    Notes: Abstract We have recently reported on the isolation of a 5.7 kb segment of Chinese hamster ovary cell genomic DNA, Expression Augmenting Sequence Element (EASE), which when used in bicistronic expression vectors allows the development of stable Chinese hamster ovary cell pools in a five to seven week time period that express high levels of recombinant protein (6–25 μg 10-6 cells/day depending on the protein). In the present study, we have mapped the activity of the EASE to a 2.1 kb region using colony forming assays and developed bicistronic expression vectors with the smaller EASE or control lambda DNA. The recovery of pools expressing the hematopoietic growth factor, FLT3 Ligand, in methotrexate-containing media took 1 to 4 weeks less when using EASE expression vectors compared with control vectors. The cell pools developed with the EASE and control vectors had similar final protein expression levels. Southern blot analysis suggested the expression cassette from the EASE containing vectors integrated in tandem arrays arranged in either head to head or head to tail fashion. By contrast, control vectors appeared to integrate with multiple interruptions to the expression vector. Thus, the EASE, within a bicistronic expression vector, appeared to facilitate tandem vector integration and reduce the time required to develop cell pools for protein expression.

    Notes: Abstract The dihydrofolate reductase-deficient Chinese hamster ovary cell line, DXB11-CHO, commonly used as a host cell for the production of recombinant proteins requires 7.5% serum-supplementation for optimal growth. Regulatory issues surrounding the use of serum in clinical production processes and the direct and indirect costs of using serum in large-scale production and recovery processes have triggered efforts to derive serum-independent host cell lines. We have successfully isolated a serum-free host that we named Veggie- CHO. Veggie-CHO was generated by adapting DXB11-CHO cells to growth in serum-free media in the absence of exogenous growth factors such as Transferrin and Insulin-like growth factor, which we have previously shown to be essential for growth and viability of DXB11- CHO cells. Veggie-CHO cells have been shown to maintain an average doubling time of 22 hr in continuous growth cultures over a period of three months and have retained the dihydrofolate reductase -deficient phenotype of their parental DXB11-CHO cells. These properties and the stability of its serum-free phenotype have allowed the use of Veggie- CHO as host cells for transfection and amplified expression of recombinant proteins. We describe the derivation a serum-free recombinant cell line with an average doubling time of 20 hr and specific productivity of 2.5 Units recombinant Flt-3L protein per 10e6 cells per day.

    Notes: Two overriding characteristics for major applications of plastics in consumer products are: - the need for high volume production and - their exposure to hostile environments.Successful application and continued growth for plastics in these products requires knowledgeable prediction of performance in the process to produce the plastic part and in the environments to which it will be exposed during the product's lifetime. Progress on establishing relationships between material properties and structure are described with emphasis on response to melt process conditions and behavior when exposed to hostile environments.Commercial plastics and the processes by which they are converted into useful applications are complex systems. A multitude of interacting variables must be dealt with in order to achieve successful implementation in commercial products. Specific examples are discussed in the area of processing for non-Newtonian flow characteristics and overt effects in injection molding and extrusion, tailoring of polymers within established rheological limits and the influence of melt processing on physical and mechanical properties. The influence of application environments is discussed and specific examples are given for environmental stress cracking, stress-environment embrittlement and the effects of hot water solutions on polymer systems.

    Description: Computations were made for those test cases of Problem 3 which were designated as laminar flows, viz., test cases 3.1, 3.2, 3.4, and 3.5. These test cases corresponded to flows over a flat plate and a compression ramp at high Mach number and at high Reynolds number. The computations over the compression ramps indicate a substantial streamwise extent of separation. Based on previous experience with separated laminar flows at high Mach numbers which indicated a substantial effect with spatial grid refinement, a series of computations with different grid sizes were performed. Also, for the flat plate, comparisons of the results for two different algorithms were made.

    Description: A development status evaluation is presented for CFD methods applicable to fuselage-integrated scramjet powerplant incorporating hypersonic vehicles; these methods are critically important due to the unavailability of experimental facilities for such elevated Mach number/high-enthalphy conditions. Advancements are required in algorithm robustness and speed, geometric flexibility, and the inclusion of more complete flow physics. The most serious deficiencies lie in turbulence modeling, the lack of complete transition-prediction methods, and combustion modeling.

    Description: An implicit multigrid method for the compressible Navier-Stokes equations is applied to a series of two-dimensional, steady, laminar hypersonic flows over a compression ramp, including both attached and separated flow conditions. The algorithm uses upwind-biased differencing for the convective and pressure terms and central differencing for the shear-stress and heat-transfer terms. An implicit spatially factored scheme is used to advance the solution in terms on a given mesh, and a V-cycle multigrid strategy is used to accelerate convergence. The multigrid algorithm provided an order of magnitude decrease in the computational time required to obtain a converged position of the separation location and enabled a substantially grid-independent result to be obtained for each of the flows considered.

    Description: Patched grid calculations within the framework of an implicit, flux vector split upwind/relaxation algorithm for the Euler equations are presented. Aspects of computing on patched grids are discussed including the effect of a metric-discontinuous interface on the convergence rate of the algorithm, and the effect of curvature along an interface. Applications to a converging-diverging nozzle including effects of choking and bypass slots in two dimensions are presented.
