    Notes: Abstract Nowick and his associates have stated that many ionic crystals and glasses exhibit a loss per cycle which is independent of frequency over an appreciable range and have suggested that such behavior constitutes a “new universality”. Furthermore, much such data seem to approach an asymptotic, nearly temperature-independent ac loss at sufficiently low temperatures. In order to further evaluate these conclusions, small-signal ac relaxation data for a CaTiO3:30% Al3+ ceramic material are analyzed in detail and the results compared to those published by Nowick and associates for the same material. It is found that a plausible conducting-system dispersion model based on the effective-medium approximation for hopping charges yields results globally similar to, but somewhat different in detail from, those of Nowick et al. But a response model which includes both such conducting-system response and dielectric-system dispersion well fits the data over a wide temperature range. To do so, it requires the presence of a non-zero high-frequency-limiting resistivity probably arising from localized charge motion. No constant-loss individual dispersions appear in the model, but it nevertheless yields approximately constant loss over a limited frequency range at low temperatures. It suggests that asymptotic behavior is associated with the nearly temperature-independent dielectric-dispersion contribution to the response at low temperatures, and it does not verify the Nowick conclusion that the slope of the ac conductivity approaches a constant value near 0.6 at high temperatures.

    Notes: Abstract Arousal from hibernation requires thermogenesis in brown adipose tissue, a process that is stimulated by β-adrenergic signals, leading to a rise in intracellular 3′,5′-cyclic adenosine monophosphate AMP (cAMP) and activating cAMP-dependent protein kinase A (PKA) to phosphorylate a suite of target proteins and activate lipolysis and uncoupled respiration. To determine whether specific adaptations (perhaps temperature-dependent) facilitate PKA kinetic properties or protein-phosphorylating ability, the catalytic subunit of PKA (PKAc) from interscapular brown adipose of the ground squirrel Spermophilus richardsonii, was purified (final specific activity = 279 nmol phosphate transferred per min per mg protein) and characterized. Physical properties of PKAc included a molecular weight of 41 kDa and an isoelectric point of 7.8 ± 0.08. A change in assay temperature from a euthermic value (37 °C) to one typical of hibernating body temperature (5 °C) had numerous significant effects on ground squirrel PKAc including: (a) pH optimum rose from 6.8 at 37 °C to 8.7 at 5 °C, (b) Km values at 37 °C for Mg.ATP (49.2±3.4 M) and for two phosphate acceptors, Kemptide (50.0±5.5 M) and Histone IIA (0.41 ± 0.05 mg/ml) decreased by 53%, 80% and 51%, respectively, at 5 °C, and (c) inhibition by KCl, NaCl and NH4Cl was reduced. However, temperature change had little or no effect on Km values of rabbit PKAc, suggesting a specific positive thermal modulation of the hibernator enzyme. Arrhenius plots also differed for the two enzymes; ground squirrel PKAc showed a break in the Arrhenius relationship at 9 °C and activation energies that were 29.1 ± 1.0 kJ/mol for temperatures 〉9 °C and 2.3-fold higher at 68.1 ± 2.1 kJ/mol for temperatures 〈9 °C, whereas the rabbit enzyme showed a breakpoint at 17 °C with a 13-fold higher activation energy over the lower temperature range. However, fluorescence analysis of PKAc in the absence of substrates, showed a linear change in fluorescence intensity and wavelength of maximal fluorescence over the entire temperature range; this suggested that the protein conformational change indicated by the break in the Arrhenius plot was substrate-related. Temperature change also affected the Hill coefficient for cAMP dissociation of the ground squirrel PKA holoenzyme which rose from 1.12 ± 0.18 at 37 °C to 2.19 ± 0.07 at 5 °C, making the release of catalytic subunits at low temperature much more responsive to small changes in cAMP levels. Analysis of PKAc function via in vitro incubations of extracts of ground squirrel brown adipose with 32P-ATP + cAMP in the presence versus absence of a PKA inhibitor, also revealed major differences in the patterns of phosphoproteins, both between euthermic and hibernating animals as well as between 37 and 5 °C incubation temperatures; this suggests that there are both different targets of PKAc phosphorylation in the hibernating animal and that temperature affects the capacity of PKAc to phosphorylate different targets. Both of these observations, plus the species-specific and temperature-dependent changes in ground squirrel PKAc kinetic properties, suggest differential control of the enzyme in vivo at euthermic versus hibernating body temperatures in a manner that would facilitate a rapid and large activation of the enzyme during arousal from torpor.

    Notes: The double melting endotherm of spunbonded isotactic polypropylene (iPP) fabrics was investigated by monitoring changes in the solid-state NMR spectrum that result from thermal annealing. The DSC melting thermogram was found to change from a double to a single endotherm at anneal temperatures ≥156°C, with a concomitant increase in percent crystallinity. All of the carbon resonances in the CP/MAS NMR spectrum of the purely crystalline phase of iPP were found to be composed of multiple peaks with relative intensities that depend on anneal temperature. By monitoring the changes in the distribution of intensity among the various peaks of a given resonance, a transition temperature of 156°C was identified. Arguments are presented that this redistribution of intensity within a given carbon resonance characterizes the transformation from the α1 to the α2 monoclinic crystal form. The exothermicity associated with this transformation is responsible for the observation of a double melting endotherm by DSC. The splitting patterns observed in the NMR spectrum are discussed in terms of interlayer distances between layers of isochiral helices and the density of exposed methyls at the contact faces of these interlayers. © 1996 John Wiley & Sons, Inc.

    Description: The baseline experiment program is concerned with future space experiments and cover the scientific disciplines of astronomy, space physics, space biology, biomedicine and biotechnology, earth applications, materials science, and advanced technology. The experiments within each discipline are grouped into functional program elements according to experiments that support a particular area of research or investigation and experiments that impose similar or related demand on space station support systems. The experiment requirements on module subsystems, experiment operating modes and time profiles, and the role of the astronaut are discussed. Launch and rendezvous with the space station, disposal, and on-orbit operations are delineated. The operational interfaces between module and other system elements are presented and include space station and logistic system interfaces. Preliminary launch and on-orbit environmental criteria and requirements are discussed, and experiment equipment weights by functional program elements are tabulated.

    Description: RAM, a family of payload carriers that operate in conjunction with the Space Shuttle, is shown to provide flexible and economical laboratories and facilities for the conduct of manned and man-tended scientific and applications investigations in near-earth orbit. Two primary mission modes of RAM are discussed in terms of capability provided, and major advantages and constraints. The Sortie mission mode uses the Shuttle orbiter as the space platform and RAM as the scientific equipment carrier to provide a flexible and economical means for conducting short-duration manned investigations. The man-tended observatory mode uses Shuttle for delivery, periodic on-orbit update and service, and retrieval of the observatory, with RAM providing the interface with the Shuttle for servicing and the space platform functions for operation.
