    Description: Multicellular masses of the cellular slime mold Dictyostelium discoideum, under conditions which ordinarily suppress cell differentiation, develop clusters of stalk cells and spore cells when implanted with Sephadex particles that had been soaked in 5 X 10(-3)M cyclic adenosine monophosphate (AMP).     Notes: Abstract Various factors that can effect thermal blooming in stagnation zone situations are examined, including stagnation-zone motion, longitudinal air motion in the neighborhood of the stagnation zone, and the effects of scenario noncoplanarity. Of these effects, only the last offers reasonable hope of reducing the strong thermal blooming that normally accompanies stagnation zones; in particular, noncoplanarity should benefit multipulse more than cw beams. The methods of treating nonhorizontal winds hydrodynamically for cw and multipulse steady-state sources are discussed. Aspects of pulse “self-blooming” are also considered.

    Notes: Abstract The computation of time-dependent three-space-dimensional laser beam propagation is described. The methods are applicable to the propagation of high energy laser beams through the atmosphere in the presence of a horizontal wind and turbulence for most situations of interest. Possible cases are propagation of cw beams through stagnation zones, multi-pulse propagation, including the self-consistent treatment of pulse self-blooming, and propagation involving transonic slewing. The solution of the Maxwell wave equation in Fresnel approximation is obtained by means of a discrete Fourier transform method, which, surprisingly, gives excellent results for diffraction problems. The latter provide a stringent test for the accuracy of any solution method. Considerable use is also made of discrete Fourier transform methods in solving the hydrodynamic equations. The treatment of turbulence is based on the generation of random phase screens at each calculation step along the propagation path. In a time-dependent calculation the random phase screens can be either made to move with the wind at a given propagation position or generated anew for each successive time.

    Notes: The isotype pattern of tropomyosin was investigated in normal and dystrophic avian pectroal muscle using two-dimensional gel electrophoresis. Previous reports have shown that adult pectoral muscle of chickens contains only the α-subunit of tropomyosin and a breast-type troponin-T (TN-T), whereas pectoral fetal muscle contains both α- and β-tropomyosin and leg-type TN-T. The change from the fetal to the adult forms begins shortly after hatching. It has been previously reported that avian dystrophic pectoral muscle contains both the leg- and breast-type TN-T; we show that in avian dystrophic muscle there is also persistent expression of the β-subunit of tropomyosin.

    Notes: Vital dyes have been extensively used in many laboratories to distinguish the prestalk and prespore zones of a migrating slug in Dictyostelium discoideum. Here we present evidence that the two zones do not form immediately following aggregation, as is sometimes assumed, but only after 1-4.5 hr of migration. When the transition to the two-toned state occurs, it changes very quickly, in a matter of approximately 10 min. Furthermore, it can be rapidly induced by submerging the slug in mineral oil. The time of appearance of the zones is correlated with previously reported changes in the distribution of cyclic AMP secretion and with changes in regulation properties along the axis of the slug.

    Notes: Ammonia, at moderate concentrations, stimulates aggregate density of Dictyostelium mucoroides. The range of stimulatory concentrations includes ammonia concentrations established by populations of amebae. At higher concentrations, ammonia inhibits aggregate density.A quantitative test of the hypothesis that ammonia is the aggregation-suppressing gas has been carried out. The concentration of ammonia established over defined populations of amebae is one or two orders of magnitude lower than the concentration of ammonia required to exert the same degree of inhibitory effect as the populations of amebae exert.An additional difference between ammonia and the aggregation-suppressing gas is the fact that increasing concentrations of the aggregation-suppressing gas cause progressively larger aggregation streams, while increasing concentrations of ammonia have no such effect.The stimulatory effect of ammonia at concentrations established by ameba populations indicates that ammonia must be included in the variables affecting the aggregation process and that this ammonia effect must be taken into account in any quantitative modelling of the aggregation process.
