    Notes: Abstract In this paper, a new scheme of stochastic averaging using elliptic functions is presented that approximates nonlinear dynamical systems with strong cubic nonlinearities in the presence of noise by a set of Itô differential equations. This is an extension of some recent results presented in deterministic dynamical systems. This method involves the laborious manipulation of Jacobian elliptic functions such as cn, dn and sn rather than the usual trigonometric functions. The use of a symbolic language such as Mathematica reduces the computational effort and allows us to express the results in a convenient form. The resulting equations are Markov approximations of amplitude and phase involving integrals of elliptic functions. Finally, this method was applied to study some standard second order systems.

    Notes: Abstract We formulate the method of averaging for perturbations of Euler's equations of rotational motion. Euler's equations are three strongly nonlinear coupled differential equations that can be viewed as a three dimensional oscillator. The method of averaging is used to determine the long-term influence of perturbation terms on the motion by averaging about the nominal rigid body motion. The treatment is applicable to a large class of motions including precession with large nutation – it is not restricted to small motions about simple spins or nearly axi-symmetric bodies. Three examples are shown that demonstrate the accuracy of the method's predictions.

    Notes: Abstract We investigate the system $$\ddot x - x\cos \varepsilon 1 + x^3 = 0$$ in which ε≪1 by using averaging and elliptie functions. It is shown that this system is applicable to the dynamics of the familiar rotating-plane pendulum. The slow foreing permits us to envision an ‘instantancous phase portrait’ in the $$x - \dot x$$ phase plane which exhibits a center at the origin when cos ε1≤0 and a saddle and associated double homoclinic loop separatrix when cos ɛ 1 〉 0. The chaos in this problem is related to the question of on which side (left (=L) or right (=R)) of the reappearing double homoclinic loop separatrix a motion finds itself. We show that the sequence of L's and R's exhibits sensitive dependence on initial conditions by using a simplified model which assumes that motions cross the instantancous separatrix instantancously. We also present an improved model which ‘patches’ a separatrix boundary layer onto the averaging model. The predictions of both models are compared with the results of numerical integration.

    Notes: Negative-ion fast atom bombardment mass spectrometry and metastable ion studies were applied to the characterization of three isomeric fluorine-containing sugars. Peculiar fragmentation processes were evidenced for each isomer, enabling their differentiation. Comparison with the behaviour of a non-fluorinated analogue allowed investigation of the role of fluorine in the decomposition processes of these compounds.

    Notes: Recent evidence suggests that the accessory olfactory system (AOS) may mediate chemoreception before birth. Such a capability may allow the fetus to begin to sample chemical stimuli from the outside world, a possibility that has important developmental and evolutionary implications. Herein we describe the development in the mouse of the duct that connects the vomeronasal organ (VNO), containing the primary receptor neurons of the AOS, with the nasal cavity and thus with external stimuli. Twenty-four mice, four at each of six different ages from the last day of gestation through 25 days of age, were fixed and embedded in glycol methacrylate. Serial sections were examined under the light microscope so that the VNO duct could be reconstructed in three dimensions. Results confirm an earlier study which demonstrated that the VNO duct is not patent before birth. The duct becomes patent sometime after the first day of life but remains in an immature condition throughout the normal prepubertal period. During this period the duct is characterized by an internal surface that is rapidly desquamating such that the lumen of the duct contains sloughed tissue. These results suggest that the VNO is unlikely to function in the prenatal period, since the route for external stimuli to reach its receptor surface is blocked. The protracted period of VNO duct development reported here is consistent with the great bulk of data on the AOS of mammals which firmly establishes its role in the detection of pheromones which coordinate reproduction. © 1993 Wiley-Liss, Inc.

    Notes: A series of 2- and 4-quinolones substituted in the hetero-ring have been evaluated using 13C NMR techniques. Substituent effects are discussed as an aid to evaluation of potential tautomerization in the 2-quinolone systems. The observed substituent shifts for systems having a hydroxy function at the 4-position in the 2-quinolones can be explained without implementing the tautomerism to a 4-quinolone.

    Notes: The interaction of insulin-like growth factors (IGFs) with the IGF-1 receptor is an important step in the control of cell proliferation and development. In particular, IGF-1 and IGF-2 are key regulators of central nervous system development, and may modulate the growth of glial tumors. We have investigated the growth factor regulation of the human glioblastoma cell line T98G. These cells growth arrested in serum-free medium at 34°C, despite their secretion of substantial amounts of bioactive IGF-1. To be stimulated to divide, growth-arrested cells required the addition of platelet-derived growth factor (PDGF) or its equivalent, 1% serum. Cell proliferation in serum-free medium could also be obtained by shifting the cells to a temperature of 39.6°C. Treatment of growth-arrested cells with PDGF or temperature shift was accompanied by a transient increase in the expression of the mRNA for the IGF-1 receptor. Transfection with a plasmid constitutively expressing the full cDNA for the human IGF-1 receptor allowed autonomous growth in serum-free medium at 34°C. By contrast, growth induction by growth factors or temperature shift was abrogated by transfection of the cells with a plasmid expressing a 300 bp segment of mRNA antisense to the IGF-1 receptor mRNA. Cloning in soft agar was also inhibited by expression of antisense IGF-1 receptor mRNA. These results demonstrate that the IGF-1 receptor is strictly required for the growth of T98G glioblastoma cells. Moreover, the autocrine interaction of IGF-1 with its receptor regulates both autonomous and anchorage-independent growth of these cells. © 1994 wiley-Liss, Inc.

    Description: Higher order chromatin structure presents a barrier to the recognition and repair of DNA damage. Double-strand breaks (DSBs) induce histone H2AX phosphorylation, which is associated with the recruitment of repair factors to damaged DNA. To help clarify the physiological role of H2AX, we targeted H2AX in mice. Although H2AX is not essential for irradiation-induced cell-cycle checkpoints, H2AX-/- mice were radiation sensitive, growth retarded, and immune deficient, and mutant males were infertile. These pleiotropic phenotypes were associated with chromosomal instability, repair defects, and impaired recruitment of Nbs1, 53bp1, and Brca1, but not Rad51, to irradiation-induced foci.     Description: Parasite-specific CD4+ T cells have been shown to transfer protection against Leishmania major in susceptible BALB/c mice. An epitope-tagged expression library was used to identify the antigen recognized by a protective CD4+ T cell clone. The expression library allowed recombinant proteins made in bacteria to be captured by macrophages for presentation to T cells restricted to major histocompatibility complex class II. A conserved 36-kilodalton member of the tryptophan-aspartic acid repeat family of proteins was identified that was expressed in both stages of the parasite life cycle.
