    Notes: The formation of polyhydrogen adducts of peptide molecules during fast atom bombardment (FAB) mass spectrometry (MS) is shown to be matrix dependent. FAB/MS analyses completed in glycerol solvent produced higher than expected peak intensities at m/z corresponding to (M + 2)+, (M + 3)+, and (M + 4)+ when the contributions of all stable isotopes present in the sample are included. Analysis of the same peptides in thioglycerol by positive or negative ion FAB yielded peak intensity distributions for the protonated molecule which matched those predicted from the natural abundance of stable isotopes. These findings suggest that thioglycerol should be the FAB matrix in analyses of peptides when determination of elemental or isotopic composition is desired.

    Notes: An extraction-derivatization method suitable for the analysis of subnanogram amounts of biogenic amines in aqueous solution has been developed. The most satisfactory procedure for the analysis of these compounds was reaction with 3,5-ditrifluorobenzoylchloride (DTFMBCl) in phosphate buffer at pH 7.2 followed by extraction of the resultant amide esters into ethyl acetate. This was followed by hydrolysis of the phenolic ester functionalities by shaking the organic layer with 10 M ammonium hydroxide. The phenolic and alcoholic hydroxyl groups were then reacted with bistrimethylsilylacetamide and the trimethylsilyl-DTFMB amides were then analysed by gas chromatography/mass spectrometry in the negative ion chemical ionization mode with methane as reagent gas. The limits of detection for these derivatives was 〈1 pg and the method was readily applicable to the extraction and analysis of 0.5 ng of a given biogenic amine.

    Notes: A new technique known as thermally-assisted fast atom bombardment (TA-FAB) has been applied to the analysis of a series of cyclic tetrapeptide mycotoxins in order to demonstrate the usefulness of the method for structural elucidation. TA-FAB uses saturated aqueous solutions of highly hydroxylated compounds, such as fructose, as alternatives to the usual viscous liquid matrices employed in conventional FAB. During the TA-FAB analysis, the probe tip is resistively heated causing diffeiences to occur in the desorption profiles for the analyte and the matrix ions enabling an optimization for analyte desorption as a function of temperature. In this study, direct comparisons are made between TA-FAB, conventional FAB, and electron impact ionization for the analysis of the Helminthosporium carbonum mycotoxins at the 1.5 μg level. The results demonstrate the superior capacity of TA-FAB to provide both molecular weight confirmation and significant fragmentation to aid in the structural elucidation of these important biomolecules.

    Notes: Prednisolone, dexamethasone and betamethasone were labelled with deuterium via a simple synthetic procedure and used as internal standards in the gas chromatographic/mass spectrometric analysis of the corresponding undeuterated compounds. The mass spectrometer was used in the negative chemical ionization mode, which gave fragmentation of the methoxime trimethylsilyl ether derivatives favourable for their quantification. The method was applicable to the quantification of synthetic corticosteroids contained in human aqueous humour in the 0.1-10-ng range.

    Notes: Pentafluorobenzyl hydroxylamine has been used as a derivatization reagent in the analysis of corticosteroids by gas chromatography/negative ion chemical ionization (GC/NICI) mass spectrometry. The resulting pentafluorobenzyloxime-trimethylsilyl (PFBO/TMS) derivatives were generally formed in moderate yield but, despite this, the use of these derivatives resulted in a ∼ 10-fold improvement in the capability of identification of corticosteroids by GC/NICI mass spectrometry in comparison with the methoxime/TMS derivatives previously employed. The NICI mass spectra of PFBO/TMS derivatives were simple, with most of the ion current being carried by the [M - C6F6CH2]-. or [M - PFB]-. ion and by a reagent-specific peak at m/z 196. The derivatives have been employed in the quantification of ∼400 pg amounts of endogenous hydrocortisone present in samples of human aqueous humour and are readily applicable to the determination of low-picogram amounts of these compounds in biological media.

    Notes: A fermentor system with an external filtration loop has been developed to control the growth and sporulation of yeast in a single vessel. Excess growth medium, instead of being removed by centrifugation, is removed by filtration and replaced with acetate sporulation medium. The technique did give 80% sporulation after 20 hr, greatly improving the rate and degree of synchrony of sporulation and it also eliminated the contamination hazard of the conventional harvest technique, centrifugation, and resuspension of vegetative cells in sporulation medium. Furthermore it permits proper control of the environmental conditions throughout the growth, exchange, and sporulation phase. In this technique 100% recycle of biomass is achieved without any packing of the cells on the filter. This technique has wide application in the study of industrial fermentation that involves microbial differentiation such as the production of ergot alkaloids, bacitracin, and cephalosporin.

    Notes: Devonian shales and other unconventional resources can be highly fractured and may have significant amounts of gas stored by adsorption. Conventional experiments are not well suited for characterizing the properties important for describing gas storage and transport in these media. Here, X-ray computed tomography scanning is used to determine gas storage in dynamic gas flow experiments on Devonian shale samples. Several important properties are obtained from these experiments, including fracture widths, adsorption isotherms, and matrix porosities and permeabilities.

    Notes: Nuclear magnetic resonance (NMR) spin-lattice relaxation measurements are used to investigate pore structures and fluid phase distributions in porous media. A new method for estimating relaxation time distribution functions from measured relaxation data is presented using a B-spline basis to represent the distribution function and Tikhonov regularization to stabilize the estimation problem. Surface relaxivity, which is required to convert relaxation time distributions to pore-size distributions of fluid phase distributions at partial saturations, is determined using pore volume-to-surface-area ratios estimated by NMR diffusion measurements. This approach was validated by analyzing certain model porous media with known pore volume-to-surface-area ratios. The method is demonstrated by determining pore-size and fluid phase distributions of sandstone and carbonate samples, as well as by comparing the pore-size distributions of chalk samples obtained by this methodology with those estimated by mercury porosimetry.
