    Beschreibung / Inhaltsverzeichnis: Zusammenfassung 17–21,5 h nach Teilhepatektomie wurde in Intervallen von 30 min der Übertritt von Hepatocyten aus der G1- in die S-Phase durch Bestimmung des Markierungsindex, der spezifischen Aktivität der DNA und der Verteilung der Radioaktivität zwischen säurelöslichem niedermolekularem Pool und DNA-Fraktion nach Applikation von tritiiertem Thymidin (3H-TdR) untersucht. 17–18 h nach Zweidrittelhepatektomie werden nur geringe Änderungen des 3H-TdR-Markierungsindex beobachtet. Zwischen 18 und 20 h nach der Operation steigt jedoch der Zellfluß aus dem G1- in das S-Kompartiment steil an; er beträgt im Mittel 8,85%/h. Anschließend nimmt der Prozentsatz markierter Hepatocytenkerne geringfügig ab und steigt 21,5 h nach Leberresektion erneut an. Änderungen in der spezifischen Aktivität der DNA und Verteilung der Radioaktivität zwischen niedermolekularer und DNA-Fraktion korrelieren weitgehend mit dem 3H-TdR-Markierungsindex. Das gleichartige Verhalten dieser drei Parameter wird diskutiert. Es kann angenommen werden, daß der Übertritt der Hepatocyten von G1 nach S diskontinuierlich verläuft und daß sich der Gehalt der Zelle an niedermolekularen DNA-Vorläufern selektiv für diejenigen Hepatocyten ändert, die in das S-Kompartiment eintreten. Durch die kombinierte Bestimmung der Zellkinetik und biochemischer Parameter können Vorgänge in einer Untergruppe auch einer nur partiell synchronisierten Zellpopulation analysiert werden.
    Notizen: Summary Transfer of hepatocytes from G1- to S-phase was studied 17 to 21.5 h after partial hepatectomy at 30 min intervals by determination of labeling index, specific activity of DNA and distribution of radioactivity between acid soluble pool and DNA fraction after application of tritiated thymidine (3H-TdR). 17 to 18 h after two-thirds hepatectomy only small changes of the 3H-TdR labeling index are observed. Between 18 and 20 h after operation, however, the cell flux from G1- to S-compartment increases rapidly; on the average it amounts to 8.85 per cent per hour. Afterwards the percentage of labeled hepatocytes decreases to a small extent and rises again 21.5 h after liver resection. Changes of specific activity of DNA and distribution of radioactivity between low-molecular and DNA fraction correlate to a high degree with 3H-TdR labeling indices. The analogous pattern of the three parameters is discussed. It can be assumed that the transfer of hepatocytes from G1- to S-progresses discontinuously, and that the cellular content of low-molecular DNA precursors changes selectively in those hepatocytes entering into the S-compartment. By combined determination of cell kinetics and biochemical parameters processes even in a subgroup of an only partially synchronized cell population can be analysed.

    Beschreibung: To evaluate changes in terrestrial and aquatic ecosystems in the Azores, we developed paleoecological and paleoclimate records from Lake Funda on Flores Island that span the last millennium. Vegetation composition (n-alkane average chain length) indicate when human activities began in the catchment area and biogenic silica tracks changes in diatom productivity. In addition, sterol hydrogenation and archaeal lipids (isoprenoid glycerol dialkyl glycerol tetraethers) trace changes in redox conditions and biogeochemical cycles, respectively. Finally, a high-resolution reconstruction using leaf wax hydrogen isotopes records changes in precipitation amount over the last millennium.
