    Notes: Summary The efficacy of different hexacyanoferrates(II) in preventing the enteral absorption of134Cs was studied in piglets. As compared to the controls, oral application of134Cs together with KFe[Fe(CN)6], NH4Fe[Fe(CN)6], or Fe4[Fe(CN)6]3 resulted in a strong reduction of the134Cs-uptake by more than 97%. The decrease in enteral absorption depends on the dose of administered hexacyanoferrate(II), whereas differences between the compounds under study were small. The biological half-life of134Cs in non-hexacyanoferrate(II) treated piglets was 21.6±3.3 days (mean±SD).

    Notes: Summary Injection i. v. of labetalol, a new adrenergic alpha- and beta-blocking agent, decreased arterial blood pressure in 9 hypertensive subjects resting in the supine position, when standing and during supine exercise. Heart rate after labetalol was unchanged in the resting supine position, and it fell in the latter two conditions. Plasma noradrenaline concentration was higher after labetalol in all three experiments as compared to a control study. Plasma adrenaline after labetalol was increased only in the standing position, when the highest plasma noradrenaline concentrations were observed. Blood glucose concentration tended to increase after labetalol, but the difference was not statistically significant. The changes in plasma noradrenaline and blood glucose after labetalol mimic findings observed after alpha-adrenergic receptor blockade. The beta-adrenergic receptor blocking property of labetalol is responsible for the reduced heart rate and it is likely to contribute to the higher plasma noradrenaline concentration observed when standing and during supine exercise.

    Notes: Summary The acute haemodynamic effects of the beta-adrenoreceptor agonist, prenalterol, were studied in six patients with chronic end-stage renal failure. Prenalterol 0.8 mg, 1.6 mg, and 3.2 mg was administered i.v. as a bolus, and after the last dose the selective adrenergic beta-1-receptor antagonist metoprolol was administered i.v. in doses of 5 and 10 mg. The haemodynamic effects of the drugs were investigated using impedance cardiography and radionuclide angiocardiography. The main haemodynamic effects were a dose-related chronotropic effect, demonstrated by an increase in heart rate (26%; 〈0.05), and an inotropic effect, shown by an increase in stroke volume index (20%;p〈0.05) and left ventricular ejection time (12%;p〈0.05); the cardiac index was increased by 47% (p〈0.05). Transmural myocardial perfusion (DPTI/SPTI ratio) was decreased by 22% (p〈0.05) after prenalterol. It is concluded that prenalterol has positive inotropic and chronotropic effects in patients with chronic renal failure, that the improvement in left ventricular performance is at the expense of a decreased transmural myocardial perfusion, and that metoprolol is a specific antidote.

    Notes: Chick embryo fibroblasts were maintained at confluency for up to 35 days in medium containing 0.5% or 0.75% fetal bovine serum or 2.5% or 5.0% horse serum. At weekly intervals cells were subcultured and serially propagated in medium containing 10% FBS until their replicative lifespans were completed. The results showed that the replicative lifespan of embryonic chick fibroblasts was dependent on the cumulative number of population doublings undergone by the culture and was not related to the calendar time cells were in culture. Further characterization of 0.75% FBS maintained chick cells returned to 10% FBS medium showed that cells had protein contents and incorporated 3H-thymidine into DNA at a rate that resembled that of young cells, despite an advanced chronological age.

    Notes: Chick embryo fibroblasts serially propagated in media containing division ratelimiting amounts of fetal bovine serum underwent premature culture senescence as illustrated by accelerated declines in the number of cells incorporating 3H-thymidine, increased population doubling times, reduced cell densities at subcultivation, and reduced replicative life-spans compared to cells grown in medium containing non-rate-limiting amounts of serum. Low serum serially propagated “senescent” cultures returned to 10% serum containing medium had proliferative rates, incorporated 3H-thymidine, and attained saturation densities at confluency similar to younger cells. “Senescent” cells serially propagated in low serum and returned to 10% serum achieved life-spans similar to cells continuously grown in the presence of 10% serum. The results of these and other studies show that cells serially propagated in the presence of division rate-limiting amounts of fetal bovine serum, or at high inoculation densities, accumulate a substantial number of cells in the population during exponential growth conditions that are not senescent but are prevented from entering DNA synthesis becuase of mitogen limitations. Our results indicate that the amount of serum mitogen in the growth medium affects only the rate at which cells express their genetically predetermined replicative potential and not the replicative lifespan per se. These results are discussed in relation to the techniques that should be employed for studying cellular aging and the mechanism of senescent cell formation.

    Notes: Groups of cDNA clones encoding abundant leaf proteins or derived from genes (gene families) with other features of interest have been selected from a barley leaf cDNA library. The characteriaztion of nine of the groups is summarized and includes information on the tissue specificity and light dependence of expression of their corresponding genes. Different types of control of gene expression are represented in the collection: leaf-specific expression, both stimulated and inhibited by light, constitutive expression, and expression that is maximal in one case in coleoptiles and in two cases in meristematic tissue. For the light-stimulated genes (gene families) encoding chloroplast proteins (Cab, RbcS, and plastocyanin), relative and absolute levels of messengers were determined as a function of cell age in sections of 7-day-old barley leaves grown under diurnal conditions. Key parameters of cell growth (protein, RNA, and DNA accumulation) were determined in the same leaf sections. The main conclusions of the expression studies are as follows: (1) Light is in no case a requirement for gene expression although it has significant stimulatory effect on some genes; (2) weak expression of some genes coding for chloroplast proteins was detected in the leaf-like, white coleoptiles, whereas expression in roots could not be detected; (3) The cab, rbcS, and plastocyanin genes are expressed very early during leaf cell differentiation, when the plastids morphologically are still in their amyloplast-amoeboid stages; (4) The expression of the cab, rbcS, and plastocyanin genes is not coordinated during leaf cell development.
