    Description: Osteoporosis and other diseases of bone loss are a major public health problem. Here it is shown that the statins, drugs widely used for lowering serum cholesterol, also enhance new bone formation in vitro and in rodents. This effect was associated with increased expression of the bone morphogenetic protein-2 (BMP-2) gene in bone cells. Lovastatin and simvastatin increased bone formation when injected subcutaneously over the calvaria of mice and increased cancellous bone volume when orally administered to rats.     Description: The classical recessive mouse mutant, Purkinje cell degeneration (pcd), exhibits adult-onset degeneration of cerebellar Purkinje neurons, retinal photoreceptors, olfactory bulb mitral neurons, and selected thalamic neurons, and has defective spermatogenesis. Here we identify Nna1 as the gene mutated in the original pcd and two additional pcd alleles (pcd2J and pcd3J). Nna1 encodes a putative nuclear protein containing a zinc carboxypeptidase domain initially identified by its induction in spinal motor neurons during axonal regeneration.     Description: Mutations in the IDH1 and IDH2 genes encoding isocitrate dehydrogenases are frequently found in human glioblastomas and cytogenetically normal acute myeloid leukaemias (AML). These alterations are gain-of-function mutations in that they drive the synthesis of the 'oncometabolite' R-2-hydroxyglutarate (2HG). It remains unclear how IDH1 and IDH2 mutations modify myeloid cell development and promote leukaemogenesis. Here we report the characterization of conditional knock-in (KI) mice in which the most common IDH1 mutation, IDH1(R132H), is inserted into the endogenous murine Idh1 locus and is expressed in all haematopoietic cells (Vav-KI mice) or specifically in cells of the myeloid lineage (LysM-KI mice). These mutants show increased numbers of early haematopoietic progenitors and develop splenomegaly and anaemia with extramedullary haematopoiesis, suggesting a dysfunctional bone marrow niche. Furthermore, LysM-KI cells have hypermethylated histones and changes to DNA methylation similar to those observed in human IDH1- or IDH2-mutant AML. To our knowledge, our study is the first to describe the generation and characterization of conditional IDH1(R132H)-KI mice, and also the first report to demonstrate the induction of a leukaemic DNA methylation signature in a mouse model.     Description: Observations at 5 to 25 microns show that the object discovered by Strom et al. (1976) at 2.2 microns within the densest part of the L1551 dark cloud is a strong source of radiation from grains as cool as 130 K. The energy distribution resembles that of infrared objects embedded within other molecular cloud cores, but implies a total luminosity of only 30 solar luminosities. The luminosity of the source and its proximity to the other T Tauri stars suggests that it is a 1-2 solar-mass premain-sequences star still swaddled within the L1551 cloud. The radio and optical evidence for mass motions around IRS 5 may mean that IRS 5 has been flaring in FU Ori-type eruptions within the last 1000 years.

    Description: The generation of tunable visible, infrared, and ultraviolet light is examined, along with the control of this light by means of novel mode-locking and modulation techniques. Transient mode-locking of the Nd:YAG laser and generation of short tunable pulses in the visible and the alkali metal inert gas excimer laser systems were investigated. Techniques for frequency conversion of high power and high energy laser radiation are discussed, along with high average power blue and UV laser light sources.

    Description: Analytical and experimental studies of the rapid transfer of stored populations from metastable states to selected target states of a different species are reported. Both laser-induced or laser-switched collision and laser-induced two-photon spontaneous emission are described. It is shown that the laser-induced collision method is particularly useful in the visible and UV spectral regions. It has applications in photochemistry, gas-phase kinetics, and in high-power, high-energy gas-phase lasers. The anti-Stokes source is useful in the VUV and soft X-ray spectral regions.

    Description: A type of high resolution XUV radiation source is described which is based upon spontaneous anti-Stokes scattering of tunable incident laser radiation from atoms excited to metastable levels. The theory of the source is summarized and two sets of experiments using He (1s2s)(1)S atoms, produced in a CW hollow cathode and in a pulsed high power microwave discharge, are discussed. The radiation source is used to examine transitions originating from the 3p(6) shell of potassium. The observed features include four previously unreported absorption lines and several sharp interferences of closely spaced autoionizing lines. A source linewidth of about 1.9 cm(-1) at 185,000 cm(-1) is demonstrated. Previously announced in STAR as N83-18422

    Description: Some preliminary IRAS results in the form of images at 12, 25, 60, and 100 microns of an 8 deg x 15 deg area around the galactic center are presented. These absolute intensity maps have unprecedented sensitivity combined with high angular resolution, wide field coverage, and large wavelength range. They give a broad view of the central galaxy revealing previously unseen details, especially in regions far from the central few arcmin. Well-defined infrared features in the nucleus correspond to the nuclear radio sources Sgr A, B2, C, and D. Extremely faint structures are detected, such as the cold molecular cloud associated with Sgr B2 which has never before been detected at wavelenths shorter than 40 microns.
