    Description: A competitive enzyme-linked immunosorbent assay (cELISA) was developed by using a whole-cell antigen from a marine Brucella sp. isolated from a harbor seal (Phoca vitulina). The assay was designed to screen sera from multiple marine mammal species for the presence of antibodies against marine-origin Brucella. Based on comparisons with culture-confirmed cases, specificity and sensitivity for cetacean samples tested were 73% and 100%, respectively. For pinniped samples, specificity and sensitivity values were 77% and 67%, respectively. Hawaiian monk seal (Monachus schauinslandi; n = 28) and bottlenose dolphin (Tursiops truncatus; n = 48) serum samples were tested, and the results were compared with several other assays designed to detect Brucella abortus antibodies. The comparison testing revealed the marine-origin cELISA to be more sensitive than the B. abortus tests by the detection of additional positive serum samples. The newly developed cELISA is an effective serologic method for detection of the presence of antibodies against marine-origin Brucella sp. in marine mammals.

    Notes: Strips of polystyrene held in a flowing O2 or N2O atmosphere have been exposed to 240-600 nm radiation. The extent of photooxidation has been followed by x-ray photoelectron spectroscopy (XPS). Although N2O is a more reactive gas than O2, it produces a less oxidized polymer surface. This surprising observation can be correlated to the photochemistry occurring at the gas/polystyrene interface. © 1992 John Wiley & Sons, Inc.

    Notes: The Community Bureau of Reference has organized a collaborative interlaboratory project to improve the analytical protocol for some specific chlorobiphenyls (CBs) within the European Community (EC). A series of test procedures were prescribed to optimize the gas chromatographic conditions for splitless and on-column injection, which substantially improved the quality of data. Important parameters included the initial column and injector temperatures and the choice of the polarity of the stationary phase. In a study with cleaned eel-fat extracts, coefficients of variation for reproducibility CV(R) ranging from 11 to 24% at the 0.04-0.3 mg/kg level per CB congener were obtained. No significant difference could be found between splitless- and on-column injection.

    Notes: A comparison of the effects on peak shape is made between a 350 and 100 microliter displaced coaxial and a 350 microliter concentric cylinder electron capture cell. Peaks as fast as W1/2 = 0.5 second were detected using electronics operating in the constant current mode. The measured peak shapes are compared to those obtained by computer simulation assuming either complete mixing or plug-like flow within ECD cell. The advantage of controlling the flow pattern through the cell to obtain plug-like flow is demonstrated. Differences in peak shape between the displaced coaxial and concentric cylinder cells of the same volume are explained in terms of the electrostatic fields within the cells.

    Notes: A new design for an electron capture detector cell is described. The cell is compatible with the requirements of high resolution capillary columns and is shown to be useful in applications that require high analysis speed. A unique method of sample introduction reduces the problems of sample loss by adsorption on the surfaces of the cell. Previously reported problems of sample loss by adsorption on active surfaces within the cell when using hydrogen carrier gas are shown to have been eliminated. Examples are shown demonstrating the increased speed of analysis that can be obtained when using hydrogen carrier gas and 100 micron diameter columns.

    Notes: A homologous series of 2,4-dichlorobenzyl alkyl ethers (DCBEs) have been synthesized and purified for use as retention index calibrants and internal standards. They are stable, sensitive to ECD and FID and ideal for GC-MS, with base peaks at m/z 159/161. The retention index calibration against the n-alkanes for a linear temperature program (LTP) series is given and recommendations made for the most suitable members of the DCBE series for use as internal standards in organochlorine (OC) residue analysis.

    Notes: The versatility of the quadrupole ion trap as a mass spectrometer in which the mass resolution may be varied over a wide range has been demonstrated. Mass resolution of 0.8 × 107 and 1.2 × 107 have been obtained for ions of m/z 414 and m/z 614, respectively, well in excess of the normal value of 3m, where m is the mass of ion. At the highest mass resolution, peak-widths at half-maximum are but 52 × 10-6 u. While it has been shown previously that enhanced mass reslution in the quadrupole ion trap can be achieved by reduction of the mass scanning rate, the range of mass resolutions reported here were achieved by reducing the mass scanning rate, in stages, by an overall factor of ca 5.5 × 104.
