    Notizen: Summary Although it is generally accepted that phagosome acidification is induced through the activity of a vacuolar proton pump (V-ATPase) present on the phagosome membrane, exactly how these pumps are delivered to the phagosomes is not well understood. To study this question inParamecium, it was necessary to first show that an authentic V-ATPase was present on their phagosomal membranes. Three antibodies raised against V-ATPases or their subunits were each found to label one or two large digestive vacuoles (DVs) inParamecium multimicronucleatum when immunofluorescence microscopy was used. Using horseradish peroxidase immunocytochemistry to increase sensitivity, about 10 DVs were shown to contain a V-ATPase. In high magnification images and cryoultramicrotomy these proton pumps were found to be located on the acidosomes, suggesting the vacuolar proton pumps on the DVs originate from the acidosomes. The authenticity of the V-ATPase was further confirmed by its sensitivity to cold temperature and to the V-ATPase specific inhibitor, concanamycin B, which at 10 nM doubled the t1/2 for vacuole acidification. Thus, we conclude that (1) acidosomes and some DVs ofParamecium have a bona-fide concanamycin B-sensitive and cold-sensitive V-ATPase, (2) the V-ATPase is delivered to the young DVs during acidosome fusion, and (3) the V-ATPase is involved in vacuole acidification. Finally, we have now determined thatParamecium has two immunologically related V-ATPases that are involved in two very different functions, (1) the acidification of phagosomes and (2) fluid segregation in the contractile vacuole complexes.

    Beschreibung: A ray-trace simulation code for the Hard X-ray Telescope (HXT) on board the Hitomi (ASTRO-H) satellite is beingdeveloped. The half power diameter and effective area simulated based on the code are consistent with groundmeasurements within 10%. The HXT observed the pulsar wind nebula G21.5-0.9 for 105 ksec. We confirmed that theencircled energy function and the half power diameter obtained from the data are consistent with the ground measurements.

    Beschreibung: Results from a Ginga observation of the AM Her type object H0538 + 608 are presented. During the first half of the observation (pulsing state) the source showed periodic modulations caused by the white dwarf rotation, and the modulation profile was characterized by a flat top and a flat bottom. During the second half, the source was in a remarkable flaring state and the periodic modulation disappeared. The dramatic change of the X-ray light curve is interpreted in terms of a change of accretion pattern between an eclipsing spot and a noneclipsing spot. The spectrum is well described throughout by a thin thermal model including an iron K-line with equivalent width of some hundred eV, supporting the standard model for the hard X-ray emission region. The temperature in the pulsing state was found to change with the spin phase of the white dwarf. In the flaring state, the temperature showed a positive correlation with the source luminosity.

    Beschreibung: Evidence from a concurrent X-ray/optical observing program is presented which indicates that the orbital period of the anomalous AM Her-type cataclysmic variable H0538 + 608 (BY Cam) differs from the rotational period by 1.3 +/- 0.1 percent. This finding would explain much of the unusual behavior previously exhibited in optical photometry and polarimetry. The model proposed by Ishida et al. (1991) is developed further using observations of accretion and the primary characteristics of optical and X-ray light curves and radial velocity measures of H-alpha. An optical QPO with a period of about 30 min was seen on two nights and possible causes of it are discussed.

    Beschreibung: Gamma Cassiopeiae is an enigmatic Be star with unusually strong hard X-ray emission. The Suzaku observatory detected six rapid X-ray spectral hardening events called "softness dips" in a approx.100 ks observation in 2011. All the softness dip events show symmetric softness-ratio variations, and some of them have flat bottoms apparently due to saturation. The softness dip spectra are best described by either approx.40% or approx.70% partial covering absorption to kT approx.12 keV plasma emission by matter with a neutral hydrogen column density of approx.(28) 10(exp 21)/sq cm, while the spectrum outside these dips is almost free of absorption. This result suggests the presence of two distinct X-ray-emitting spots in the gamma Cas system, perhaps on a white dwarf (WD) companion with dipole mass accretion. The partial covering absorbers may be blobs in the Be stellar wind, the Be disk, or rotating around the WD companion. Weak correlations of the softness ratios to the hard X-ray flux suggest the presence of stable plasmas at kT approx 0.9 and 5 keV, which may originate from the Be or WD winds. The formation of a Be star and WD binary system requires mass transfer between two stars; gamma Cas may have experienced such activity in the past.
