Abstract The role of isopentenyl adenine (2ip) in cereal tissue and plant regeneration was analyzed by addition of this compound into various cultural media. Preculture of rice calli in the presence of certain levels of 2ip before transferring onto a normal tissue culture regeneration medium increased significantly the rate of embryogenesis as well as plant regeneration. Different combinations of 2ip and 2,4-D in preculture media had profound effects on cell differentiation. The procedures of embryogenesis and plant regeneration can be separated by manipulating the ratio of 2,4-D to 2ip in the culture media. Various possible functions of 2ip, 2,4-D and KT in cereal regeneration are also suggested.
