end-capping DER732, the interfacial zone of an undeformed rubber particle and the degree of cavitation of the rubber cavity on the fracture surface were greatly increased. At the maximal addition of DER732, fracture energy (GIc) for this toughened epoxy resin containing 10phr CTBN rubber increases up to 2.4 fold compared to that of a conventional CTBN-toughened epoxy resin, but the thermal and the mechanical properties remained quite unaffected. The modification on the interfacial property provides a new technique in the improvement of fracture toughness of a rubber-toughened epoxy resin.

    Notes: Liquid hydroxyl-terminated cis-polybutadiene (cis-HTPB) having various molecular weights were cured with trimethylol propane (TMP) and isophorone diisocyanate (IPDI) by the one-shot method and the physical properties of these elastomers obtained were measured. The effect of pseudo-crosslinks was not observable and the calculated value of the number average molecular weight of the network chain (Mc) between each TMP crosslink unit was found to be pertinent in determining the tensile strength, modulus, tear strength, and hardness. The mechanical properties increased with increase in the 1/Mc value and showed a linear relationship with the high 1/Mc values (Mc below about 3000). On the other hand, chain entanglement becomes more important in affecting the mechanical properties when Mc was higher than 3000, and physical properties were less dependent on 1/Mc values in the lower range.
