    Description: Current vaccine development strategies for malaria depend on widespread immunological responsiveness to candidate antigens such as the zygote surface antigens and the sporozoite coat protein, the circumsporozoite (CS) protein. Since immunological responsiveness is controlled mainly by genes mapping within the major histocompatibility complex (MHC), the humoral immune response to the zygote surface antigens and the cytotoxic T lymphocyte (CTL) response to the CS protein were examined in MHC-disparate congenic mouse strains. Only two of six strains responded to the 230-kilodalton zygote surface antigen and another two strains responded to the 48/45-kilodalton surface antigen. From two mouse strains, expressing between them five different class I MHC molecules, there was recognition of only a single CTL epitope from the CS protein, which was from a polymorphic segment of the molecule. The restricted CTL response to this protein parallels the restricted antibody response to this protein observed in humans and mice.     Description: The circumsporozoite (CS) protein of Plasmodium falciparum is the focus of intense efforts to develop an antisporozoite malaria vaccine. Localization of sites for T-cell recognition on this molecule is critical for vaccine design. By using an algorithm designed to predict T-cell sites and a large panel of H-2 congenic mice, a major nonrepetitive T-cell site was located. When a synthetic peptide corresponding to this site was covalently linked to the major B-cell site on the molecule, an immunogen capable of eliciting a high-titer antibody response was formed. This peptide sequence could prime helper T cells for a secondary response to the intact CS protein. The new helper T-cell site is located outside the repetitive region of the CS protein and appears to be the immunodominant T site on the molecule.     Description: This is the Limnological survey of the Cheshire, Shropshire, and Staffordshire Meres: Interim data report produced by the University of Liverpool in 1992. This report looks at the Limnological survey data from Cheshire, Shropshire and Staffordshire Meres. Limnological data of the report covers: changes in water conductivity, Phenolphthalein Alkalinity, Total Alkalinity, pH, Chloride concentrations, Soluble reactive Phosphorus, Total Phosphorus, Nitrate Nitrogen, Ammonium Nitrogen, Silicate, Chlorophyll, Carotenoids, Secci disk depth, changes in Trophic Score, changes in DAFOR scores for submerged and floating plants and Oxygen saturation during summer. This report also contains Seasonal maps of different Meres. The more important limnological data are plotted as seasonal means in relation to the sampling sites. Conductivity is shown as μSiemens per cm, alkalinity as milliequivalents per litre. Total and soluble reactive (available inorganic) phosphorus are shown in terms of P in μg per litre. Nitrate and ammonium are shown in terms of N in mg per litre. Chlorophyll a is given as μg per litre. A profile of oxygen saturation is shown. These profiles were obtained towards the middle of the day in August and September.

    Description: This is the report on Lakes – Classification and Monitoring, a strategy for the classification of lakes by the National Rivers Authority.This report describes a scheme for the assessment and monitoring of water and ecological quality in standing waters, greater than about 1ha in area, in England and Wales although it is generally relevant to Northwest Europe. Thirteen hydrological, chemical and biological variables are used to characterize the standing water body in any current sampling. Statistical testing on the chemical variables showed that at least six samples during a year would be needed to produce a representative sampling mean; but in this scheme the choice of variables minimizes logistic cost by not using boat sampling and time costs by not demanding extensive taxonomic work. Standing waters are classified in a state-changed system in which the contemporary values of the variables are compared with a reference baseline state and then placed in categories of percentage change from this baseline. The scheme is presently designed for use at about five year intervals on all lakes greater than 2ha area plus additional lakes of significant amenity or conservation interest.

    Description: Biomanipulation is a form of biological engineering in which organisms are selectively removed or encouraged to alleviate the symptoms of eutrophication. Most examples involve fish and grazer zooplankton though mussels have also been used. The technique involves continuous management in many deeper lakes and is not a substitute for nutrient control. In some lakes, alterations to the lake environment have given longer-term positive effects. And in some shallow lakes, biomanipulation may be essential, alongside nutrient control, in re- establishing former aquatic-plant-dominated ecosystems which have been lost through severe eutrophication. The emergence of biomanipulation techniques emphasises that lake systems are not simply chemical reactors which respond simply to engineered chemical changes, but very complex and still very imperfectly understood ecosystems which require a yet profounder understanding before they can be restored with certainty.
