    Experimental investigation of transport of strongly retained species by soil columns (1997)

    Notes: Abstract Column experiments have been extensively used in transport studies of major cations but few investigations are available on migration through soils of strongly retained species that are environmentally relevant (like heavy metals). By presenting some selected experiments (lead and proton step-breakthrough tests in different conditions), this study shows that the soil-column technique is also applicable in the case of species which exhibit very large retention factors. The use of very small soil columns (about 0.4 mL of pore volumes) combined with relatively high flow rates (0.1–0.3 mL min−1 ) allows to observe the entire breakthrough curve (adsorption and desorption front up to 5000 pore volumes) in reasonable experimental time, in reproducible conditions and without experimental drawbacks. In the adopted experimental conditions no kinetic effects, related to diffuse transport and sorption reaction were recognized; moreover, Peclet number was higher than 60. Consequently, it was possible to calculate the equilibrium isotherms from the diffuse fronts of the breakthrough. Knowledge that can be derived, concerning the reversibility of the adsorption process, the influence of complexation on the adsorption, the kinetics of complex formation, and the effect of dissolution on proton transport, is also discussed.

    Notes: Summary We previously demonstrated that the lipopeptide of the myelin basic protein (MBP) immunodominant epitope in Lewis rat Palm-GpMBP(74-85) (Gp: guinea pig), which induced experimental autoimmune encephalomyelitisin vivo strongly increased the T cell proliferative responsein vitro. We extended this study to the human immunodominant epitope hMBP(83-99), synthesizing different lipophilic peptides bearing a hydrophobic chain linked through an amide or a C-C bond. To this aim, we developed a synthetic pathway for (±)-N-Fmoc-Ahd-OH (Ahd: 2-aminohexadecanoic acid) which was used to synthesize diastereomeric peptides which were successfully separated by reverse-phase high-performance liquid chromatography. MBP-specific T cell lines recognizing the immunodominant epitope hMBP(83-99) have been generated from patients affected by multiple sclerosis. Their proliferative response to the native peptide and to some lipoderivatives has been investigated. In contrast to the animal model, none of the investigated lipopeptides exhibited ‘superagonist’ activity.

    Notes: Summary The adamantane moiety was introduced in the tachykinin NK2 receptor-selective agonist [β-Ala8]-NKA(4–10) (H-Asp-Ser-Phe-Val-β-Ala-Leu-Met-NH2, MEN 10210) and in different positions of the NK2 receptor antagonist MEN 10376 (H-Asp-Tyr-d-Trp-Val-d-Trp-d-Trp-Lys-NH2) in order to investigate how this substitution affects their biological activity at tachykinin NK1, NK2 and NK3 receptors. 1-Adamantaneacetic acid (1-Ada-CH2COOH) was directly conjugated in the solid phase as the preformed OBt active ester to the N-terminal position of MEN 10210, obtaining MEN 10586 (1-Ada-CH2CO-Asp-Ser-Phe-Val-β-Ala-Leu-Met-NH2). The Pfp ester of adamantaneacetic acid (1) was prepared and used for the acylation of the N-terminal position of MEN 10376, yielding MEN 10606 (1-Ada-CH2CO-Asp-Tyr-d-Trp-Val-d-Trp-d-Trp-Lys-NH2). Compound 1 was then used to obtain the building block Fmoc-Lys(1-Ada-CH2CO)-OH as a modified amino acid for the synthesis of MEN 10818 [H-Asp-Tyr-d-Trp-Val-d-Trp-d-Trp-Lys(1-Ada-CH2CO)-NH2]. In order to investigate the biological activity of the peptide bearing the adamantane group together with the free N-terminal amino function, we synthesised MEN 10676 [H-Asp(O-2-Ada)-Tyr-d-Trp-Val-d-Trp-d-Trp-Lys-NH2] using Fmoc-Asp(O-2-Ada)-OH, in which 2-adamantanole was the protecting group of the aspartate β-COOH moiety during the peptide synthesis and survived the final peptide cleavage and deprotection carried out under controlled conditions. MEN 10586 showed an agonist activity comparable to that of the parent compound MEN 10210 at NK1 and NK2 receptors of guinea pig ileum, rabbit isolated pulmonary artery and hamster isolated trachea preparations, while it showed a 25-fold higher agonist activity at NK3 receptors of rat isolated portal vein. The three modified antagonist analogs displayed similar or reduced affinity at NK1, NK2 and NK3 receptors as compared to MEN 10376. The drop was particularly evident (〉2 log units) at the NK2 receptors of the rabbit isolated pulmonay artery.

    Description: This chapter takes into consideration two Russian translations of the Vita of Cimabue. One of these presents a selection from Le Vite de’ più eccellenti pittori, scultori, e architettori by Giorgio Vasari and is dated 1933; the other is the first complete Russian translation of Vasari’s Vite published between 1956 and 1971. The comparison of these two translations focuses on morphological, lexical and syntactical aspects of the original version. Particular attention is devoted to the translation of inter-phrasal and intra-phrasal connectors, of the coniunctio relativa and of gerunds. The chapter also reflects on the translation of Latin inscriptions, toponyms and proper nouns.
